Abstract: Skeletal muscle stem cells (MuSC) are crucial for tissue homeostasis and repair after injury. Following activation, they proliferate to generate differentiating myoblasts. A proportion of cells self-renew, re-enter the MuSC niche under the basal lamina outside the myofiber and become quiescent. Quiescent MuSC have a primary cilium, which is disassembled upon cell cycle entry. Ex vivo experiments suggest cilia are important for MuSC self-renewal, however, their role in muscle regeneration in vivo remains poorly understood. Talpid3 (TA3) is essential for primary cilia formation and Hedgehog (Hh) signalling. Here we use tamoxifen-inducible conditional deletion of TA3 in MuSC (iSC-KO) and show that regeneration is impaired in response to cytotoxic injury. Repeat injury exacerbates the regeneration phenotype in TA3iSC-KO mice, indicating depletion of MuSCs. Single cell transcriptomics of MuSC progeny isolated from myofibers identifies components of several signalling pathways, which are deregulated in absence of TA3, including Hh and Wnt. Pharmacological activation of Wnt restores muscle regeneration, while purmorphamine, an activator of the Smoothened (Smo) co-receptor in the Hh pathway, has no effect. Together, our data suggest that TA3 and primary cilia are important for MuSC self-renewal, and that pharmacological treatment can efficiently restore muscle regeneration. A la edad de trece años, Abba y su familia fueron deportados al gueto de Kovno. Luego fue deportado al campo de concentración de Stutthof y más tarde a un subcampo del campo de concentración de Dachau. Tuvo que hacer un duro trabajo en estos campos, hasta que en la primavera de 1945 se vio obligado a realizar la Marcha de la Muerte hasta el subcampo Kaufering V en Utting am Ammersee. Allí fue liberado por el ejército estadounidense. Después de la Segunda Guerra Mundial emigró a la tierra de Israel. Tiene dos hijos, cinco nietos y diez bisnietos.

Umso erstaunlicher ist´s dass sie das Lied heuer auch im evangelischen Karfreitagsgottesdienst in Utting singen wird, den kein geringerer als der Landesbischof persönlich halten wird. An interview with Brittany Utting and Daniel Jacobs of HOME-OFFICE. In this episode, Brittany and Daniel discuss their latest projects and how architecture overlays with labor, ecology, and the politics of collectivity.

Liebe Freunde und Gäste, unser Schiff, die "Sir Shackleton", ist in den frühen Morgenstunden des 18.08.2020 an ihrem Bojenliegenplatz vor St. Alban gesunken. Der Schrecken war für uns sehr groß, als wir die Nachricht dazu bekommen haben. Aus dem Wasser ragten nur noch die beiden Masten, das Schiff kam auf rund 7 Metern Tiefe im Schlick zum liegen. Die Feuerwehr Dießen rückte an und hat unter Aufsicht des Wasserwirtschaftsamtes in Weilheim eine Ölsperre ausgebracht, um auslaufende Treib- und Betriebsstoffe aus dem Motor aufzufangen. Die Versicherung schickte einen Gutachter, der die Bergung organisiert und koordiniert hat. Da wir am Ammersee nicht so viele Schiffe mit einem Gewicht von knapp 10 Tonnen haben, ist das für die Bergung notwendige Material hier am See nicht verfügbar. Der Gutachter hat daraufhin ein Team der Wasserrettung Nußdorf am Attersee in Oberösterreich mit dem Heben der "Sir Shackleton" beauftragt. Die Bergung des Schiffes fand am 23.08.2020 statt. Rund 30 Einsatzkräfte waren mit insgesamt 7 Booten von ca. 09:00 Uhr ca. 17:30 Uhr an der Unfallstelle im Einsatz. Die Bergung war kompliziert. Die Taucher aus Österreich mussten zum Teil blind am Schiff arbeiten, da aufgewirbelter Schlick die Sicht auf 0 reduzierte. Um ca. 16:00 Uhr tauchte das Schanzkleid aus dem Wasser auf und um ca.16:45 Uhr konnten wir selbst an Bord und haben die Flagge der "Sir" an der Saling gehisst. Nachdem das Schiff ausgepumpt und stabilsiert wurde, nahm es die Wasserwacht Dießen in Schlepp und brachte es nach Utting zur Steinlechner Bootswerft, die unser Schiff seit 2011 betreut. Um das Gewicht auf dem Slipwagen zu reduzieren, musste die "Sir" noch im Wasser komplett leergeräumt werden. 4 Helfer der Werft und die Besatzung des Wasserwachtbootes aus Dießen haben kräftig angepackt und nach einer Dreiviertelstunde rollte die "Sir" auf dem Slipwagen in das Werftgelände. Das Schiff wurde dort in den folgenden Tagen professionell gereinigt und begutachtet. Die Ursache des Untergangs war ein gebrochenes Teil der Kühlwasserversorgung. Die Schäden vor allem im Innenbereich sind erheblich. Wie es nun mit der "Sir Shackleton" weitergeht, wird sich in den nächsten Tagen ergeben. Wir halten Euch hier in jedem Fall auf dem Laufenden. This episode was recorded at the Micam shoe fair in Milan, Italy, back in February. I sat down with Richard Utting, Export Manager at Loake, the famous British manufacturer of Goodyear welted shoes. Loake was a brand that, contrary to many other Northampton based companies, first got big on the domestic market and then went on to grow internationally. Richard Utting has been a large part of this journey, and we had a very interesting talk about everything from how to succeed internationally and why it’s beneficial to be on different markets, to how brand building has increased importance also for makers of classic shoes and of course about Brexit.
