Recently, I reached out across a number of Facebook groups for wrestling fans to talk to for an upcoming feature called 'I Was There When...', mainly focusing on any major events or angles they have witnessed live in person. The 3 interviews I conducted and took highlights from for episode 13 of this podcast ('I Was There When...' - The Facebook Fans Edition) were so good that I thought they each deserved to be put out in full as episodes in their own right. Henry Rock II is the founder and director of City Startup Labs, an entrepreneur's academy for African American millennials that develops human, social and economic capital within inner-city America. In partnership with Ventureprise, Inc., a non-profit venture development organization, CSL operates a Center of Excellence for Entrepreneurial Competency, Innovation and Leadership at the University of North Carolina at Charlotte. Henry has over two decades of experience in media sales, advertising and marketing. His career includes business development and strategic planning for small black-owned and operated ventures. He has given a TEDxCharlotte Talk on creating a new class of African American entrepreneurs. Henry does more than inspire young men and women to live powerful and rewarding lives. He gives them the tools and shares the mindsets to make it happen. This episode is perfect for anyone interested in the education of entrepreneurs, a discussion on black cultural nationalism, and a basis for hope in our lives. IN THIS EPISODE Henry shares what City Startup Labs is and what it is designed to do. He explains the four modules that make up CSL that guide young entrepreneurs from acceleration to incubation to market. He discusses the development of his curriculum and how he makes use of lean startup principles and Barry Gordy's autobiography to reach his students. Henry talks about the mindset of young black men and how perceptions of them affect them psychologically and physically. He shares what has surprised him leading CSL. Henry describes himself as a young man, why he chose to take courses in economics, and his transition into a career in media and advertising. He talks about an amazing period in black media and how it informs what he is doing now. He discusses guiding forces in his life and why he is hopeful about the future. Esta noche segundo especial “Chicas del Rock” en Corsarios: Ya os podemos desvelar el contenido del programa de esta noche en Corsarios. Nos lo cuentan ellos mismos, como siempre. Hoy “Especial Chicas del Rock II”: Esta semana, concretamente el 8 de marzo se celebraba el Día Internacional de la Mujer. Ya sabéis que nosotros no somos de diferenciar entre chicos y chicas, hombres ni mujeres ni penes ni vulvas. Preferimos hablar de personas, amigos, trabajadores, válidos y con un gran cerebro y corazón para sacarlo todo adelante, con el rock por bandera a ser posible. Lo defendemos (y predicamos con el ejemplo) en Redhardnheavy, en Corsarios y en nuestra vida diaria, en el trabajo, con la familia, amigos y demás… Pero eso no quita para que nos sirva de buena excusa para reivindicar las bandas femeninas y el trabajo de todas las chicas y mujeres en el rock que han roto muchas barreras y han servido de ejemplo e inspiración (y lo siguen siendo) para que a día de hoy ver féminas en nuestro mundillo sea la norma y no la excepción. Ya lo hemos hecho otras veces, y lo seguiremos haciendo siempre que haga falta y nos apetezca. Así, pincharemos muchos grupos en los que las chicas tengan un lugar preponderante en la banda de turno, sea mayoría de chicos o no. Además, nos acordaremos de alguna de las pioneras, por supuesto, y a su vez de l nueva hornada de chicas guerreras que contribuyen aún más a que el mundo del rock y el heavy metal no tenga sexo a la hora de tener que calificarlo o valorarlo. Además, nos acordaremos de los conciertos que hemos visto estos últimos días, muchos y muy buenos, destacando por razones obvias (por la temática del programa) el de BATTLE BEAST ayer en Madrid. Boy Scouts looked a little different in those days and if you believed everything you read in those books you might be shocked. Die akute Pankreatitis beginnt in den exokrinen Azinuszellen des Pankreas und wird durch verschiedene, bisher nicht vollständig geklärte, intrazelluläre Vorgänge ausgelöst. Das Hormon Cholezystokinin stimuliert Signaltransduktionskaskaden, welche über eine Reorganisation des Aktinzytoskeletts zu einer akuten Organentzündung führen. In dieser Arbeit wurde untersucht, ob ein über das Enzym RhoA vermittelter intrazellulärer Signalweg zu Aktin-bindenden Proteinen im Pankreas diese Reaktion hervorruft und durch Cholezystokinin reguliert werden kann. Die Ergebnisse der vorliegenden Arbeit bringen den Nachweis der Existenz der im Folgenden beschriebenen Signaltransduktionskaskade in exokrinen Azinuszellen: RhoA führt über eine Aktivierung von ROCK II zu einer Phosphorylierung der Zieluntereinheit MYPT1 der Myosinphosphatase und somit zu einer Hemmung des Gesamtenzyms. Dadurch transloziert die sowohl in der Zytosol- als auch in der Zytoskelettfraktion vorkommende, unphosphorylierte Form MYPT1 vollständig ins Zytosol. Die Myosinphosphatase führt zu einer Dephosphorylierung der MLC von Myosin. Die fast vollständig in der Zytoskelettfraktion exprimierte phosphorylierte Form pMLC transloziert im dephosphorylierten Zustand ins Zytosol. Durch die Interaktion mit MYPT1 kann MLC zu einer Aktinmyosinkontraktion und somit zu einer Reorganisation des Aktinzytoskeletts führen. Über alternative Signalwege bewirkt RhoA eine Aktivierung von mDia, welches mittels Profilin zu einer Aktinpolymerisation führt. Über ROCK II wird eine Aktivierung der LIMK durch RhoA vermittelt. Dadurch wird Cofilin vermehrt phosphoryliert, wodurch die Depolymerisation der Aktinfilamente gehemmt wird. Durch eine dosis- und zeitabhängige Stimulation mit physiologischen und supraphysiologischen Dosierungen Cholezystokinin wird der Signalweg über RhoA gehemmt. Dadurch kann eine Kontraktion des Aktinzytoskeletts stattfinden und es zu einer Fusion von Vesikeln und zu einer Inhibierung des regulären Sekretionsmechanismus der Pankreaszellen kommen. Da die Hemmung von Aktin-modulierenden Proteinen eine bedeutende Rolle bei der Organfunktion und Entwicklung der akuten Pankreatitis spielt, trägt diese Arbeit dazu bei, sowohl die physiologischen als auch die pathophysiologischen Vorgänge innerhalb der Azinuszellen näher zu charakterisieren. Dies könnte zu einem besseren Verständnis der dieser Erkrankung zugrundeliegenden Mechanismen führen und somit einen therapeutischen Ansatz bei der akuten Pankreatitis darstellen.
