Abstract: Long-term memories are stored as stable configurations of neuronal ensembles, termed engrams. While investigation of engram cell properties and functionality in memory recall has been extensive, less is known about how engram cells are affected by forgetting. We describe a form of interference-based forgetting using an object memory behavioral paradigm. By using activity-dependent cell labelling, we show that although retroactive interference results in decreased engram cell reactivation during recall trials, optogenetic stimulation of the labelled engram cells is sufficient to induce memory retrieval. Forgotten engrams may also be reinstated via the presentation of similar or related environmental information. Furthermore, we demonstrate that engram activity is necessary for interference to occur. Taken together, these findings indicate that retroactive interference modulates engram expression in a manner that is both reversible and updatable. Retroactive inference may constitute a form of adaptive forgetting, where in everyday life new perceptual and environmental inputs modulate the natural forgetting process. A., Skelton, S., McKissick, O. P., Bladon, J. H., Ramirez, S. Abstract: Memory engrams are both necessary and sufficient to mediate behavioral outputs. Defensive behaviors such as freezing and avoidance are commonly examined during hippocampal-mediated fear engram reactivation, yet how reactivation of these cellular populations across different contexts engages the brain to produce a variety of defensive behaviors is relatively unclear. To address this, we first optogenetically reactivated a tagged fear engram in the dentate gyrus (DG) subregion of the hippocampus across three distinct contexts. We found that there were differential amounts of light-induced freezing depending on the size of the context in which reactivation occurred: mice demonstrated robust light-induced freezing in the most spatially restricted of the three contexts but not in the largest. We then utilized graph theoretical analyses to identify brain-wide alterations in cFos co-activation during engram reactivation across the smallest and largest contexts. Our manipulations conferred greater positive cFos correlations and recruited regions spanning putative fear and defense systems as hubs in the respective networks. Moreover, reactivating DG-mediated engrams generated network topologies across experimental conditions, emphasizing both shared and distinct features. By identifying and manipulating the circuits supporting memory function, as well as their corresponding brain-wide activity patterns, it is thereby possible to resolve systems-level biological mechanisms mediating memory's capacity to modulate behavioral states. Abstract: Memories are thought to be stored in neural ensembles known as engrams that are specifically reactivated during memory recall. Recent studies have found that memory engrams of two events that happened close in time tend to overlap in the hippocampus and the amygdala, and these overlaps have been shown to support memory linking. It has been hypothesised that engram overlaps arise from the mechanisms that regulate memory allocation itself, involving neural excitability, but the exact process remains unclear. Indeed, most theoretical studies focus on synaptic plasticity and little is known about the role of intrinsic plasticity, which could be mediated by neural excitability and serve as a complementary mechanism for forming memory engrams. Here, we developed a rate-based recurrent neural network that includes both synaptic plasticity and neural excitability. We obtained structural and functional overlap of memory engrams for contexts that are presented close in time, consistent with experimental studies. Moreover, we showed that enhancing the initial excitability of a subset of neurons just before presenting a context biases the memory allocation to these neurons. We then explored the role of inhibition as a way of controlling competition among neurons from two ensembles. This work suggests mechanisms underlying the role of intrinsic excitability in memory allocation and linking, and yields predictions regarding the dynamics of memory engrams. Abstract: Alcohol withdrawal directly impacts the brain's stress and memory systems, which may underlie individual susceptibility to persistent drug and alcohol-seeking behaviors. Numerous studies demonstrate that forced alcohol abstinence, which may lead to withdrawal, can impair fear-related memory processes in rodents such as extinction learning, however the underlying neural circuits mediating these impairments remain elusive. Here, we tested an optogenetic strategy aimed at mitigating fear extinction impairments in male c57BL/6 mice following exposure to alcohol (i.e., ethanol) and forced abstinence. In the first experiment, extensive behavioral extinction training in a fear-conditioned context was impaired in ethanol-exposed mice compared to