Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2023.03.06.531272v1?rss=1 Authors: Zuhlsdorff, K., Verdejo-Roman, J., Clark, L., Albein-Urios, N., Soriano-Mas, C., Cardinal, R., Robbins, T., Dalley, J. W., Verdejo-Garcia, A., Kanen, J. W. Abstract: Cognitive flexibility refers to the ability to adjust to changes in the environment and is essential for adaptive behavior. It can be investigated using laboratory tests such as probabilistic reversal learning (PRL). In individuals with both Cocaine Use Disorder (CUD) and Gambling Disorder (GD), overall impairments in PRL flexibility are observed. However, it is poorly understood whether this impairment depends on the same brain mechanisms in cocaine and gambling addictions. Reinforcement learning (RL) is the process by which rewarding or punishing feedback from the environment is used to adjust behavior, to maximise reward and minimise punishment. Using RL models, a deeper mechanistic explanation of the latent processes underlying cognitive flexibility can be gained. Here, we report results from a re-analysis of PRL data from control participants (n=18) and individuals with either GD (n=18) or CUD (n=20) using a hierarchical Bayesian RL approach. We observed significantly reduced stimulus stickiness (i.e., stimulus-bound perseveration) in GD, which may reflect increased exploratory behavior that is insensitive to outcomes. RL parameters were unaffected in CUD. We relate the behavioral findings to their underlying neural substrates through an analysis of task-based fMRI data. We report differences in tracking reward and punishment expected values (EV) in individuals with GD compared to controls, with greater activity during reward EV tracking in the cingulate gyrus and amygdala. In CUD, we observed reduced responses to positive punishment prediction errors (PPE) and increased activity following negative PPEs in the superior frontal gyrus compared to controls. Thus, an RL framework serves to differentiate behavior in a probabilistic learning paradigm in two compulsive disorders, GD and CUD. Copy rights belong to original authors. My interview today is with Mike Kanen, Regional Crosschecker with the San Diego Padres. I have known Mike for several years going back to when our facility was in Sloatsburg, NY, where he used to run scouting events for the Indians inviting only a handful of players and their families for closeup meetings and evaluation.I have interviewed many college coaches during the past year and I am very interested to hear how college and pro ball perspectives can differ on the recruiting and scouting front. Join us as we dig into a variety of topics, including:Scouting HS players vs. college playersGoing to college vs. pro ballSpecific traits when scouting a young athleteHow scouting has changed over the past 5-10 yearsMike is a graduate of John Hopkins University where he played baseball for 4 years. He joined the scouting department of the Washington Nationals straight out of school. Thereafter, he was with Cleveland Indians in various scouting capacities for nearly 8 years before joining the Padres about a year and a half ago.Welcome to the Show Mike… Ready to take your game to the next level? Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2023.02.15.527569v1?rss=1 Authors: Luo, Q., Kanen, J. W., Bari, A., Skandali, N., Langley, C., Knudsen, G. M., Alsio, J., Phillips, B. U., Sahakian, B. J., Cardinal, R. N., Robbins, T. W. Abstract: Serotonin is critical for adapting behavior flexibly to meet changing environmental demands. Cognitive flexibility is important both for successful attainment of goals, as well as for social interactions, and is frequently impaired in neuropsychiatric disorders, including obsessive-compulsive disorder (OCD). However, a unifying mechanistic framework accounting for the role of serotonin in behavioral flexibility has remained elusive. Here, we demonstrate common effects of manipulating serotonin function across two species (rats and humans) on latent processes supporting choice behavior during probabilistic reversal learning using computational modelling. The findings support a role of serotonin in behavioral flexibility and plasticity, indicated, respectively, by increases or decreases in choice repetition ('stickiness') or reinforcement learning rates depending upon manipulations intended to increase or decrease serotonin function. More specifically, the rate at which expected value increased following reward and decreased following punishment (reward and punishment 'learning rates') was greatest after sub-chronic administration of the selective serotonin reuptake (SSRI) citalopram (5 mg/kg for 7 days followed by 10 mg/kg twice a day for 5 days) in rats. Conversely, humans given a single dose of an SSRI (20mg escitalopram), which can decrease post-synaptic serotonin signalling, and rats that received the neurotoxin 5,7-dihydroxytryptamine (5,7-DHT), which destroys forebrain serotonergic neurons, exhibited decreased reward learning rates. A basic perseverative tendency ('stickiness'), or choice repetition irrespective of the outcome produced, was likewise increased in rats after the 12-day SSRI regimen and decreased after single dose SSRI in humans and 5,7-DHT in rats. These common effects of serotonergic manipulations on rats and humans - identified via computational modelling - suggest an evolutionarily conserved role for serotonin in plasticity and behavioral flexibility and have clinical relevance transdiagnostically for neuropsychiatric disorders. In the fourth episode of the Passive Investing Show, J talks to Alexander Kanen about how crypto and blockchain are ripe to revolutionize the private equity and real estate industries. They talk about the benefits brought about by these new technologies, how we as investors can maximize its potential, and the developments that will be coming in the very near future. Technology has continuously paved the way in reinventing