How do new insights from the XARENO study impact treatment options for patients with AF and advanced CKD? Abstract: Tissue clearing is currently revolutionizing neuroanatomy by enabling organ-level imaging with cellular resolution. However, currently available tools for data analysis require a significant time investment for training and adaptation to each laboratory's use case, which limits productivity. Here, we present FriendlyClearMap, an integrated toolset that makes ClearMap1 and ClearMap2's CellMap pipeline easier to use, extends its functions, and provides Docker Images from which it can be run with minimal time investment. We also provide detailed tutorials for each step of the pipeline. For more precise alignment, we add a landmark-based atlas registration to ClearMap's functions as well as include young mouse reference atlases for developmental studies. We provide alternative cell segmentation method besides ClearMap's threshold-based approach: Ilastik's Pixel Classification, importing segmentations from commercial image analysis packages and even manual annotations. Finally, we integrate BrainRender, a recently released visualization tool for advanced 3D visualization of the annotated cells. As a proof-of-principle, we use FriendlyClearMap to quantify the distribution of the three main GABAergic interneuron subclasses (Parvalbumin+, Somatostatin+, and VIP+) in the mouse fore- and midbrain. For PV+ neurons, we provide an additional dataset with adolescent vs. adult PV+ neuron density, showcasing the use for developmental studies. When combined with the analysis pipeline outlined above, our toolkit improves on the state-of-the-art packages by extending their function and making them easier to deploy at scale. Abstract: Synthetic sugar analogs are widely applied in metabolic oligosaccharide engineering (MOE) and as novel drugs to interfere with glycoconjugate biosynthesis. However, mechanistic insights on their exact metabolism in the cell and over time are mostly lacking. We developed sensitive ion-pair UHPLC-QqQ mass spectrometry methodology for analysis of sugar metabolites in organisms and in model cells and identified novel low abundant nucleotide sugars in human cells, such as ADP-glucose and UDP-arabinose, and CMP-sialic acid (CMP-NeuNAc) in Drosophila. Dynamic tracing of propargyloxycarbonyl (Poc) labeled analogs, commonly used for MOE, revealed that ManNPoc is metabolized to both CMP-NeuNPoc and UDP-GlcNPoc. Finally, combined treatment of B16-F10 melanoma cells with antitumor compound 3Fax-NeuNAc and 13C-labeled GlcNAc revealed that endogenous CMP-NeuNAc levels started to decrease before a subsequent decrease of ManNAc 6-phosphate was observed. This implicates 3Fax-NeuNAc first acts as a substrate for cytosolic CMP-sialic acid synthetase and subsequently its product CMP-3Fax-NeuNAc functions as a feed-back inhibitor for UDP-GlcNAc 2-epimerase/N-acetylmannosamine kinase. Thus, dynamic analysis of sugar metabolites provides key insights into the time-dependent metabolism of synthetic sugars, which is important for the rational design of analogs with optimized effects. Barbara Rossing first came to Holden as Waitri in 1973 and also served as Village pastor 1986-88. She is professor of New Testament at the Lutheran School of Theology at Chicago, where she also directs the seminary's environmental ministry emphasis. Her publications include The Rapture Exposed: The Message of Hope in the Book of Revelation (Basic Books, 2004); Journeys Through Revelation: Apocalyptic Hope for Today (Presbyterian Church USA, 2010); “Revelation” in The Fortress Commentary on the Bible (2014), and articles and book chapters on the Bible and ecology, including the Lutheran World Federation's publication "Creation-- Not For Sale." She chairs the Society of Biblical Literature's Ecological Hermeneutics section, and is active in public theology in both the scholarly academy and the church, working on climate change and the Bible. She loves working on the 500th anniversary of the Reformation in 2017, including ecumenical responses to Pope Francis's encyclical letter on climate change. She received the MDiv degree from Yale University Divinity School, and the ThD from Harvard University. This podcast features Barbara Rossing of Leavenworth, who has been a Lutheran pastor for 35 years. She was pastor for three years in the late 1980s at Holden Village and currently teaches New Testament at the Lutheran School of Theology in Chicago. Rossing is passionate about the future of the natural environment, how we care for our most vulnerable citizens and about finding common cause with folks of other faiths. This is the 500th anniversary of the Protestant Reformation, which split Christianity. Efforts are being made globally and locally to repair relationships. On Sunday night, October 1, a joint service will be held at the St. Joseph's Catholic Church led by local Lutheran pastors. Rossing and Father Osmar Aguirre will speak. During our interview, we spoke of her admiration for Pope Francis and his outreach to other faiths and the shared values of both churches. We also spoke of the values that she learned at Holden Village and how those have inspired her activism with the local Climate Conversations group, involvement with Upper Valley MEND and other endeavors. "The more the church can lead on reminding us who we are as compassionate people – as community builders – the more we will be in a better world," Rossing told me. "Community is much more than just a house. Where we experience the love of God and the love of one another rooted in this beautiful, wonderful place we call home," she said. She's a fascinating speaker.

What was the message that Jesus came to share with the world? What was it that Jesus seemed to always be talking about? What was the gospel he preached, embodied and then sent his disciples out to proclaim? Many seem to believe Jesus' message was primarily about information on how to go to heaven after you die. A rather prominent Christian pastor was quoted in a popular magazine as saying this about the Jesus we find in Revelation: “In Revelation, Jesus is a pride-fighter with a tattoo down His leg, a sword in His hand and a commitment to make someone bleed. This is a guy I can worship. I cannot worship the hippy, diaper, halo Christ, because I cannot worship a guy I can beat up.” Sadly, his thoughts about the Jesus of Revelation are not entirely unique. Some have used the book of Revelation to depict a violent God, who supports violent acts and who ultimately brings the world to a violent end. Every couple of decades or so it seems there is a disease that the American Church gets afflicted with. Some have called it "Apocalyptic Fever." Books, films and conferences pop up around the country giving rise to wild speculations and predictions about the end of the world, which most seem sure will happen soon. The one and only thing they all seem to have in common is each cites the book of Revelation as their source. How are we to understand these things as Christians?
