Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2023.03.06.531336v1?rss=1 Authors: Banerjee, S., Gadpayle, M. P., Das, S., Samanta, S., Datta, R., MAITI, S. Abstract: PDZ protein interacting specifically with Tc10 or PIST is a mammalian trans-Golgi resident protein that regulates subcellular sorting of plasma membrane receptors. PIST has recently been found to play an important role in regulating viral pathogenesis. Alteration in PIST expression is linked to the reprogramming of cell surface receptors which is crucial in determining herpes simplex virus1 infection. In this context, PIST is crucial in triggering autophagy via Beclin 1 -PI3KC3 pathway. However, there is complete lack in our knowledge on the role of this protein in any parasitic infection. Leishmania parasites infect their host macrophage cells via phagocytosis and their survival within the parasitophorous compartment has recently been found to be dependent on host autophagy by a yet to be identified mechanism. Using Leishmania major (L. major)- macrophage infection model system we, for the first-time report here that in infected macrophages Golgi resident PIST protein migrates towards the parasite containing compartment. We have also found that PIST associates with Beclin 1, however, not with LC3 within the L. major parasite containing compartment of infected macrophages. Further, we performed genetic ablation of PIST by siRNA and observed that knockdown of macrophage PIST in turn helps in parasite replication. Contrary to this, overexpression of PIST in macrophages restricted the multiplication of L. major. Collectively, our study for the first time reveals that PIST is essential in regulating intracellular parasite, L. major infection within macrophage cells. Copy rights belong to original authors. Today we explore the social dynamic of a thru-hike. If you've listened to previous seasons shows on this subject, you might think that you already know the answer to this question. However, let us not forget, our trekkers are hiking against the current. Whereas the vast majority of thru-hikers- regardless of which triple crown trail they're on- hike northbound, our hikers are trekking south. For that reason, the bubbles that one experiences on the AT or PCT may look a lot different. Or will it? In addition to the number of fellow hikers a person might encounter, we also hear about the quality of the companionship. Our Correspondents are more than a month into their journey and we'll learn the role their fellow trekker is playing on the enjoyment of their own journey. Roughly one month into their journey, our correspondents are now at the point of learning about just how much they don't know about their trail, themselves, and thru-hiking as a whole. After all, even the most experienced thru-hikers have gaps in their research, preparation, and occasionally fail to pack needed items of gear. This episode is all about the twists, turns, and surprises that are an intrinsic part of life on trail. While we won't provide too many spoilers, our correspondents regale us with their stories of combating relationship issues (with those on trail and those at home), difficult conditions (foreseeable and not), and learning to be adaptable. Don't worry though, this episode isn't all serious - sometimes the spontaneity of trail life leads to wonderful moments - both heartwarming and belly-laugh-inducing alike. It's Wildcard time! A few times a season, we challenge our correspondents' to be creative and discuss whatever they want to! Our trail correspondents lived up to the challenge that the wildcard presents. They regal us with a variety of stores, give us topics to ponder, and provide us an insight as to what "real life" on trail looks like. Turn's out, our correspondents get quite philosophical when they get to deal their own hand! Well into the first few weeks of hiking, most of our correspondents will be calling in with 100+ miles under their boots. At this point, many of our correspondents have now experienced both the highs and the lows of thru-hiking. From the struggles of having to separate from trafamlies to impromptu hotel room interviews, this episode is jam-packed newfound thru-hiker wisdom. Tune in to hear what our correspondent came up with for our first wildcard episode! Get 10% off at Greenbelly.co by using code "TC10" at checkout. - Manuel Mesa. Piloto.
What's up guys, pretty high scoring week this week. Probably the highest week for the whole league ever, in total. Good work. Lots of big-time individual performances will be covered. In other news, Henry lost a nail-biter, despite scoring 120+, to fall to 1-4, and also unfortunately no Joe Twitter rant of the week. Forgot to check it. Was in a semi-rush recording. Also shoutout Henry's mom for cheering me on at TC10 on Sunday. Forgot to shout her out at the end of the pod. Anyways, hope all is well. Peace & love --Ben

Tom Cairney’s thunderbolt against Leeds was just one of the highlights of what could turn out to be a pivotal week in Fulham’s season, as a last-gasp point on Tuesday night was eclipsed by an away domination of league leaders Newcastle United. There’s also time for us to wax lyrical about both TC10 and man of the moment, Ryan Sessegnon, as we dissect both games and examine just how good Fulham’s playoff credentials are. On top of that, we preview two crucial but winnable home games against Blackburn Rovers and Wolverhampton Wanderers, before turning our hand to some of the best questions that we’ve got in the mailbox this week. As ever, if you’re interested in getting involved with Fulhamish let us know - hit us up at our sparkly new email address: pod@fulhamish.co.uk See acast.com/privacy for privacy and opt-out information.

Vale a pena escutar porque o programa ficou bem divertido.
