On July 5, 2000, Ricardo Aguilera was shot and wounded in a gang related drive-by shooting in Los Angeles, California. Several witnesses identified 25-year-old Rafael Madrigal Jr. in a photo lineup as either the shooter or driver of the car involved. Those witnesses testified against Rafael at trial. Rafael, who maintained his innocence throughout the ordeal, had been at work at Proactive Packaging, a 50-minute drive away, at the time of the shooting. A co-worker could have confirmed his alibi, and his boss could have testified that he was certain Madrigal was at work because he was the only one who knew how to operate one of the machines in the production line. But Rafael’s defense attorney only called a single co-worker to the stand at the trial, and did not present a recording of Rafael’s co-defendant admitting that Rafael was not involved. On January 18, 2002, a jury convicted Rafael of attempted murder and he was sentenced to 53 years to life in prison. https://www.wrongfulconvictionpodcast.com/with-jason-flom Wrongful Conviction is a production of Lava for Good™ Podcasts in association with Signal Co. “¿Por qué esta gente busca una señal?”Del santo Evangelio según san Marcos: 8, 11-13Lectura y reflexión: Pbro.

RNA Therapies and mRNA Challenges: Industry Shifts

Good morning from Pharma Daily: the podcast that brings you the most important developments in the pharmaceutical and biotech world. Today, we delve into a series of transformative events shaping the landscape of drug development, regulatory scrutiny, and corporate strategies. At the forefront is Madrigal Pharmaceuticals' strategic acquisition of Ribo Therapeutics' preclinical siRNA programs, valued at $4.4 billion. This move aims to fortify Madrigal's liver disease drug portfolio alongside its promising candidate, resmetirom. By expanding into RNA-based therapies, Madrigal highlights an industry trend focused on gene silencing techniques to target genetic diseases more precisely.Turning to Moderna, it faces a regulatory hurdle as the FDA issued a refusal-to-file letter for its mRNA-based flu vaccine. The regulator's concerns about the trial design, specifically the use of a licensed standard-dose seasonal influenza vaccine as a control arm, emphasize the complexities of advancing mRNA technologies beyond COVID-19 applications. This situation underscores the necessity for meticulous trial designs that align with evolving regulatory standards. In cell therapy, allogeneic CAR-T treatments are gaining attention as companies strive to make these therapies more accessible by using T cells from healthy donors, rather than modifying a patient's cells. Despite technical challenges like graft-versus-host disease and immune rejection, these treatments promise streamlined manufacturing and reduced costs, marking a significant evolution from the pioneering autologous CAR-T success with Emily Whitehead in 2012. Eli Lilly's entry into CAR T-cell therapy through a $2.4 billion acquisition of Orna represents an ambitious expansion into autoimmune therapies. This strategic move exemplifies a broader trend where companies diversify portfolios to include emerging therapeutic modalities promising transformative impacts on patient care.In respiratory medicine, Upstream Bio's phase 2 trial of its TSLP receptor agonist showed encouraging results in reducing asthma exacerbations, comparable to Tezspire. However, falling short of best-case scenarios leaves room for competitors to present more compelling data. This illustrates the competitive nature of asthma treatment development and the ongoing quest for superior therapeutic options.A critical regulatory update arises from the NIH's decision to halt the Xarelto arm of a stroke prevention trial due to safety concerns. This decision highlights the indispensable role of independent monitoring committees in ensuring patient safety and meaningful clinical trial outcomes. On the corporate front, AstraZeneca has articulated an ambitious goal to achieve over 25 blockbuster drugs by 2030 as part of its strategy to reach $80 billion in revenue. This vision underscores the importance of innovation and strategic planning in sustaining growth within an increasingly competitive market.Fujifilm Biotechnologies' completion of its £400 million expansion project in the UK is another notable development, signaling robust investment in antibody production capabilities. This expansion positions Fujifilm as a key player in biopharmaceutical contract manufacturing and underscores the growing demand for flexible production technologies. The biotech sector is also witnessing significant activity with Pelage making strides in addressing hair loss through promising candidate developments. The market's enthusiasm for innovative solutions beyond traditional treatments reflects a broader demand for cutting-edge approaches to longstanding medical challenges. In obesity treatment, Novo Nordisk and Eli Lilly continue to lead with notable advancements. Novo Nordisk's recent developments with its Wegovy pill have been positively received, yet analysts question if this will suffice in maintaining their competitive edge given the dynamic nature of this therapeutic areSupport the show

Gloria a ti, Señor Jesús.

