Abstract: Synaptotagmin-9 (Syt9) is a Ca2+ sensor mediating fast synaptic release expressed in many parts of the brain. The presence and role of Syt9 in retina is unknown. We found evidence for Syt9 expression throughout the retina and created mice to conditionally eliminate Syt9 in a cre-dependent manner. We crossed Syt9fl/fl mice with Rho-iCre, HRGP-Cre, and CMV-cre mice to generate mice in which Syt9 was eliminated from rods (rodSyt9CKO), cones (coneSyt9CKO), or whole animals (CMVSyt9). CMVSyt9 mice showed an increase in scotopic electroretinogram (ERG) b-waves evoked by bright flashes with no change in a-waves. Cone-driven photopic ERG b-waves were not significantly different in CMVSyt9 knockout mice and selective elimination of Syt9 from cones had no effect on ERGs. However, selective elimination from rods decreased scotopic and photopic b-waves as well as oscillatory potentials. These changes occurred only with bright flashes where cone responses contribute. Synaptic release was measured in individual rods by recording anion currents activated by glutamate binding to presynaptic glutamate transporters. Loss of Syt9 from rods had no effect on spontaneous or depolarization-evoked release. Our data show that Syt9 is acts at multiple sites in the retina and suggest that it may play a role in regulating transmission of cone signals by rods. Copy rights belong to original authors. We just need to move forward.""We probably spent six weeks negotiating that. Well, the first offer we spent about three, two, or three weeks on and just couldn't figure out a way to do it. That second offer, we spent about six weeks really hammering out terms, wanting a lot of assurances about how things were gonna move forward."Christina: Getting guarantees for their people was a top priority"One of the things we negotiated, we spent about three weeks negotiating, is a clause in the contract that said you have to keep our people for at least six months. You absolutely cannot change their pay, you can only increase it, and you have to give them these benefits you're promising them. Dr. Mitchell first explains how he met his co-founder, interventional radiologist Andy Thoreson, while they were both practicing medicine at the Keesler Air Force base. They connected due to common interests in business and investing. Eventually, they began to discuss clinical needs that they saw in their everyday lives and potential devices to address them. To protect their medical device ideas, they filed IPs and addressed patents to a company, rather than themselves as individuals. Dr. Mitchell emphasizes that this method is highly favorable when there are multiple inventors of a device, since assigning the patent to a separate entity will prevent legal issues and de-risk the business for future acquirers. When forming a new company, Dr. Mitchell believes that good corporate governance is key. This involves asking fundamental questions about the business goals, location of incorporation, type of corporation, and fundraising strategies. Eventually, Drs. Mitchell and Thoreson launched Redsmith, which was purchased by BD. Due to external factors, the purchase took two years. Next, the co-founders embarked on a mission to develop Oncodisc, a port that would automatically notify doctors when cancer patients' physiological signs indicated high risk for sepsis. Dr. Mitchell described a new mindset for this second product– He wanted to build a business out of this technology instead of selling it to a larger company. Taking a product through commercialization would allow him to have more influence over the final product and result in a larger impact on the healthcare system. Additionally, he wanted to explore applications of Oncodisc beyond cancer care, since ports are utilized in many chronic conditions such as end-stage renal disease and congestive heart failure. What started as a simple addition to chemotherapy ports became a larger digital health mission. Finally, Dr. Mitchell discusses the mechanics of his fundraising process. The seed round mainly included friends and colleagues who were clinicians, since they recognized the utility of Oncodisc. Having this network of investors also allowed the co-founders to obtain clinical advice during R&D. The subsequent Series A round presented different challenges, since Dr. Mitchell was presenting to investors from highly specialized business backgrounds, and little clinical expertise. He had to learn how to address extensive questions over regulatory strategy and effectively communicate his clinical ideas during his pitches. For those who may have missed last week's show, David Thoreson is an explorer, storyteller, photographer, and ocean and climate advocate. He is the first American explorer in history to sail the NW Passage in both directions. David has sailed more than 70,000 nautical miles, including below the Antarctic Circle, 3 times across the Atlantic, 8 times over the Arctic Circle, twice around Cape Horn and in 2009-2010, was the photographer on board OceanWatch that sailed the 28,000-mile circumnavigation of North and South America. His documentary of that voyage was nominated for an Emmy and published in the beautiful photo book: One Island, One Ocean. Exploring the Arctic led Thoreson to see the issues of climate change and water quality, and he hopes now to inspire others to explore their connections to nature and address climate issues locally and globally.

I recommend you do a little bit of research on your own. Look up different types of meditation and some scientific studies that show the benefits of meditating. Please don't say “I can't meditate because I can't clear my mind”. Of course, you can't you are only human. Mindfulness meditation isn't about “clearing your head”.Give meditation a try and let me know how it works out for you in the comments in a couple weeks! I am also advocating for mindfulness throughout your daily life. When you are walking do you feel your feet move back and forth to move you across a massive space rock? When you are eating pay attention and smell and taste your food. Don't watch or listen to anything. Start practicing mindfulness in all your activities. Today on the Tales of a Red Clay Rambler Podcast I have an interview with Thor Thoreson. A student of Marguerite Wildenhain from 1970-73, Thor talks about his time at the Pond Farm School and the influence of Wildenhain’s modernist teachings. He came to Pond Farm at the suggestion of Dean Schwartz, a ceramics teacher at Luther College who sent many students to study with Wildenhain. Schwartz later compiled the comprehensive text on Pond Farm, titled Marguerite Wildenhain and the Bauhaus: An Eyewitness Anthology. In addition to the interview, this episode features an audio tour of the surviving buildings at the Pond Farm School given by Thoreson. Wildenhain immigrated to the US after studying at Walter Gropius’s Bauhaus school in Weimar, Germany. While there she studied with sculptor Gerhard Marcks and potter Max Krehan, later taking this master/student teaching style to Pond Farm. The school was founded in the early 1940’s seventy-five miles north of San Francisco by Gordon and Jane Herr.
