Our guests are Karina Smith and Michelle Culbert, from South Florida, who both have Optic Nerve Glioma, a rare eye condition affecting sight that typically affects young children. It is a slow-growing brain tumour in or around the optic nerve, which connects the eye to the brain. As the tumour progresses, it presses on the optic nerve.Karina and Michelle believe they are the only two adults in Florida with this rare eye condition. Sadly, there is very little knowledge, advice, and guidance on how to treat optic nerve glioma in adults. They both explain how what the sudden onset of sight loss was like for each of them, physically and emotionally and the steps that they had to take to weather the mental health impact it took. For over 50 years, this dark, isolated road has swallowed up women and girls in the night from 1969 to present. Abstract: In normal tissue homeostasis, bidirectional communication between different cell types can shape numerous biological outcomes. Many studies have documented instances of reciprocal communication between fibroblasts and cancer cells that functionally change cancer cell behavior. However, less is known about how these heterotypic interactions shape epithelial cell function in the absence of oncogenic transformation. Furthermore, fibroblasts are prone to undergo senescence, which is typified by an irreversible cell cycle arrest. Senescent fibroblasts are also known to secrete various cytokines into the extracellular space; a phenomenon that is termed the senescence-associated secretory phenotype (SASP). While the role of fibroblast derived SASP factors on cancer cells has been well studied, the impact of these factors on normal epithelial cells remains poorly understood. We discovered that treatment of normal mammary epithelial cells with conditioned media (CM) from senescent fibroblasts (SASP CM) results in a caspase-dependent cell death. This capacity of SASP CM to cause cell death is maintained across multiple senescence-inducing stimuli. However, the activation of oncogenic signaling in mammary epithelial cells mitigates the ability of SASP CM to induce cell death. Despite the reliance of this cell death on caspase activation, we discovered that SASP CM does not cause cell death by the extrinsic or intrinsic apoptotic pathway. Instead, these cells die by an NLRP3, caspase-1, and gasdermin D (GSDMD)-dependent induction of pyroptosis. Taken together, our findings reveal that senescent fibroblasts can cause pyroptosis in neighboring mammary epithelial cells, which has implications for therapeutic strategies that perturb the behavior of senescent cells. This is part two of an afternoon's conversation with Taylor Culbert. Taylor is a PhD candidate in Theatre and Performance at the Graduate Center at the City University of New York. She is writing her dissertation on 17th century animal performances in Europe, looking specifically at how cultural narratives about animals shaped the ways that humans and animals responded to one another. In Part 1 we began with stag hunts and some surprising revelations about animal intelligence. It was a conversation with many interesting twists and turns. Taylor described the challenge of finding accounts from the 17th century about the relationship performers developed with the animals they worked with. I invited Dominique to share what she had observed in the artists and trainers who performed in the Cavalia shows. It's been a few years since that show was on tour, so Dominique began by describing the show. Cavalia is like Cirque du Soleil only with horses. In traditional circuses, horses are worked in the round always within reach of a long whip. In Cavalia they wanted to break away from this traditional image of superiority of humans over animals to portray a relationship built around friendship. Part 2 begins with Dominique sharing some stories from the show describing the relationship that developed between the artists and their equine performing partners. From there we circled back into the 17th century to Taylor's research and that brought us back around to the importance of positive reinforcement training and the role it plays in creating safety nets under horses.

When she isn't writing, Taylor works part-time training horses at a rescue in upstate NY. Taylor is enrolled in my on-line clinics so I've been getting to know her. I asked recently what her PhD dissertation is on and was so intrigued by her answer, I knew I wanted to learn more, so I invited her to join us for an afternoon's conversation. We began with stag hunts and some surprising revelations about animal intelligence. It's a conversation with interesting twists and turns. We enter a time warp between 17th century Europe and our current time which prompts Dominique to share stories about Cavalia, the theater company she co-founded that brought horses, acrobats, and other artists together in performance. This is the second half of our conversation with Christa Culbert. Christa is an artist, a horse trainer, and an educator. She is pursing a career in the horse world, and dreams of starting a classical dressage clicker training horse rescue someday. Christa has been participating in my on-line clinics so I've been watching her progress with her two older horses. In this episode she introduces us to Huck, a horse she rescued when he was so lame he could barely walk. We talk about the choices she made for him, including the role that training has had in his recovery. We also talk about Christa's experiences living with her horses in their pasture. That's a life style I know many people would love to have with their own horses. Other topics include using clicker training to teach young children. Christa asks an important question: What kind of leaders do we need? Her answer: “We need people who are creative, who look to science, who are good listeners, and when something is not going their way, they break things down or try a different way - all things that clicker training teaches. In the recent Dressage Camp one of the standouts for me during the introductions were how many of the participants were working with older horses. It was so good to hear how many people were choosing to rescue older and often unsound horses. They wanted to know how they could help their horses, so clearly this was a great topic for the podcast. I invited one of the Dressage Camp participants, Christa Culbert, to spend the afternoon with us in conversation. Christa is an artist, a horse trainer, and an educator. She is pursing a career in the horse world, and dreams of starting a classical dressage clicker training horse rescue someday. Christa has been participating in my on-line clinics so I've been watching her progress with her two older horses. Both came from a horse rescue. In this episode we talk about Christa's background both in the horse world and her search for a way of working with horses that feels morally right. That search brought her to clicker training. In so many of the podcasts we talk about what we can learn from the science of behavior analysis. In this conversation Christa shares how her training as an artist has informed her horse training.

If you were around for the mid 2000's Metal explosion, Bleed The Sky should be familiar to you (or is one of your fav's like it is mine).  With their recent line up change by adding Rudy Flores to vocals (So This Is Suffering) and dropping their newest single "Rot in Flesh", Belled The Sky is ready to get back out on the dance floor and mix it up with their fans.Formed in 2003 in Cali, they quickly signed to Nuclear Blast in 2004 and dropped "Paradigm in Entropy" in 2005.  What followed were multiple tours, another album and then a hiatus in 2009 to 2018(ish).David Culbert talks with me about the band back then, present day, and what's to come.  David has ties to Central Texas; San Antonio and Austin/Round Rock area.  So hopefully we get so see them head this way for somewhat of a reunion.

Dr. Gabriel Culbert is an Assistant Professor in the College of Nursing at the University of Illinois Chicago and Dr. Agung Waluyo is the Directorate of Community Engagement & Empowerment at the Universitas Indonesia. Gabe and Agung describe how they met in Jakarta in 2003 and how their research partnership has developed since then. Agung reflects on what it was like to learn about drug use and the HIV epidemic after a sheltered childhood, recalls being warned that he could be sent to jail for doing stigma research focused on Indonesian healthcare providers, and shares how he navigates conversations about politically sensitive research with government officials. Gabe describes the reasons why the HIV epidemic became concentrated in prisons in Indonesia, questions why Indonesia has one of the highest HIV mortality rates despite having the largest universal healthcare system in the world, and envisions an expanded role for nurses in HIV prevention and treatment. Valerie and Carly nominate Gabe and Agung to be poster children for international research partnerships.
