Today's crossword, by veteran cruciverbalist Paolo Pasco, had its ups and downs, but that was by design, and the payoff - which we discuss in today's episode but don't want to spoil by revealing here - was an absolute delight. The clues were on the trickier side, as befits a Thursday - 33D,  So called "African Unicorn", OKAPI, comes to mind (well, it does now ...), as does 25D, Noted feature of "The Brothers Karamazov", EPILOG, and 48D, Lake that feeds the Truckee River, TAHOE (who? what? where?). A fine Friday crossword by Juliana Tringali Golden, with arguably the most interesting clue of the month, 53D, Country where almost half the world's zippers are made, JAPAN (wow!). Of course, there were many other worth pondering -- 25D, Yawl pole, MAST (say what?), 59A, Immediate threat to capture in a game of Go, ATARI (OK, if you say so), and 55A, "Platoon" but not "Dunkirk", IAMB (possibly the most NYTimes-ish crossword clue ever).  All in all a great crossword, 5 squares on the JAMCR scale for sure!

I'm looking forward to sharing with you some of our community's questions that have come in over the past few weeks… Let's get started!    Nadia: Dear Doctor Cabral, thank you so much for all you do to help the community and for your very interesting podcast! I would like to ask if you could indicate me in which direction we could look regarding my dad's health. He is 62 years old, 5feet7 and weighs now less then 125 pounds. He has always been thin (even though he eats 3 times more then the rest of the family) but muscular. But now he really has just skin on his bones even though he still eats a lot (and healthy). He goes to the bathroom at least once a day, sometimes after every meal, which is very good. And his blood work came back normal. He's been trying to gain weight for years but he just gets more and more skinnier and is pretty preoccupied. He is very active all day, always working around the house, in the garden, loves hiking and biking. A Nutritionist told him years ago that it will be impossible for him to gain weight and that it's much easier for overweight people to lose weight. Do you have any advice on how my dad could get to a normal weight again or what we should look at? Thanks so much for your time and help, Have a great day, Nadia Erin: Vitamin c help!! Ok so i was told only to take food based vitamin c but all the other is synthetic and depletes copper....so can you please clear this up for me. I really appreciate it!! Ryan: Hello! I have seen recently a somewhat controversial opinion on D3 supplementation and would like your opinion. Usually, I'd gloss over something like this, but considering the high reputation of the source in the functional/integrative space, I thought I'd get your highly valued opinion. Quote… "D3 is not what they measure in a vitamin D test. They measure 25D. The problem is, they should be measuring 1,25D, and better yet, the ratio between the two. Supplementing D3 will raise your 25D levels which shuts down your vitamin D receptors and innate immune system function. Supplementing D3 above what we naturally get in food will massively increase calcium absorption. In a hard-drinking water world, that causes hypercalcemia, magnesium and K2 deficiency.” Jamie: Hi Dr Cabral thank you for all you do. I have been listening for a very long time. I'll try to keep this short as I did try to search if you have talked about this before but couldn't find anything. I currently see a Dr who claims to be an integrative doctor local to me. I'm 33 year old female. I weigh around 130#. My symptoms have been thinning hair, nail fungus, zero libido, zero energy and pms 2 weeks prior to menstruating, (this has been for many years) increase of stress the past two years due to my job. . He did lab work and my thyroid levels are low, and vitamin d was low. He also told me that I should take Progesterone 100mg qhs for the pms symptoms / low energy. Long story short I'm taking t3/t4 15mcg daily (along with fish oil, vitamin d, b complex, magnesium from EquiLife) I had more lab work done recently and my IGF-1 is 208 (says it should be 280) and he is recommending Sermorelin therapy. I googled it and it sounds fantastic. All the things I need (energy, libido etc) but my question is being only 33 (no kids) is going to harm me in the long run taking these hormones? I know you can't tell me to stop, I just need advice on really what this Sermorelin therapy is and if it is going to hurt me in the long run taking progesterone “forever”. Ross Trudeau steps up to the plate, swings, and it's a hit -- because Ross Trudeaus' crosswords are always a hit. This is a more high-brow crossword than is typical for the New York Times, as there were 3 clues that tied in to 28D, Literary trio found in the answers to this puzzle's starred clues, THEBRONTES. Those clues -- 6D, Chain known for its soft pretzels, AUNTIEANNES; 25D, Prominent left-leaning political action committee, EMILYSLIST;  and the rather lengthy 19A, Children's book whose title character says, "If I can fool a bug, I can surely fool a man. It's a gigantic crossword! No, not really, it is just 15x15, but it did manage to fit a giant's trademark FEEFIFOFUM into the answers. It also slipped in one of our favorite shows, 25D, Discovery Channel program that debunked popular beliefs, MYTHBUSTERS, and therefore EARN our respect. We were glad to see a shoutout to 26A, Island nation in the western Pacific, PALAU, as well as one of our all-time favorite deities, 49D, Greek goddess of wisdom, ATHENA. A nice crossword - Ross Trudeau is inherently incapable of writing any other kind - we give it a 5 squares on the JAMCR scale.

