We eat for a variety of reasons, most importantly to sustain life. Unsurprisingly, the decisions we make about what we consume can have a huge impact on our health. Equally important is how our bodies eliminate the waste created by what we ingest. After all, what goes in must come out. Are there hard and fast rules about what constitutes a healthy …let's just say it, poo? Sean Gibbons, a microbiologist at the Institute for Systems Biology in Seattle, joins The Excerpt to discuss what bowel movements, or a lack of them, reveal about our health. Over the past 15-20 years, Peter has produced so much groundbreaking research in the endlessly fascinating field of the gut microbiome. His work is not limited to DMPK by any means, but in this episode, we discuss drug metabolism by gut bacteria and its impact on pharmacokinetics. Clinically, reduced efficacy and/or increased side effects observed in some patients can be attributed to the gut microbiome, including narrow therapeutic index drugs such as digoxin. You will come away with an appreciation for scientific creativity and the sometimes unexpected benefits of doing an experiment that everyone thinks is going to fail. We speculate on how much there is to learn about the possible interactions between the gut microbiome and drugs for obesity, including GLP1 agonists. Finally, we talk about some practical considerations in terms of studying bacterial drug metabolism in vitro. The episode explores the following: How important is the gut microbiome in terms of drug metabolism? What do we know about the balance between control of the microbiome by the host and control of the host by the microbiome? What are some of the challenges of studying gut bacterial drug metabolism in vitro? Could gut bacteria play a role in the efficacy and/or side effects of GLP1 agonists such as semaglutide, used to treat obesity and diabetes? Speaker:Peter J. Turnbaugh – Professor, Department of Microbiology & Immunology at the University of CaliforniaPeter J. Turnbaugh, Ph.D., is a Professor in the Department of Microbiology & Immunology, the G.W. Hooper Research Foundation, and the Benioff Center for Microbiome Medicine at the University of California, San Francisco. He is also a CZ Biohub-San Francisco Investigator. For the past two decades, his research has focused on the metabolic activities of the trillions of microbes that colonize our adult bodies. Dr. Turnbaugh and his research group use interdisciplinary approaches in preclinical models and human cohorts to study how the gut microbiome influences nutrition and pharmacology. He received a B.A. in Biochemistry, Biophysics, and Molecular Biology from Whitman College and a Ph.D. in Microbial Genetics and Genomics from Washington University in Saint Louis. From 2010-2014 he was a Bauer Fellow in the FAS Center for Systems Biology at Harvard University, where he established an independent research group before starting his faculty position at the University of California, San Francisco. It's always exciting to see flashy headlines about the next revolutionary drug approved by the FDA or how a biotech startup is making millions for a diagnostic test. However, it's equally important to not lose sight of what gets us to the most important discoveries in the first place: basic research. Today, we're interviewing Dr. Marc Kirschner, a renowned cell biologist and biochemist known for his research on the cytoskeleton, cell cycle, and embryo signaling. Join us to hear his thoughts on how scientific disciplines have become more intertwined over time, the crucial role of technological advancements such as electron microscopy in the field of biochemistry, and the dire need for approaching scientific discovery with curiosity instead of going straight for the money. Dr. Marc Kirschner is the founding chair of the Department of Systems Biology at Harvard Medical School and is known for major discoveries in cell and developmental biology. He has also coauthored 2 books on the biological basis of evolution and helped launch the peer-reviewed journal PLOS (Public Library of Science) Biology.

