Abstract: ApoE4 isoform contributes to increased risk for Alzheimers Disease (AD) over the life course of individuals. Much remains unknown about the biological pathways that connect APOE4 genotype with the development of pathology that eventually leads to AD, nor do we know how early in life these cellular alterations begin. To answer these questions, we derived neural precursor cells (NPCs) from induced pluripotent stem cells (IPSCs) that were CRISPR-edited at the APOE locus. We intended to characterize the protein expression landscape in the NPCs subsequent to targeted deletion of E4 from a parent IPSC line of APOE3/4 genotype. Differentially expressed proteins (DEPs) following mass spectrometric analysis were determined from the protein abundance fold change values obtained for each protein. Proteins which showed greater than 1.5-fold difference with FDR adjusted P-value less than 0.05 were considered differentially expressed. DEPs were mapped to the STRING database (v11.5) for retrieval of interacting proteins and functional enrichment. CRISPR-editing of E4 from the parent line revealed 98 differential expressed proteins. Of these, 54 were upregulated, and 44 were downregulated. Further analysis of the DEPs via STRING database showed that these changes primarily affect pathways linked to RNA processing, plasma membrane repair, and cytoskeleton organization. Indeed, we find the effects of E4 extend beyond proteins considered central to AD pathology. Knowing more about the protein interactions regulated by ApoE, in an isoform-specific manner, can reveal new mechanistic insights into development of AD. Copy rights belong to original authors. A reportagem "Colônia dos Excluídos" exigiu da jornalista Iara Diniz um mergulho em um período no qual pessoas diagnosticadas com hanseníase no Estado eram obrigadas a se isolar na Colônia de Itanhenga, uma verdadeira minicidade construída em 1935 em Cariacica. De lá, ninguém tinha permissão para sair. A série de reportagem é destaque em A Gazeta desde a última segunda-feira (23). Em entrevista à Rádio CBN Vitória, a jornalista falou sobre a reportagem. Ouça a conversa completa! O Brasil é o segundo país com maior número de casos de hanseníase, ficando atrás apenas da Índia. Nos últimos dez anos, 312 mil pessoas foram diagnosticadas com a doença. Apesar disso, a hanseníase permanece negligenciada, com pouco investimento em pesquisas e, ainda, sem uma vacina. A mudança do nome lepra para hanseníase foi feita em 1976, por meio de decreto federal, e se tornou lei em 1995. A medida veio com o objetivo de diminuir o estigma milenar associado à doença e considerada por alguns estudiosos como "a ação mais eficiente no combate da doença". O preconceito, no entanto, ainda está presente na sociedade e é apontado pela médica como um dos principais fatores que dificultam o diagnóstico precoce da hanseníase. Em entrevista à CBN Vitória, a doutora Patrícia Deps, especialista em hanseníase e professora titular e chefe do Departamento de Medicina Social e do Programa de Pós-graduação em Doenças Infecciosas da Universidade Federal do Espírito Santo (Ufes), fala sobre o assunto. Data and Reporting Standards for digital events have been needed and missing for many years, and the shift to digital since the beginning of the pandemic made it even more necessary! The recent cyber attacks on US companies have brought to light the importance of cyber security. A new set of guidelines for securing these components and software as a whole will be released soon, impacting not just software developers but the users as well. The Cloud OnAir event will breakdown these new guidelines and educate attendees on steps to take to ensure more secure software and software components. Internally, Google has been optimizing their software supply chain security for years with solutions like BeyondCorp and internally developed solutions that Google has since adapted for their clients. These solutions will be discussed in detail in the Cloud OnAir event. Victor goes on to explain the three areas of supply chain security and how they fit into the overall security of online platforms. Software projects are often built using many small pieces of software sourced from third parties, which can create vulnerabilities. The new guidelines will help ensure quality and security at all levels of development for software and its pieces, thus strengthening security at every level of the supply chain. Nikhil and Victor talk about issues that contribute to supply chain security, including the risks that a microservices architecture can introduce and the use of open source software and their dependencies. We hear about Google’s contributions to the supply chain security effort, like OpenSSF that strives to bring the open source community together toward the goal of cyber security. Yves vit dans le XXe Arrondissement de Paris. C'est également là qu'il a grandi et qu'il enseigne, depuis 20 ans, l'Education Physique et sportive. Ce qui m'a intrigué chez lui, c'est d'avoir eu la chance de voir le travail qu'il mène avec ses élèves autour de la danse. Et ce sont les rues de Paris qui sont le théâtre du ballet improbable que nous offrent ses élèves. Il définit cette pratique comme une étape sur le chemin de sa réflexion sur son rôle dans cette société, dans cette école et sur l'EPS. Christian Le Pape se serait bien vu comme nombre de ses copains de la baie de la Forêt dans les années 1980 - les Le Cam, Jourdain, Guillemot - devenir coureur à plein temps. Mais la vie de saltimbanque sans statut qui allait avec ne l'emballait pas trop et il a préféré devenir prof de gym. De ce choix cornélien de sa jeunesse, le Finistérien gardera une trace qui l'amènera, en 1990, a créer une structure qui va profondément transformer et organiser la voile de compétition en France : le Pôle Finistère Course Au Large de Port-La-Forêt. Trois décennies plus tard, Christian Le Pape, 61 ans, raconte dans cet épisode le cheminement qui amène des marins - qui, longtemps, ont préféré investir dans le matériel - vers l'entraînement et la professionnalisation de leur pratique. Trente années passées à survoler les podiums et à se construire un palmarès incroyable, d'abord en Figaro (la Solitaire n'a pas échappé depuis 2002 à un coureur du pôle) puis en Imoca (idem pour le Vendée Globe depuis 2000-2001), qui font de Portlaf', comme on dit, une référence en or massif de la course au large. L'ex prof d'EPS n'occulte pas les difficultés, les discussions, les dissensions sur la route du succès mais évoque aussi le chercheur de pépites qu'il est, ayant détecté tour à tour Franck Cammas, Armel Le Cléac'h, François Gabart, Sébastien Simon. Plus de 2 heures d'émission pour découvrir comment fonctionne, non pas la Star Ac' de la voile, comme on a pu parfois appeler le Pôle, mais plutôt le Harvard de la course au large !

