Abstract: Clinically, there is a positive correlation between BMI and the risk of tendinopathy. However, the underlying mechanisms are not understood. Dyslipidaemia and increased circulating free fatty acids (FFA) are associated with increased BMI. We hypothesised that increased FFA concentrations negatively affect rat tendon-derived cells (rTDCs) through mitochondrial-mediated mechanisms. rTDCs were isolated and treated with oleic acid (OA), stearic acid (SA), and palmitic acid (PA). Cell viability was assessed using AlamarBlue assay, and gene expression using real-time PCR. Cell respiration and reactive oxygen species (ROS) production were measured using high-resolution respirometry and MitoSox staining. PA transport into the mitochondria was blocked by pre-treatment with 50 M etomoxir. Treatment with SA and PA at 10 g/ml decreased rTDC viability by 40% and 60%, respectively. PA decreased the gene expression of the tendon markers Scx and Tnmd, and increased the expression of Mmp3, Mmp13, and Ptgs2 (encoding Cox-2). FFA treatment increased the expression of Cpt1 and Pdk4, indicating an increase in mitochondrial FFA oxidation. PA, at 10 g/ml, increased cellular respiration and ROS production. Pre-treatment with etomoxir partially inhibited the effects of PA on cell viability, Mmp3 gene expression, ROS production, and cell respiration, but did not affect PA-induced inhibition of Scx or Tnmd expression. We found that increased saturated FFA concentrations in the microenvironment reduce cell viability and alter ROS production, respiration, and gene expression. Blocking PA transport into mitochondria partially reversed the negative effects of PA. Overall, an increase in saturated FFA concentrations may contribute to poor tendon health. Abstract: Skeletal stem and progenitor cells are critical for bone homeostasis and healing, but their identity and diversity in humans are not well understood. In this study, we compared stromal populations in matched tissues from the femoral head and neck of 21 human participants using spectral flow cytometry of freshly isolated cells. High-level analysis indicated significant differences in marker distribution between periosteum, articular cartilage, endosteum and bone marrow stromal populations, and identified populations that were highly enriched or unique to specific tissues. Periosteum-enriched markers included CD90 and CD34. Articular cartilage, which has very poor regenerative potential, showed enrichment of multiple markers, including the PDPN+CD73+CD164+ population previously reported to represent human skeletal stem cells. We further characterized periosteal populations by combining CD90 with other strongly expressed markers. CD90+CD34+ cells sorted directly from periosteum showed significant colony-forming unit fibroblasts (CFU-F) enrichment, rapid expansion, and consistent multi-lineage differentiation of clonal populations. In situ, CD90+CD34+ cells include a perivascular population in the outer layer of the periosteum and non-perivascular cells closer to the bone surface. In conclusion, our study indicates considerable diversity in the stromal cell populations in different tissue compartments within the adult human skeleton, and suggests that periosteal stem cells reside within the CD90+CD34+ population. Over his 5 years in Bath, primarily as a student, Saumya Masrani, who chose his last name as his moniker, involved himself in his immediate British electronic music scene, becoming the resident DJ for local promoters Origin Sounds. Subsequently, the Mumbai-born UK-based talent found the opportunity to open for the likes of Scuba, Or:la and DJ Stingray as JUKEBOXX, and later followed the trail to his hometown with support slots for Cinnaman and Lilly Palmer. All the nights of partying seeped into his work as a DJ and earlier this year he showcased the same as a producer with the release of his dark dance track ‘Growling Hunter’. Bringing the same musical knowledge rooted in the British-scene to his Wild City mix, Masrani starts at the disjointed fringes of house and techno and quickly edges into leftfield bass and jungle. There is a pre-headline slot sensibility to the selections which balance atmosphere, with the likes of rRoxymore and Kosh, against energy, through numbers by Cain and Bolam. The presence of tribal and hip-hop inflexions keeps the palette varied and the interest intact while the intensity of a full-blown simmers under the surface of the mix. In 1965, Major Joseph Gervais was invited to speak at a gathering of retired pilots. While he was there, he was introduced to Mrs. Irene Bolam by one of Amelia Earhart's closet friends, Viola Gentry. Gervais felt he instantly recognized her as an older version of Amelia Earhart and commenced to research her past. Using Gervais' research, author Joe Klaas documented his assertion in his book Amelia Earhart Lives released in 1970. Bolam denied being Earhart, filed a $1.5 million lawsuit and submitted a lengthy affidavit refuting the claim. The book's publisher McGraw-Hill pulled Klaas' book from the market shortly after it was released, and court records indicate they made an out of court settlement with her after she refused to give a set of fingerprints to the court upon her victory.  On Bolam's death, Gervais sought permission to photograph and fingerprint the body, but permission was denied. After Amelia Earhart Lives was published in 1970, three additional books were subsequently published that continued to support the Irene Bolam hypothesis as it’s come to be known. Those books were titled, Stand by to Die by Robert Myers and Barbara Wiley, Amelia Earhart Survived by Colonel Rollin C. Reineck (2003), and in January 2016, Amelia Earhart: Beyond the Grave by W. C. Jameson was published. The authors of these books continued to promote the theory that Bolam and Earhart were one and the same. Shortly after the passing of Irene Bolam in 1982 researcher and author Bob Meyers, who we mentioned earlier and who had known Amelia Earhart as a young teenager had multiple conversations with Irene’s closest friend and confidant Diana Dawes about the role that Irene played in the repatriation of Amelia Earhart. Recently courtesy of aviation pioneer Ann Holtgren Pellegreno who herself completed Amelia’s world flight in a Lockheed 10 we received those tapes. Conversations never before heard by the general public. Until now.  Friends of Amelia Earhart like Bob Meyers, Art Kennedy and many of the Zonta believed that Amelia Earhart returned and that Irene was Amelia. While denying it on public platforms, Irene herself played coy about her background and past in private; even going so far as to tease family and friends that there was more to her than meets the eye. Irene’s own son Larry Heller stated emphatically that when Irene passed away in 1982, the woman pictured on the funeral program was not the woman who raised him. That’s because according to the theory, that woman didn’t exist before 1940. At least not as Irene.    For most people, this theory is the most far-fetched of all the many theories in this case. Geoff Bolam is an automotive artist who comes from a long line of artists, going back 5 generations with a distant relation to the great William Morris. An early fascination with cars and machines was nurtured by his father Denis, a Marine Engineer, who taught him how to work on cars. Trained as a graphic designer. he was a freelance illustrator and lettering artist who became a creative director and design consultant in central London for nearly 20 years. Geoff started painting for pleasure in 2005 and made the transition to full-time painting in 2014. His unique style is painting oils onto aluminum panels using various surface treatments including etching, polishing, and wire brush techniques. His work has been featured in numerous automotive magazines and for the last 3 years, his art has been selected for the RAC’s ‘Art of Motoring’ exhibition, which showcases the ‘finest artists working in the genre’. I had the pleasure of meeting Geoff and enjoying his art at Automobilia Monterey this past summer.
