Abstract: Pavlovian conditioning is thought to involve the formation of learned associations between stimuli and values, and between stimuli and specific features of outcomes. Here we leveraged human single neuron recordings in ventromedial prefrontal, dorsomedial frontal, hippocampus and amygdala neurons while patients performed a sequential Pavlovian conditioning task containing both stimulus-value and stimulus-stimulus associations. Neurons in the ventromedial prefrontal cortex encoded predictive value along with the amygdala, but also encoded predictions about the identity of stimuli that would subsequently be presented, suggesting a role for neurons in this region in encoding predictive information beyond value. Unsigned error signals were found in dorsomedial prefrontal areas and hippocampus, potentially supporting learning of non-value related outcome features. Our findings implicate distinct human prefrontal and medial temporal neuronal populations in mediating predictive associations which could partially support model-based mechanisms during Pavlovian conditioning. Abstract: Extinction is a fundamental form of inhibitory learning that is important for adapting to changing environmental contingencies. While numerous studies have investigated the neural correlates of extinction using Pavlovian fear conditioning and appetitive operant reward-seeking procedures, less is known about the neural circuitry mediating the extinction of appetitive Pavlovian conditioned responding. Here, we aimed to generate an extensive brain activation map of extinction learning in a rat model of appetitive Pavlovian conditioning. Male Long-Evans rats were trained to associate a conditioned stimulus (CS; 20 s white noise) with the delivery of a 10% sucrose unconditioned stimulus (US; 0.3 ml/CS) to a fluid port. Control groups also received CS presentations, but sucrose was delivered either during the inter-trial interval or in the home-cage. After conditioning, 1 or 6 extinction sessions were conducted in which the CS was presented but sucrose was withheld. We performed Fos immunohistochemistry and network connectivity analyses on a set of cortical, striatal, thalamic, and amygdalar brain regions. Neural activity in the prelimbic cortex, ventral orbitofrontal cortex, nucleus accumbens core, and paraventricular nucleus of the thalamus was greater during recall relative to extinction. Conversely, prolonged extinction following 6 sessions induced increased neural activity in the infralimbic cortex, medial orbitofrontal cortex, and nucleus accumbens shell compared to home-cage controls. All these structures were similarly recruited during recall on the first extinction session. These findings provide novel evidence for the contribution of brain areas and neural networks that are differentially involved in the recall versus extinction of appetitive Pavlovian conditioned responding. In today's episode I take a closer look at Respondent Conditioning, also known as Classical Conditioning or Pavlovian Conditioning. I really tried my best to make it simple, but the amount of times I said the words "conditioned, unconditioned, stimulus, and response" started to make my head spin! Today's main focus was on Respondent Conditioning with just a sprinkle of Operant Conditioning, and in my next episode I will pick up where I left off and go into more details about Operant Conditioning and review reinforcement and punishment. Maneesh Sethi is the founder and CEO of Pavlok (https://pavlok.com/), an incredibly interesting behavioural modification tool that's been used by thousands to quit bad habits like smoking, biting fingernails, over-eating, getting out of bed late, and so much more. Based on the theory of Pavlovian Conditioning put forward by Russian physiologist Ivan Pavlov, it works on the principle that some of our deeply conditioned behaviours can be quite easily changed with a mild shock, or tap to the wrist, where the Pavlok device is worn. For biohackers, changing our behaviour is a big deal, and something that most high-performers want to implement more in our lives, to override old programming, negative habits, and everything in between. And uh, if you make it through this episode, you will be able to answer this riddle. So here it goes. You're in a room. Let's say it's a pretty small room with two people. One is a baby and the other is an adult, let's say it's a man and he doesn't speak English and you don't speak his language. Now he's holding a basketball and you want to teach him the meaning of the words “drop it.” Now, so just to recap, we have three people in the room, you, a baby, a man who doesn't speak English and is holding a basketball and that's it. There's no dictionary that you can use to translate. There's no dry erase board that you can write a diagram on and you are not allowed to really do any kind of a miming in order to try and get him to drop it. You also cannot touch him. So that's the setup. What are you going to do? Today's episode is specifically about teaching “drop it.” Of course we're going to focus on how to teach “drop it” to a dog, but teaching “drop it” to a dog is a little bit like teaching “drop it” to someone who doesn't speak your language and we're also going to talk about “come.” Now I know at a first glance it might not seem like these two behaviors have very much in common, but, again, by the end of this episode I think you will see what they do have in common and why I wanted to pair these two very important behaviors