Ann Kennedy is Associate Professor at Scripps Research Institute and runs the Laboratory for Theoretical Neuroscience and Behavior. Among other things, Ann has been studying how processes important in life, like survival, threat response, motivation, and pain, are mediated through subcortical brain areas like the hypothalamus. She also pays attention to the time course those life processes require, which has led her to consider how the expression of things like proteins help shape neural processes throughout the brain, so we can behave appropriately in those different contexts. You'll hear us talk about how this is still a pretty open field in theoretical neuroscience, unlike the historically heavy use of theory in popular brain areas throughout the cortex, and the historically narrow focus on spikes or action potentials as the only game in town when it comes to neural computation. We discuss that and I link in the show notes to a commentary piece Ann wrote, in which she argues for both top-down and bottom-up theoretical approaches. I also link to her papers about the early evolution of nervous systems, how heterogeneity or diversity of neurons is an advantage for neural computations, and we discuss a kaggle competition she developed to benchmark automated behavioral labels of behaving organisms, so that despite different researchers using different recording systems and setups, analyzing those data will produce consistent labels to better compare across labs and aggregated bigger and better data sets. Dr. Beth Stroupe is a Professor in the Department of Biological Science and the Institute of Molecular Biophysics at Florida State University (FSU). She is also Director of the Molecular Biophysics Graduate Program there. Research in Beth's lab focuses on understanding what molecules and proteins look like. This is important for better understanding how these molecules work, both from a basic science perspective and with an eye towards controlling molecules for clinical applications. They use a variety of different techniques that allow them to visualize these very tiny molecules. When she's not working, Beth and her partner enjoy spending quality time with their two cats, as well as growing black-eyed peas and other vegetables in their garden, kayaking, hiking, and exploring the great outdoors. Beth completed her undergraduate training at Wake Forest University where she majored in chemistry and minored in music. She was awarded her PhD in biochemistry from the Scripps Research Institute. Afterwards, Beth conducted postdoctoral research at Brandeis University before joining the faculty at FSU where she is today. In our interview, Beth shares more about her life and science.

Vidcast:  https://www.instagram.com/p/DJHoaqqROQk/An FDA-approved psoriasis drug turns help those addicted to alcohol turn the corner.  That drug is apremilast, a phosphodiesterase 4 inhibitor sold under the brand name Otezla.Preclinical testing of the drug at San Diego's Scripps Research Institute using mouse lines that are prone to over consume alcohol shows that apemilast not only reduces craving for the drink but also lessens a characteristic accompanying heightened pain sensitivity known as mechanical allodynia.  These beneficial effects occurred in both female and male mice.Seeking to explain the drug's effects, the investigators found that apemilast increases signaling by the amino acid neurotransmitter GABA, gamma-aminobutyric acid.  GABA is known to slow down and calm brain activity thereby reducing stress, anxiety and fear.  This drug is already FDA-approved for psoriasis and psoriatic arthritis nd that's a plus for getting it into the clinic for curbing alcoholism.The next step will be clinical trials of apemilast.  Once completed, this drug may well help those with alcohol use disorder kick the habit and suppress the heightened pain response that accompanies it.https://insight.jci.org/articles/view/189732https://www.sciencedaily.com/releases/2025/04/250422155830.htm#apremilast #otezla #alcohol #aud #pain

In 2022, the Nobel Prize in Chemistry went to Dr. Carolyn Bertozzi of Stanford University, Dr. Morten Meldal of the University of Copenhagen, and Dr. K. Barry Sharpless of the Scripps Research Institute “for the development of click chemistry and bioorthogonal chemistry.” In “click chemistry,” molecular building blocks snap together quickly and efficiently to let chemists build more complicated molecules. But bioorthogonal chemistry takes that work one step further, allowing the technique to be used within living organisms without damaging cells.“When someone is thinking outside the box, or in a very different way, we like to think of that as orthogonal thinking,” Bertozzi explained. “So biorthogonal means not interacting with biology. Totally separate from biology.” Her research began with an interest in developing ways to see specific sugar molecules on the surface of cells. Dr. Karine Gaelle Le Roch is a Professor in the Department of Molecular, Cell and Systems Biology and Director of the Center for Infectious Disease and Vector Research at the University of California, Riverside. Karine's research focuses on parasites, and much of her work has focused on the parasite that causes malaria in humans. She is working to better understand the life cycle of the parasite, how the molecular components in its body work together, and how the parasite's function can be disrupted after a person has been infected in order to treat and cure malaria. When she's not doing science, Karine enjoys sailing in the San Diego Bay, skiing in the winter, spending time with her son, and watching him play soccer. She received her BA in Biochemistry and Microbiology from the University Paris-Sorbonne, her MS in host-parasite interactions between Oxford University in the UK and the University of Lille in France, and her PhD in Parasitology and host-parasite interactions from the University Paris-Sorbonne. Afterwards, she conducted postdoctoral research at The Scripps Research Institute in La Jolla and subsequently she was an Institute Fellow at the Genomics Institute of the Novartis Research Foundation. Karine joined the faculty at UCR in 2006. She has earned various honors and awards throughout her career, including the Academic Senate Regent Award for Outstanding Research from the University of California, she was a participant in the Provost Faculty Leadership Program at UCR and the University of California's Women's Initiative for Professional Development. Published in the June 2023 issue of the journal JCI Insight, the study adds to growing preclinical evidence that the glucagon-like peptide-1 (GLP-1) system plays a role in alcohol and other substance use disorders and that GLP-1 receptor agonists show potential for treating people with alcohol use disorder (AUD). GLP-1 is a gut hormone that stimulates insulin secretion after eating. This promotes a feeling of fullness, helps regulate blood sugar, and reduces hunger cravings. Semaglutide and other GLP-1 agonists—medications that bind to GLP-1 receptors and mimic their effects—are currently used as treatments for diabetes and obesity. “Parts of the brain that drive eating behaviors overlap extensively with the drive to use alcohol or other substances,” explained Lorenzo Leggio, M.D., Ph.D., and Leandro Vendruscolo, Pharm.D., Ph.D., two of the study's senior authors. They added that there is also an overlap between the brain mechanisms that regulate overeating and those that contribute to the development and maintenance of substance use disorders, including AUD. Dr. Stacey Harmer is a Professor of Plant Biology in the College of Biological Sciences at the University of California, Davis. Stacey studies different biological rhythms and the circadian clocks within organisms that create and maintain those rhythms. Circadian clocks can be found in various organisms, from bacteria to people. In particular, Stacey is interested in understanding why plants have circadian clocks, how these clocks work in plants, and what aspects of physiology and development these clocks control. Stacey likes to take her mind off research when she's not in the lab by doing yoga and road biking. She and her husband also enjoy cooking and eating their delicious kitchen creations. She received her BA in Biochemistry from the University of California, Berkeley and was awarded her PhD in Biochemistry from the University of California, San Francisco. Afterwards, Stacey conducted postdoctoral research at the Scripps Research Institute in La Joya before joining the faculty at UC, Davis where she has been since 2002. She is the recipient of the American Society of Photobiology's New Investigator Award and was selected as a Chancellor's Fellow at UC Davis. Stacey is with us today to share her exciting experiences in life and science.