old processes and concepts. Find out the exciting things that crypto and blockchain currently offer and have in store! Here are some power takeaways from today's conversation: Tokenization brings liquidity and price discovery into the real estate industry. Blockchain redefines ownership, making it sovereign and programmable. We should refrain from exaggerating the value of tokenization. Standards and secondary markets are needed to truly bring out the value of tokenization. Episode Highlights: [02:33] Down the Bitcoin Rabbit Hole Alex started dipping his toes into bitcoin after accommodating a certain client. Gradually, they designed a protocol to conduct a real estate closing using digital asset transactions. While passionate about crypto, Alex is more focused on the technology side of things and advises people to seek professional advice before using crypto. [07:39] Crypto and Technology in Real Estate Blockchain brings the benefits found in public markets to the private equity and real estate space. Equity tokens are digital counterparts to existing analog systems and instruments. They fractionalize the entity that owns the real estate, not the real estate itself. Tokenization allows people to enjoy liquidity and price discovery. When you tokenize 40% of an asset, it grants you access to larger capital pools. It lessens the friction in the paperwork process once you find an investor. It also allows you to automate compliance, building it into the token itself. [26:36] A Word of Caution Syndicating to millions of investors may sound tempting, but there are regulatory requirements you need to comply with. Do not exaggerate the value of tokenization and liquidity. Industry-wide standards and secondary markets are necessary for liquidity to become a reality. [31:53] Current State of the Real Estate Industry Tokenization is progressing fast in real estate. However, secondary markets and universal standards are still needed to attain true liquidity. Regulation will keep the processes in real estate on track. With crypto and blockchain, ownership becomes sovereign and programmable. Notable Quotes from the Episode: [20:04] “Tokenization offers you to take a piece of your asset. Don't sell the whole thing.” [27:34] “It's important not to exaggerate the value of tokenization. And it's important not to exaggerate the value of liquidity in some of these cases.” [36:16] “Little by little, [the advent of AI and blockchain is] transforming our understanding of ownership. Like we said before, it's sovereign, programmable ownership. It is to disintermediate the gatekeepers, disintermediate the centralized points of vulnerability if you will.” Resources Mentioned: LinkedIn Kanen Law

Ok, I'll be honest, I am FREAKING OUT right now over this interview! This women needs no introduction, she's the star of six international TV shows, the author of six books and the changer of families across the globe! Since then, I've been referring clients to her videos and sharing her techniques with moms for years. She IS the guide to parenting and her methods WORK! I encourage you to go watch her show Supernanny, streaming on Hulu, Youtube, ABC and her new season on Lifetime. Here's just a few of the staples of my parenting that I learned from watching it over the years: Consistent, fair, firm and loving discipline (search up "the knotty spot") Sleep training for babies and young kids Schedule and routine Positive reinforcement The importance of PLAYING with your kids Chores and responsibilities for kids Bedtime routine How to be present with your kids and so much more! You can watch full episodes on specific topics on Youtube PLEASE share this episode with every parent you know, and especially every single mom, mom's need to know there is help for whatever they are going through with their kids. BBQ Holdings, Inc. (BBQ) a national restaurant company engaged in the ownership and operation of casual and fast dining restaurants. BBQ Holdings has four brands with 145 “brick and mortar” locations in 31 states and three countries, including 47 company-owned and 98 franchise-operated restaurants. In addition, the Company opened five Company-owned Famous Dave's ghost kitchens operating out of its Granite City locations, and six Famous Dave's franchisee ghost kitchens operating out of the kitchen of another restaurant location or a shared kitchen space. While BBQ Holdings continues to diversify, it was founded with the principle of combining the “art and science” of barbecue to serve up the very best of the best to barbecue lovers.

The infamous live show is HERE! 2 of our favorite guests, @BillSchulz and @JoNosuchinsky from @morninBillandJo on @compoundmedia rejoined us to do one of the 80s biggest darlings, The Karate Kid. Perfect timing right smack in the middle of the Cobra Kai popularity. Bill and Joanne have no love for this Jersey boy getting jumped every 5 minutes who becomes an indentured servant to a Japanese handyman. And Bill plays a message from a star from one of the Karate Kid films as he's none too pleased that this film is taking fire. But can this duo convince @KevinGootee and @KevinIsrael_NJ they're the best...around. NOTHIN'S EVER GONNA KEEP THEM DOWN? @GTSCpodcast on twitter, Gutting the Sacred Cow on Twitter, FB, Tumblr, and TikTok Guttingthesacredcow.com every day for new articles to give you the pop culture news that makes you look smart, funny, and more tolerable as a person. Come listen to a WUU service!250 years ago, John Murray, a Universalist preacher who had lost his faith after the death of his wife and child, sailed from England to New York. Or, at least, he meant to. He ended up stranded along the coast of New Jersey. There, he met a Universalist who changed his life...and our lives too. Today we celebrate this momentous anniversary and look for signs of renewal in our own lives. Also today, the Wisdom Story will be in the form of an amazing video by the Craig Family!Rev.