controls. In the second experiment, neuronal ensembles processing a contextual fear memory in the dorsal hippocampus were tagged and optogenetically reactivated repeatedly in a distinct context in ethanol-exposed and control mice. Chronic activation of these cells resulted in a context-specific, extinction-like reduction in fear responses in both control and ethanol-exposed mice. These findings suggest that while ethanol can impair fear extinction learning, optogenetic manipulation of a fear engram is sufficient to induce an extinction-like reduction in fear responses. Abstract: The hippocampus processes both spatial-temporal information and emotionally salient experiences. To test the functional properties of discrete sets of cells in the dorsal dentate gyrus (dDG), we examined whether chronic optogenetic reactivation of these ensembles was sufficient to modulate social behaviors in mice. We found that chronic reactivation of dDG cells in male mice was sufficient to enhance social behaviors in a female exposure task when compared to pre-stimulation levels. However, chronic reactivation of these cells was not sufficient to modulate group differences in a separate subset of social behaviors, and multi-region analysis of neural activity did not yield detectable differences in immediate-early gene expression or neurogenesis, suggesting a dissociation between our chronic stimulation-induced behavioral effects and underlying neural responses. Together, our results demonstrate that chronic optogenetic stimulation of cells processing valent experiences enduringly and unidirectionally modulates social interactions between male and female mice. What is learning and memory? Is amnesia really a loss of stored memories or something more? In this interview, we will discuss where memory goes and if there is a way to retrieve them after trauma. Recent studies have shown that spatial memories are encoded as sparse populations of cells that are activated during learning and are necessary for the retrieval of specific memories. We refer to these cells as "memory engram cells" and the focus of Tomás J Ryan and his team is to understand how engram cells are able to store specific memories as information. Here is a quick summary/breakdown of the video above. The old storage theory of amnesia is WRONG. The problem is the ability to retrieve the memory. Short term memory lasts a few hours. It is very disruptable (long term is not). Long term memory lasts your entire lifetime. It requires transcription and translation of many different genes. We have many different genes that regulate memory. They are different for short term and long term - playing roles at different stages. Most popular was CREB (turns on other genes). Engrams cells are structures in the brain where memories are stored. R They used optigenetics to prove engram cells were important for memory. Engram cells only account for 4-6% of the brain cells in the hippocampus (of mice). V Consolidation idea - a short term memory (unstable) forms into a long term memory (stable), thus stabilizing the engram. Optogenetics - let's us express photoactivatable opsins in whatever region of the brain we want. The brain is electrically active. They put certain opsins into brain cells and then stimulate them with light. These opsins can conduct ions - putting positively charged cations from the external environment of the cell into the neuron. This depolarizes the cell, which results in an action potential (creating neuronal activity). What they did at MIT was integrate optogenetics and memory engram cells. They tested parts of the brain using a transgenetics - they took immediate/early genes (genes that show activity as a function of neuronal spike) and used opsins to only activate those genes. Amnesia and Memory Learning is the enhancement of synaptic connections. Memory is a stable thing in the brain and can last for your entire life. Learning and recall are not "things", they are activities. Learning = process of making a memory. Recall = process of retrieving a memory. So the problem is not memory itself. Amnesic engram cells have a loss of synaptic strength. Increasing the synaptic strength helps get the memory (which is commonly seen in Alzheimer's (AD), Huntington's (HD)). If you increase the synaptic potentiation (increasing the dendritic spine density), esp. of the amnesic engram cells, you can get natural access to those memories. Neurotrophic factors (BDNF, NGF, CNTF, GDNF) do this, but are not localized nor memory specific. Learning something new (about something you are amnesic about) will train your brain to those particular engram cells. Once you activate the engram cells (with optogenetics), you can restore access to old information and add new information on those amnesic memories. Without dealing with the cause, you probably won't be able to really restore memories (in diseases like AD, HD, etc.) In late stages of neurodegeneration, you may actually lose memories.