together. Now before I get too much into the nitty gritty of teaching these two important behaviors, I want to do a little review of the two kinds of learning that I'm always talking about on this show and that are so important to dog training: operant and conditioning and classical conditioning. So operant conditioning is what we tend to think about when we're thinking about dog training. Operant conditioning is really anything that fits into like a “if then” equation. If my dog's butt hits the ground, then I'll give him a treat. Or if my dog pulls, then we will stop walking. Of course, humans are very much affected by operant conditioning too, just think of everything in your life that you can put into this a kind of equation, right? Domjan, Michael – Pavlovian Conditioning: It Is Not About the CR, But About Modification of a Behavioral System- Pavlovian conditioning is typically thought of in terms of the common example of a dog salivating to a cue that occurs reliably before the delivery of meat powder. Conditioned salivation (the conditioned response or CR) was viewed ad reflecting an association of the cue and the meat powder, and Pavlovian conditioning became a favorite method of scientists whose primary interest was to elucidate the mechanisms of association learning. Investigators worked hard to find situations in which the conditioned response was convenient to measure and occured reliably. They focused on iidentifying experimental conditions which would predictably increase or decrease the target CR. I will argue that this focus on a target conditioned response misses the broader biological significance of Pavlovian conditioning, which is to enable organisms to interact more effectively with significant biological events or unconditioned stimuli (Uss). The broader perspective suggests that Pavlovian conditioning, produces a wide range of behavioral and physiological adjustments that enable the orgsnism not only to better prepare for the impending occurence of the unconditioned stimulus but to also deal with the US more effectively at both the behavioral and physiological level. Thus, Pavlovian conditioning produces a reorganization of the biobehavioral system that is activated by the US. This broader perspective will be illustrated with examples from appetitive, aversive, and sexual conditioning.

Stout, Steven – Cue Competition in Pavlovian Conditioning - In recent decades researchers in the field of Pavlovian conditioning have focused on how conditioned responding to a target conditioned stimulus (CS) is affected by the presence of non-target CSs. A common observation is that target and non-target CSs compete for control over conditioned responding in the sense that their response potentials are in-verse1v correlated. In the three and a half decades since the theoretical model of Rescorla and Wagner inspired a wealth of research into cue competition, investigators have uncovered a number of interesting empirical regularities. Unfortunately, the dissemination of these regularities to a wider community outside associative learning circles has been obscured by the tendency of Pavlovian investigators to discuss their research in a heavily theory-laden language. The purpose of this tutorial is to introduce undergraduates to the field of cue competition who have been otherwise put off by constructs sue: as positive and negative associations, memorial representations, and comparator processes. In particular, I will consider what happens to conditioned responding when non-target CSs are presented before, interspersed among, or after the target CS-US pairings, and whether those non-target CSs are discrete or contextual. Conditions under which cue competition, or its opposite, cue facilitation, are observed will be discussed.

An understanding of this learning process requires analyses of the necessary conditions for learning, the contents of that learning, and how learning is expressed in performance. Abstract: The ventral pallidum (VP) is a major hub interfacing striatopallidal and limbic circuits, conveying information about salience and valence crucial to adjusting behavior. However, how neuron populations of the VP with different firing properties represent these variables is not fully understood. Therefore, we trained mice on auditory Pavlovian conditioning and recorded the activity of VP neurons while mice were performing the task. Many VP neurons responded to punishment (51%) and reward (44%), either by firing rate increase or decrease. Additionally, 20% of cells responded to outcome-predicting auditory stimuli, showing larger responses to reward-predicting cues compared to those that signaled likely punishment. We found that a large subset of VP neurons showed burst firing based on their auto-correlograms, while a small population exhibited fast rhythmic discharge in the beta/gamma frequency range. Some bursting neurons exhibited distinct response properties of their bursts and single spikes, suggesting a multiplexed coding scheme in the VP. However, non-bursting, non-rhythmic neurons were the most sensitive to reward and punishment. Finally, we demonstrate the presence of synchronously firing neuron assemblies in the VP. Neurons participating in such assemblies were particularly responsive to reinforcing stimuli. This suggests that a synchronous, non-bursting, non-rhythmic neuron population of the VP is responsible for the lion's share of ventral pallidal salience representation, likely important for reinforcement learning.