The EPITOME of what a Monday crossword should be -- not too difficult, with a GLOB of humor (6D, Try, try again?, REHEAR, and 25D, Shelters for shelties, KENNELS), and a few novel words (41D, Theatergoer's break, ENTRACTE, and the slightly more common but even more difficult to spell word, 67A, Garlicky sauce, AIOLI). Jean was able to get through all of this with her usual APLOMB, while Mike wasn't quite as TIMELY. We give it a 5 squares on the JAMCR scale.

There was once a game called Advent Rising. It was hyped up as the next great science-fiction adventure that would transcend games and become something more. Unfortunately, it released in 2005 with a multitude of bugs in an era where patching post-release was a rarity at best. Advent Rising caused the implosion of its development studio, GlyphX Games. A group of individual developers escaped the studio's downfall, banding together to form Chair Entertainment. The newly minted indie studio went on to develop and release Shadow Complex in 2009.  The 2.5D metroidvania side-scroller adopted a more realistic aesthetic and spawned a series of novels authored by Orson Scott Card. The game released and seemed to fill a niche in the indie gaming world that hadn't been filled in quite that same way before. With a recent remaster, it seems like a perfect time to ask the question: Is Shadow Complex one of the best games period? Det hade kostat henne allt hon ägde, men ingenting hade hjälpt, snarare hade hon blivit sämre. 27Hon hade hört vad som berättades om Jesus, och nu kom hon bakifrån i hopen och rörde vid hans mantel, 28för hon tänkte att om hon bara fick röra vid hans kläder skulle hon bli hjälpt. 29Och genast stannade blodflödet, och hon kände i kroppen att hon var botad från sitt onda. 30När Jesus märkte att det hade gått ut kraft från honom vände han sig om i hopen och frågade: ”Vem rörde vid mina kläder?” 31Lärjungarna sade: ”Du ser väl hur folk tränger på, och ändå frågar du vem som har rört vid dig!” 32Han såg sig omkring efter henne som hade gjort det. 33Kvinnan, som visste vad som hade hänt med henne, kom rädd och darrande fram och föll ner för honom och talade om hur det var. 34Han sade till henne: ”Min dotter, din tro har hjälpt dig. Gå i frid. Du är botad från ditt onda.”

Vitamin D kann entweder durch die Nahrung aufgenommen oder durch UV- Bestrahlung der Haut aus 7-Dehydrocholesterol gebildet werden. Das entstehende inaktive Vitamin D3 wird anschließend durch die 25-Hydroxylase (CYP2R1) in der Leber zu 25-Hydroxyvitamin D3 (25D) metabolisiert. 25D ist die vornehmlich zirkulierende Form von Vitamin D, es selbst besitzt allerdings nur etwa ein Tausendstel der Aktivität der endgültigen, aktiven Form des Hormons, 1α,25- Hydroxyvitamin D3 (1,25D), das überwiegend in der Niere durch die mitochondriale Hydroxylase CYP27B1 produziert und im Plasma durch Interaktion mit DBP (plasma vitamin D binding protein) stabilisiert und transportiert wird. Die Hauptfunktion von 1,25D besteht offenbar in der Bindung an das intrazelluläre Protein Vitamin D Rezeptor (VDR), welches daraufhin typischerweise mit einem zweiten Protein, dem Retinoid X Rezeptor, heterodimersisiert und dann gemeinsam mit diesem an sogenannte Vitamin D response elements (VDREs) in der DNA bindet. Die weiteren Konsequenzen dieser Bindung sind nicht in allen Einzelheiten verstanden, führen aber entweder zur transkriptionellen Aktivierung oder Reprimierung einer großen Anzahl von Gensequenzen. Die E3 Ubiquitin-Ligase und Transkriptionsrepressor MDM2 ist ein potenter Inhibitor der p53 Familie von Transkriptionsfaktoren, Stoffwechselregulatoren und Tumorsuppressoren. Es konnte mit dieser Arbeit gezeigt werden, dass der VDR ein weiterer Transkriptionsfaktor, Stoffwechselregulator und Tumorsuppressor ist, welcher ebenfalls von MDM2 gebunden und inhibiert wird. Es stellte sich heraus, dass der VDR in der Zelle zum Teil durch MDM2 ubiquityliert wird, seine Steady-State Level durch das Proteasom kontrolliert werden und ein Knockdown von endogenem MDM2 die VDR-Level erhöht. Ein Knockdown von MDM2 führte zu einer signifikanten Erhöhung des Transkripts der Gene CYP24A1 und p21, klassische zelluläre Ziele der Transaktivierung durch ligandengebundenen VDR. Die Ergebnisse dieser Arbeit legen nahe, dass MDM2 analog zu p53 den VDR negativ reguliert.