Send us a textTwo Amaretto Sours and a wild idea spark a full-blown caper: what if Disney characters ran a classic mafia? We pour, we taste, and we build an underworld—with Boss Mickey at the top, a heated consigliere debate (Minnie's steel vs Goofy's chill), and capos assigned to every park. It's satire, it's structure, and it's strangely satisfying as we map crews, enforcers, and “fronts” that fit each park's energy. We start at the bar, comparing a scratch Amaretto Sour to a Bartesian version and unpack why an amaretto-forward balance works even for bourbon skeptics. Then we lay out the family tree. Disneyland gets old-school charm with Roger or Oswald and a transportation racket that kicks tribute upstairs. DCA runs on engines and muscle—Lightning or Luigi in charge, with Mr. Incredible and Spider-Man tightening the bolts. Magic Kingdom becomes a jewel operation with the Seven Dwarfs and a sly cash wash through comedy clubs, while Donald or Carousel John keep order in their own unforgettable ways.Things turn shadowy at Hollywood Studios where Scar, Darth Vader, and Loki bring enforcement and intrigue. Kermit offers a gentler alternate leadership style, backed by a toy box crew that improvises for the greater good. Epcot scales up with Figment's unpredictability or Scrooge's iron ledger, calling in Remy, Baymax, Elsa, the Three Caballeros, the Phoenicians, and even Patrick Warburton's legendary Soarin' voice for intimidation by tone alone. Animal Kingdom closes strong with Judy Hopps' discipline, Mufasa's authority, and the Madrigal family's specialized “import/export” skill set.Beyond the laughs, there's a real takeaway on leadership and systems: the right roles, trusted lieutenants, and a balance of force and finesse can run any kingdom—imagined or real. Pour a glass, press play, and then tell us who your Disney underboss would be.Enjoyed the ride? Follow the show, leave a review, and share this episode with a Disney fan who loves a clever twist.

Good morning from Pharma Daily: the podcast that brings you the most important developments in the pharmaceutical and biotech world. Today, we delve into a series of transformative updates reshaping drug development and patient care across the globe.Starting with a major collaboration between Roche and Medilink Therapeutics that has captured industry attention. Roche is making a strategic move by committing $570 million to partner on an antibody-drug conjugate, or ADC, targeting the B7-H3 immune checkpoint protein. This partnership underscores a broader industry trend focusing on immuno-oncology. ADCs have become pivotal due to their ability to deliver cytotoxic agents precisely to tumor cells, reducing systemic exposure and minimizing side effects. This precision not only enhances efficacy but also improves patient experience, marking a significant stride in cancer treatment modalities. In another strategic alliance, Pfizer has entered into a $50 million deal with Madrigal Pharmaceuticals for a DGAT2 inhibitor. Madrigal plans to pair this with its liver disease treatment, Rezdiffra, aiming to amplify therapeutic outcomes in liver conditions. This highlights an increasingly popular approach in medicine: polypharmacy and combination therapies. By tackling diseases from multiple angles, these therapies promise more comprehensive management of complex conditions, reflecting a shift towards more personalized and effective treatment strategies.Turning our attention to the startup landscape, "Baby KJ" Scientist has launched a new personalized CRISPR therapy company with $16 million in initial funding from Menlo Ventures. This venture signals growing interest in CRISPR technology for crafting bespoke genetic therapies. The potential for CRISPR lies in its ability to edit genomes precisely, opening possibilities for treating genetic disorders at their root cause and tailoring interventions to individual patients' genetic profiles. On the financial front, Aktis Oncology's successful IPO stands out, raising an impressive $318 million through an upsized offering. This achievement not only illustrates investor confidence in biotech but also suggests a more favorable atmosphere for upcoming biotech ventures seeking public funding. A robust financial ecosystem is crucial for fostering innovation and bringing cutting-edge therapies from the lab bench to the bedside. In another exciting development, Airnexis Therapeutics has secured $200 million to advance its COPD treatment asset in collaboration with a Chinese pharmaceutical firm. The total deal could reach up to $955 million in what's known as biobucks—a term used for milestone payments in biotech partnerships. This collaboration exemplifies the globalization of biotech partnerships, emphasizing the strategic importance of tapping into diverse markets for drug development and commercialization.Meanwhile, Ollin Biosciences is making waves with promising results from its bispecific antibody trial. The antibody showed superior efficacy in treating diabetic macular edema compared to Genentech's Vabysmo. This success potentially sets a new benchmark for retinal disease therapies and highlights the rapid evolution of bispecific antibodies. These agents can engage multiple targets simultaneously, offering enhanced therapeutic potential across various conditions.Leadership dynamics are also influencing the sector. Charles Fuchs has transitioned from his role as Roche's Global Head of Oncology and Hematology Product Development to Tubulis. Such moves suggest potential shifts in strategic focus for both companies involved, reflecting broader trends in leadership realignments within the industry. Additionally, Illumina's appointment of Eric Green as Chief Medical Officer showcases another instance of experienced leaders taking pivotal roles within companies at the forefront of genomic research. Such appointments underscore the imSupport the show