We talk about modeling in science, the theoretical foundations of social science, mathematical models and agent-based models, and fine-grained and coarse-grained models. We discuss assumptions about human psychology, and we then explore examples of social dynamics that can be modeled, like contagion and the spread of innovation; opinion dynamics, and consensus and polarization; cooperation; norms; and science as a social phenomenon. Finally, we discuss how to turn ideas into models, and the limitations of models. -- A HUGE THANK YOU TO MY PATRONS/SUPPORTERS: PER HELGE LARSEN, JERRY MULLER, BERNARDO SEIXAS, ADAM KESSEL, MATTHEW WHITINGBIRD, ARNAUD WOLFF, TIM HOLLOSY, HENRIK AHLENIUS, FILIP FORS CONNOLLY, ROBERT WINDHAGER, RUI INACIO, ZOOP, MARCO NEVES, COLIN HOLBROOK, PHIL KAVANAGH, SAMUEL ANDREEFF, FRANCIS FORDE, TIAGO NUNES, FERGAL CUSSEN, HAL HERZOG, NUNO MACHADO, JONATHAN LEIBRANT, JOÃO LINHARES, STANTON T, SAMUEL CORREA, ERIK HAINES, MARK SMITH, JOÃO EIRA, TOM HUMMEL, SARDUS FRANCE, DAVID SLOAN WILSON, YACILA DEZA-ARAUJO, ROMAIN ROCH, DIEGO LONDOÑO CORREA, YANICK PUNTER, CHARLOTTE BLEASE, NICOLE BARBARO, ADAM HUNT, PAWEL OSTASZEWSKI, NELLEKE BAK, GUY MADISON, GARY G HELLMANN, SAIMA AFZAL, ADRIAN JAEGGI, PAULO TOLENTINO, JOÃO BARBOSA, JULIAN PRICE, EDWARD HALL, HEDIN BRØNNER, DOUGLAS FRY, FRANCA BORTOLOTTI, GABRIEL PONS CORTÈS, URSULA LITZCKE, SCOTT, ZACHARY FISH, TIM DUFFY, SUNNY SMITH, JON WISMAN, WILLIAM BUCKNER, PAUL-GEORGE ARNAUD, LUKE GLOWACKI, GEORGIOS THEOPHANOUS, CHRIS WILLIAMSON, PETER WOLOSZYN, DAVID WILLIAMS, DIOGO COSTA, ALEX CHAU, AMAURI MARTÍNEZ, CORALIE CHEVALLIER, BANGALORE ATHEISTS, LARRY D. Starting at the young age of 10, porn became a habitual escape for me. Over time, I understood that it failed to provide lasting satisfaction, instead wiring my brain for lust. Your gut microbiome consists of trillions of microbiota and is a critical health determinant, affecting your immune system, mood, energy level, and much more. As a scientific field, microbiome research is new to the scene, but the intricate relationship between our gut and our overall health is clear – and getting clearer. In April, Netflix started streaming Hack Your Health, an informative documentary about the gut microbiome, gut health, and the science of eating. In this collaborative event between Town Hall Seattle and the Institute for Systems Biology, Hack Your Health Director Anjali Nayar will sit down with gut microbiome specialist Dr. Sean Gibbons, a scientific advisor on the film, to discuss the project, interesting developments in microbiome research, and much more. Anjali Nayar is an Indian-Canadian director, former climate scientist, and tech founder. Anjali's newest film, a Netflix Original called Hack Your Health: The Secrets of Your Gut is streaming on Netflix, and her fantasy short Closer has over 5 million views and won the 2022 Prism Prize Audience Award (Canada's top music video awards). As of 2024, she is developing a slew of scripted projects and a series with the Golden State Warriors. Her prior films have been supported by Cinereach, Sundance, and Tribeca, won countless awards, jury prizes, gone theatrical, and been acquired by Netflix and Amazon. Sean Gibbons, Ph.D., is associate professor at Institute for Systems Biology. He received his PhD in biophysical sciences from the University of Chicago in 2015, winning a prestigious EPA STAR Graduate Fellowship. He completed his postdoctoral training in the Department of Biological Engineering at MIT and The Broad Institute in 2018. He joined ISB as Washington Research Foundation Distinguished Investigator and assistant professor in 2018. His research on the human microbiome has been published in top scientific journals, including Nature, Science, and Cell. Presented by Town Hall Seattle and the Institute for Systems Biology.
