hVIVO PLC CEO Yamin 'Mo' Khan talked with Proactive's Stephen Gunnion about the company's involvement in a world-first Phase 3 human challenge trial for a pertussis (whooping cough) vaccine candidate. The trial, in partnership with ILiAD Biotechnologies, is expected to start in the second half of this year. Khan highlighted the importance of this milestone for hVIVO, noting that it will be the company's largest challenge trial to date and its first bacterial challenge trial. The trial data aims to secure FDA marketing authorisation, marking a pivotal moment for the utility of human challenge trials in vaccine development. According to Khan, "The goal is to get a better vaccine to patients faster. Pertussis is a severe disease, particularly for infants, and there's an urgent need for new vaccines." He further explained that hVIVO's state-of-the-art Canary Wharf facility will play a critical role in supporting this trial, equipped with expanded laboratory capabilities for bacterial analysis. The trial comes as cases of pertussis are on the rise, increasing sixfold between 2023 and 2024. ILiAD Biotechnologies has also secured FDA fast-track status and an MHRA innovation passport for this vaccine candidate. Khan emphasised the partnership's goal of addressing this unmet medical need and delivering an effective vaccine to patients worldwide. Recce Pharmaceuticals Ltd (ASX:RCE, OTC:RECEF) CEO James Graham joins Proactive's Tylah Tully to discuss the approval the company has received from Indonesia's Drug and Food Regulatory Authority (Badan POM) for a registrational Phase 3 clinical trial of RECCE® 327 topical gel (R327G) for treating diabetic foot infections (DFIs). The trial will start in mid-December and represents a significant milestone for the company in the ASEAN region. It will be conducted as a double-blinded, placebo-controlled study, enrolling up to 300 patients, with results expected in late 2025 and potential commercialisation in 2026. The trial has support from Indonesian stakeholders, including the Ministry of Health and PT Etana Biotechnologies as well as the Australian Government. The project will cost approximately US$2 million, partially offset by Australia's 43.5% R&D rebate. This will be one of the largest DFI studies globally and the first in Indonesia, addressing urgent health challenges in a country with more than 19.5 million adults living with diabetes. In Australia, Recce plans to launch a Phase 3 trial of R327G in 2025, aligning with its ASEAN-focused strategy. Known as 4D-150, the wet AMD gene therapy performed well in a Phase 2b clinical trial.

In a conversation with CancerNetwork®, Nader Sanai, MD discussed the current state of the glioblastoma field, highlighting ongoing research efforts to help improve outcomes among patients with this disease.  Sanai is the director of the Ivy Brain Tumor Center and J.N Harber Professor of Neurological Surgery, Francis and Dionne Najafi chair for Neurosurgical Oncology, and chief of neurological oncology at Barrow Neurological Institute. Specifically, Sanai described plans to assess treatment with niraparib (Zejula) compared with temozolomide (Temodar) in a population of patients with newly diagnosed MGMT unmethylated glioblastoma as part of the phase 3 Gliofocus study (NCT06388733).1 He contextualized the rationale for conducting this study by focusing on findings from a proof-of-concept hybrid study (NCT05076513) and detailing how they supported additional investigation into the utility of niraparib.  According to findings from this proof-of-concept study presented at the 2024 American Society of Clinical Oncology (ASCO) Annual Meeting, the median overall survival (OS) was 20.3 months among patients who received niraparib in combination with radiotherapy.2 Additionally, data showed that niraparib reached drug concentrations in Gadolinium-nonenhancing newly diagnosed glioblastoma tissue exceeding those of any other evaluated PARP inhibitors; investigators identified no new safety signals after combining niraparib with radiotherapy in this population. With the Gliofocus trial, Sanai and co-investigators aim to provide a clinically meaningful quality of life benefit with niraparib-based therapy beyond a marginally valuable statistical advantage. By evaluating treatment with niraparib, investigators look to improve historical survival rates reported with standard-of-care options among patients with unmethylated disease. “What we're looking to do with this trial is set a benchmark that's clinically relevant for patients and providers. The [OS] target for the study is 18 months, which is to effectively convert [a] 12-month natural history to a natural history closer to the methylated glioblastoma population,” Sanai said. “We think that is a meaningful transformation of a difficult patient population, a significant chunk of survival time that would be beneficial to patients, providers, and caregivers. The document, a “February 2024 Board Memo” prepared for board members of the American Clean Power Association, is striking in how specific and aggressive it is in detailing plans for its members to push Republican lawmakers to oppose any GOP effort to repeal all or parts of Biden's inaptly named Inflation Reduction Act (IRA). The IRA, which passed during Biden's second year as president, did not lower inflation but did aggressively expand government spending, including perhaps most controversially on the left's radical green energy agenda. The American Clean Power Association, or ACP, is a trade association made up of companies supportive of or benefitting from the left's green energy push. On its website, the organization describes itself as a “leading voice of more than 800 companies from across the clean power sector that are committed to meeting America's national security, economic and climate goals with fast-growing, low-cost, and reliable domestic power.” This 66-page document obtained by Breitbart News is marked on nearly every page as “ACP Confidential Information.”  Source: breitbart.com The CBO Revised The Cost Of Biden's Energy Policies Up By $466 Billion The cost estimate of Biden's Inflation Reduction Act coupled with EPA mandates, just jumped by $466 billion. The Committee for a Responsible Federal Budget and the latest Congressional Budget Office outlook conclude IRA Energy Provisions Cost Could Double With New Emissions Rule   At the time of passage, CBO and the Joint Committee on Taxation (JCT) estimated the IRA's energy and climate spending and tax breaks would cost about $400 billion through Fiscal Year (FY) 2031 and would be more than fully offset by other parts of the law. Since then, the combination of higher inflation, greater demand for credits, and looser-than-expected regulations significantly boosted the cost of those credits. Last June, we estimated the cost of the IRA energy provisions had grown by two-thirds, to $660 billion through 2031. Assuming the new vehicle emissions rule proposed by the EPA is finalized, we now estimate the cost of the provisions will more than double to $870 billion through 2031, or $1.