Abstract: Colorectal cancer (CRC) is a malignant tumour with high morbidity and mortality worldwide. Efficient screening strategies for CRC and pre-cancerous lesions will promote early medical intervention and treatment, thus reducing morbidity and mortality. Proteins are generally considered key biomarkers of cancer. Herein, we performed a quantitative, tissue-original proteomics study in a cohort of ninety patients from pre-cancerous to cancerous conditions by liquid chromatography-tandem mass spectrometry. A total of 134,812 peptides, 8,697 proteins, 2,355 (27.08%) union differentially expressed proteins (DEPs), and 409 shared DEPs (compared with adjacent tissues) were identified. The number of DEPs showed a positive correlation with increasing severity of illness. The union and shared DEPs were both enriched in the KEGG pathway of focal adhesion, metabolism of xenobiotics by cytochrome P450, and drug metabolism - cytochrome P450. Among the 2,355 union DEPs, 32 were selected for identification and validation by multiple reaction monitoring from twenty plasma specimens. Of these, three proteins, transferrin receptor protein 1 (TFR1), adenosylhomocysteinase (SAHH), and immunoglobulin heavy variable 3-7 (HV307), were significantly differentially expressed and displayed the same expression pattern in plasma as observed in the tissue data. In conclusion, TFR1, SAHH, and HV307 may be considered as potential biomarkers for screening of CRC. Copy rights belong to original authors. Visit the link for more info

Abstract: The polyphagous pest Apolygus lucorumhas become the dominant insect in Bacillus thuringiensis (Bt) cotton fields. The hormone 20-hydroxyecdysone (20E) regulates multiple events in insect development and physiology. 20E responses are controlled by pathways triggered by phospholipase C (PLC)-associated proteins. However, 20E-modulated genes whose expression is affected by PLC remain unknown. Here, isobaric tag for relative and absolute quantitation (iTRAQ) and immunoblot were carried out for comparing differentially expressed proteins (DEPs) in A. lucorumin response to 20E and the PLC inhibitor U73122, respectively. Totally 1624 DEPs were, respectively, found in the 20E/control, U73122/control, and 20E+U73122/control groups. Venn diagram analysis further revealed 8 DEPs that were shared among the three groups. Immunoblot validated these findings, which corroborated and highlighted the reliability of proteomics. KEGG enrichment analysis showed that the DEPs were included in diverse signaling pathways. The largest portion of DEPs among the three groups were categorized in metabolic pathways. In addition, DEPs among the three groups were also found to regulate the Ras-MAPK and PI3K-AKT pathways. This is the first time that iTRAQ was carried out to assess proteome alteration in A. lucorum nymphs in response to 20E and a PLC inhibitor. These findings provide novel insights into protein expression in A. lucorumin response to 20E, and a more comprehensive understanding of the function of PLC in 20E signal transduction. Google Cloud Platform (GCP) turned off a customer that it thought was doing something out of bounds. This led to an Internet outrage, and GCP tried to explain itself and prevent the problem in the future. Today, we’re talking to Daniel Compton, an independent software consultant who focuses on Clojure and large-scale systems. He’s currently building Deps, a private Maven repository service. As a third-party observer, we pick Daniel’s brain about the GCP issue, especially because he wrote a post called, Google Cloud Platform - The Good, Bad, and Ugly (It’s Mostly Good). Some of the highlights of the show include: Recommendations: Use enterprise billing - costs thousands of dollars; add phone number and extra credit card to Google account; get support contract Google describing what happened and how it plans to prevent it in the future seemed reasonable; but why did it take this for Google to make changes? GCP has inherited cultural issues that don’t work in the enterprise market; GCP is painfully learning that they need to change some things Google tends to focus on writing services aimed purely at developers; it struggles to put itself in the shoes of corporate-enterprise IT shops GCP has a few key design decisions that set it apart from AWS; focuses on global resources rather than regional resources When picking a provider, is there a clear winner? AWS or GCP?
