BUFFALO, NY- December 10, 2024 – A new #research paper was #published in Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 21 on November 18, 2024, entitled, “Prostaglandin E2 regulates senescence and post-senescence neoplastic escape in primary human keratinocytes.” Researchers Elise Srour, Nathalie Martin, Claire Drullion, Clémentine De Schutter, Joëlle Giroud, Adrien Pioger, Julie Deslé, Laure Saas, Joe Nassour, Julien Théry, Gauthier Decanter, Nicolas Penel, Chantal Vercamer, Clara Salazar-Cardozo, Corinne Abbadie, and Olivier Pluquet from CNRS, University of Lille, the Oscar Lambret Center, and the University of Colorado School of Medicine have revealed how a molecule called Prostaglandin E2 (PGE2) influences skin aging and cancer risk. Their study shows that PGE2 not only drives skin cells to age but also enables some of these aging cells to bypass natural limits and develop into pre-cancerous cells. This process provides insights into why older skin is more susceptible to cancer. The study focused on keratinocytes, the primary cells in the outer layer of the skin. As these cells age, they enter a state called senescence, where they stop dividing to prevent damaged cells from turning cancerous. While this process typically serves as a protective mechanism, the researchers found that, in certain cases, some senescent cells can escape this state, re-enter the cell cycle, and acquire characteristics of early cancer. By examining keratinocytes from donors of different ethnicities and ages, the researchers identified the PTGS2/PGE2/EP4 pathway as a key driver of this escape process. The researchers show that blocking PGE2 or its associated pathway reduced the chances of aged cells becoming precancerous. This suggests that drugs targeting this pathway, including some anti-inflammatory medications already in use, might be repurposed to slow skin aging and prevent early-stage skin cancers. Additionally, the study also found that PGE2 levels increase in the skin as it ages, further supporting its role in skin health and disease. "These results indicate that the PTGS2/PGE2/EP4 pathway is required to induce and maintain the senescent phenotype of NHEKs, and that PGE2 level is a potential determinant of the initial steps of the age-related oncogenic process." The team also highlighted the broader implications of their work. The PTGS2/PGE2/EP4 pathway is not only linked to skin health but also to age-related inflammation, a condition that contributes to several diseases. By addressing this pathway, researchers hope to address not only skin aging but other health challenges linked to aging and chronic inflammation. In conclusion, this study reveals important molecular drivers of skin aging and early cancer, leading the way for new approaches that can promote healthier skin. The King has returned! For the first time since 2023, Carlos Vela is back on the pitch for LAFC! For the 10th time in two years, LAFC takes on Vancouver. One of those things we love, the other not so much, but we talk about it all on Episode 335!LAFC, as they have done for much of the last two seasons, dominated a lesser-talented Vancouver side over the course of 90+ minutes. A penalty goal by Denis after great combination play from Bogusz and Giroud set the tone for the first half, putting LAFC up 1-0. Just before the hour mark, excellent build-up and ball movement allowed Cristian Olivera to double the leads. LAFC looked to be cruising when, in the 86', Carlos Vela jogged over to the bench after warming up in front of the North End, signaling the return of El Rey!Neither Mateusz Bogusz hitting the woodwork twice nor a late, stoppage-time goal for Vancouver could damper the mood at BMO. Our #10 was back, LAFC takes the first playoff match of 2024, and now... we play Vancouver again. Oy.Also on the pod, CWM Woodwork is a thing. On Episode 334, Filly and Scarf recap an absolutely unbelievable Decision Day, both at BMO and in Houston! The subs save the day in LA while a former Carsonite sinks their ship at the death! It was a roller coaster of emotions for about 15 minutes, but LAFC comes out on top in the West as we get set to kick off the Western Conference Playoffs!Things looked bleak for the first 45' for LAFC, as an early goal by Jackson Yueill, off an ill-advised pass from Eddie Segura, gave the road side a lead and meant that LAFC would need at least three on the day. Another offside goal from Giroud and an anemic first half attack (0 shots on target) sent the club into the halftime break down a goal. But in Houston, an Ezequiel Ponce goal just before the halftime whistle saw the home side take the lead, and left everything to play for at BMO!LAFC opened the second half a little better, but it was just after the hour mark that LAFC truly woke up. A Denis Bouanga banger off a great Tillman cross leveled things at home, and the reinforcements came on soon after. Kei Kamara, Eduard Atuesta, and, later, Marlon, would all factor in the win. Atuesta's first few touches were magic, as he took a great 1-2 ball from Kei and rocketed it home past Daniel to give LAFC the 2-1 lead! With Aaron Long cramping in the 80', the Carlos Vela homecoming was put on hold, as LAFC used their 5th sub to bring on Marlon for Long. Serendipity doesn't always announce itself right away, but it would take Marlon just 10 minutes to become an LAFC hero! Off a great Atuesta free kick, Marlon, running in the same channel as Chanot, got his head to the cross and flicked it past Daniel and into the back of the net to give LAFC the 2-goal advantage it needed to move into 1st in the West! With our result secure, everyone turned to their phones, and turned into Dynamo fans! Alas, hopes were dashed in the 6th minute of stoppage time, as a Puig penalty call turned into a Pec rocket, and the score was leveled at 1. With the top spot seemingly gone, LAFC had to hope for a miracle, and we got one in the form of Burbank's own Daniel Steres! An 11th-minute stoppage-time header gave Houston an improbable 2-1 win, and handed LAFC the top spot in the West. It was an unreal scene at BMO!Also on this episode, we remember jewelry made from a plane and wish our LAFC2 boys well in their first playoff match. What a night at BMO! Now, as Ilie said, "the best is yet to come!"

So much for tuning up and getting in form for the 2024 U.S. Open Cup Final. LAFC drops their road match to Dallas, 3-1, behind another lackluster performance amid roster rotation. LAFC may not have won a match in their last 5, but a chance to win a trophy comes to BMO on Wednesday, in the form of SKC. Let's chat about it on Episode 328.An awful turnover right to the feet of Dallas' best goal scorer, Petar Musa, sees the home side take a 1-0 lead into the half. LAFC looked to stem the tide at the hour mark, subbing on Bouanga and Giroud, but in the next few minutes that followed, all hell broke loose for the Black and Gold. Two goals in two minutes from Dallas saw the lead balloon to 3, and if not for an Atuesta goal late, LAFC would be shut out by Texas for a third straight match.LAFC turns its focus to the Open Cup Final on Wednesday, and everyone looked to come out of this one injury-free. Back at BMO, LAFC misses an opportunity to clinch a playoff spot but definitely doesn't miss a chance to welcome back Carlos Vela!LAFC played an excellent first half defensively... right up until it didn't. One mistake by Aaron Long, one pass by Bukari, and one shot on target was all Austin needed to take a lead into the locker room. An hour in, LAFC brings on Ilie and the teenager, David Martinez, and it pays dividends. Giroud, Atuesta, and Hollingshead combine for a great buildup, finding a streaking Martinez in the box to make it 1-1. Hugo did what he needed to to preserve a point, but LAFC couldn't find the winner and the match ended in a draw. Los Angeles Football Club (LAFC) officially welcomed French international forward to the club with a press conference on Thursday afternoon at the InterContinental Los Angeles Downtown. A 2018 World Cup Champion and France's all-time leading goal scorer, Giroud joins LAFC as a Designated Player and has signed with the team through the 2025 MLS season with an option through 2026. Giroud will be eligible for roster selection to the Leagues Cup Round of 32 match next week