We share an easy home recipe and talk flavor: how vanilla rounds tart fruit, why the color skews blush instead of neon, and simple tweaks for batching or going zero-proof without losing the festive mood. From there, the sleigh takes off. Snow White harmonizes with forest friends and accidentally bakes forty pies. Cinderella observes midnight rituals in France and hosts a slipper-chic party after a wrapping spell goes wild. Pocahontas crafts light from berries and river stones. Mulan battles snarled lights like a seasoned general. Rapunzel becomes the tinsel queen and gifts lanterns under winter skies. Our tour lands in Polynesia with Moana's outrigger Santa and Maui's fire-roasted feast, then dives undersea for Ariel's clam-shell gifts and under-the-sea carols. Send us a textIn June, the Fatty Liver Foundation officially launched the Stephen A. Harrison Patient Advocacy program and announced its first class of 20 Fellows. In today's conversation, Louise Campbell and Roger Green interview the founders to learn what motivated them to launch this program, and we meet five of the initial Fellows. Program Co-ordinator Elena Samsonova opens the conversation by celebrating last week's onboarding of 20 inaugural Fellows and the chemistry between them. She goes on to introduce five of them:Steven Rodrigues (ALD, UK citizen) shares his recent experience giving a 15-minute national interview on BBC Radio 5 discussing ALD clinical insights. Silvana Lesidrenska (Hepatitis B, Bulgaria) shares her desire to learn more about SLD as part of her advocacy leading a liver patients association in Bulgaria. Julie Peyout (MASH, Canada) is a great-grandmother who was recently found free after therapy for Non-Hodgkin's Lymphoma. Pam Miller (MASH cirrhosis, US) learned 24 years ago that she had MASH cirrhosis when her surgeon aborted a bariatric procedure. Since then, she has lived with cirrhosis and experienced hepatic encephalopathy (HE). She has participated in clinical trials, lobbied legislators, and collaborated with professional societies. Melanie Smith  (MASH cirrhosis, US) has lived with stage-4 MASH cirrhosis since age 36 (sixteen years ago) and experiences hepatic encephalopathy (HE). She has led support groups for over a decade. Next, Fatty Liver Foundation President Wayne Eskridge recounts the program's foundational story. He describes Stephen as a man with a genuine commitment to patients and his passing as a "tremendous loss." This led Wayne to discuss this idea with Rebecca Taub of Madrigal, "another one of my heroes." (Madrigal supports this program.) Elena describes how she joined the FLF to lead this program. Roger asks the Fellows where and how they intend to make a difference. Their goals vary, from working in a Biopharma company advocating for patients to "going to the top," perhaps even lobbying the HHS Secretary, to expanding their own education and knowledge about the various forms of SLD.

Their goals vary, from working in a Biopharma company advocating for patients to "going to the top," perhaps even lobbying the HHS Secretary, to expanding their own education and knowledge about the various forms of SLD.  00:34:46 - Part II: Goals of the Harrison Fellowship ProgramThis conversation focuses on the program's goals and scope for its first year. Wayne points out that, while the FLF originally focused on NAFLD and NASH, the new nomenclature has led them to broaden their focus to all forms of SLD. (He plans to rebrand as the Steatotic Liver Foundation.)Elena, it describes the program's four "pillars": Mentorship - Having an experienced advocate guide each Fellow as they pursue the next steps as Advocates.Education - Learning more about SLD.Networking - Connecting with other advocates and industry decision-makers at conferences.Fellowship - Supporting each other as they grow new skills and have new experiences. 00:55:09 - Experts: Louise and Roger Discuss This RoundtableRoger and Louise share their enthusiasm for the unprecedented sense of empowerment they felt among patient advocates while leading this discussion. Both comment on the identity shift from “MASH patient” to “person living with MASH.” Louise mentions that doctors or APPs still minimize disease by characterizing MASLD as “a bit of fat” that requires no action. Roger suggests that focusing on the total patient will include not only the efficacy of medicines but also potential side effects or safety concerns. Louise comments that advanced disease and cirrhosis occur most often in post-menopausal women with cirrhosis, and discusses implications. Both foresee a growing “army” of advocates that can drive earlier detection, smarter trial design, and better everyday care.01:11:11 - Business Report

Take a minute to get the scoop!Fun FactIn Weelkes' madrigal "As Vesta Was from Latmos Hill Descending", the word “descending” is literally sung with downward scales—meanwhile, “ascending” climbs right back up. Even “running down” gets a rapid, breathless passage. It's one of the earliest—and cheekiest—examples of word painting pushed to delightful extremes.About Steven, HostSteven is a Canadian composer & actor living in Toronto. On March 25, I interviewed Alexis Madrigal and Noni Session in front of a sold out crowd at Spire in West Oakland. Madrigal is the author of an essential new book called “The Pacific Circuit: A Globalized Account of the Battle for the Soul of an American City.” He is also host of KQED's Forum, a longtime journalist, and a dear friend. Noni Session is a third generation West Oaklander and the executive director of East Bay Permanent Real Estate Cooperative, a community-funded organization that challenges displacement with cooperative economic strategies. Through her groundbreaking work with EB PREC, Session is rebuilding local institutions, such as Esther's Orbit Room, and also removing housing from the speculative market to create permanently affordable, community-controlled homes. Our discussion explored themes that connect Madrigal's book and Session's revitalization projects, the history of West Oakland's role in the global economy, and much more.
