Earlier this month, the organizations EverythingALS and Vision 2030 announced a partnership with the Allen Institute to accelerate research into the progression of amyotrophic lateral sclerosis, also known as Lou Gehrig's disease or ALS. The disease is a progressive neurodegenerative condition where patients lose the ability to walk, use their hands, talk and eventually breathe. But the new investment will be geared towards finding using AI-powered technology to look for a cure at the cellular level. ALS remains a devastating disease, but the field is now at an inflection point with a growing pipeline of precision therapies, trial-ready sites, and a new generation of clinician scientists - yet barriers persist. There is disease heterogeneity, limited trial access for patients, and rising development costs. For an update on ALS clinical trials and the Healey ALS MyMatch program, a centralized platform that matches participants to trials, we spoke with Dr. Suma Babu. Dr. Suma Babu is an associate professor of neurology at Harvard Medical School and co-director of the Neurological Clinical Research Institute at Mass General Brigham. We need more – likely many more – effective treatments to end ALS. Progress in ALS research has been too slow for too long but, in 2024, we're witnessing significant advances and a new pace of progress. The past few years have see more treatments for ALS in clinical trials than ever before. Still, most drugs that reach trial fail, in any disease. However, the more experimental treatments that make it to trials – with good preclinical evidence to support them – the more likely there are to be successes. Recently, we have even seen approvals for new ALS treatments, such as tofersen, a genetic treatment developed by Biogen for SOD1-related ALS, sold under the brand name Qalsody. Dr. Stephanie Fradette was one of the people behind milestone. As the VP Head of Neuromuscular Development at Biogen, she oversees the company's research and development of treatments for ALS and related diseases. During this year's alumni reunion Partio, we caught up with some alums right on campus. One of those alumni was Daniel Barvin ‘18.Daniel has held many different roles, in both financial advising and oil and gas sectors, and eventually earned an MBA from Rice Business in 2018. But in December of that same year that he tested positive for the C9orf72 gene expansion, which means may develop ALS later in life. Daniel's father, aunt, and uncle all have ALS. In March 2023, ALS TDI announced the launch of the ALS Research Collaborative (ARC). ARC's goal is to learn more about the underlying biology of ALS to help accelerate treatments. The study is powered by the participation of people with ALS, who generously share data about their disease, their genetics, and their progression. This data then informs ALS TDI's research to find treatments for ALS. It is also shared with the broader ALS research community through the ARC Data Commons, an online portal allowing researchers to access years of data dating back to when the program was founded as the Precision Medicine Program, in 2014. This program is powered by participants – people with ALS all over the world who have chosen to share their data with ALS TDI in order to help accelerate research. ALS TDI works hard to make sure that participation is as easy as possible for these volunteers – nearly every aspect of the program can be completed from participants' homes. For more than 20 years, riders have been coming to the Tri-State Trek for an unforgettable weekend of riding bikes, seeing friends, and supporting ALS Research. The Trek is ALS TDI's signature fundraiser, and it has raised more than $10 million dollars to support our research to end ALS. It has also become something of a family affair, with riders who have been showing up for years, or even decades. Rinaldo and Sara Dorman have been an essential part of the Trek for 11 years. Rinaldo is the captain of Team America, one of the Trek's biggest and most dedicated teams. His wife Sara is a longtime volunteer who serves as the leader of the lunch crew – a job that requires feeding hundreds of hungry, sweaty cyclists every day. EPISODE 192: COUNTDOWN WITH KEITH OLBERMANN A-Block (1:43) SPECIAL COMMENT: "A group of Trump guys surrounded an Antifa kid and started pounding the living shit out of him," Tucker Carlson texted to his producer on January 6, 2021. "It was three against one, at least. Jumping a guy like that is dishonorable obviously. It's not how white men fight." THAT is the thing Fox found, at the last moment, just as its trial with Dominion was about to begin, that it could not afford to let out publicly; not, at least, while Tucker Carlson was still its employee. I raised the likelihood that there was something just like this, something even worse than everything that had already come out. Got that one right. That's not only why Murdoch settled - it's a major reason that Murdoch fired Carlson. It also suggests Fox is the source of the text (Exhibit 276 to be precise) because it serves a purpose for Murdoch now. It dirties up his EX-employee on his way out and makes him a huge risk for anybody else to hire. Unfortunately it also segues into CNN's disastrous decision to double-down on its live Town Hall next week with Trump. Not only was its moderator, Kaitlan Collins, still working FOR Tucker Carlson at The Daily Caller seven years ago, but a cache of her articles there shows grotesque indifference to human suffering to say nothing of Islamophobia. Her "ICE BUCKET CHALLENGE HIPSTERS: THESE GUANTANAMO DETAINEES DID IT FIRST" not only says Khalid Sheik Mohammed "took the challenge a record 183 times" but mocks the 'challenge' itself - which was to raise money for ALS Research and Treatment. She also mocked Syrian refugees in another article, and authored a pair of homophobic tweets in college. Yet it is Kaitlan Collins that CNN's political director thinks can stand up to Trump lying in real time. She can "prod, ask questions, follow up, and try to get as revealing answers as possible." It is this David Chalian quoted in a Vanity Fair piece explaining "we obviously can't control what Trump says - that's up to him" and how Trump's sedition and crimes "does not make our approach any different" to him. CNN is standing on a precipice. If it does not cancel the Town Hall, fire Licht and Chalian, and fire or demote Collins, both its remaining journalistic credibility and any possible re-sale value it still has, will be erased. Because its last line of defense against Trump was getting a pay check as recently as seven years ago from Tucker "It's not how white men fight" Carlson to mock ALS, torture, and refugees. B-Block (18:30) POSTSCRIPTS TO THE NEWS: When Kevin McCarthy goes to the White House Tuesday to dictate terms on the debt ceiling, Biden may have a surprise for him: defaulting on US debt may be literally unconstitutional. And Chuck Schumer supposedly talked to Dianne Feinstein on Monday, except he didn't actually TELL anybody that. Politico merely read it of a photograph of the talking points he was carrying in his hand! I was so grateful to tour the lab and attend the ALS TDI Summit which was a day of updates on the progress of ALS Research. I wrapped up the weekend by attending their White Coat Affair gala which is a powerful evening that brings together and honors those living with ALS and remembers those whom we've lost.I catch up with 5 inspirational guests as we chat about the love, passion and hope behind the lab doors of this ALS research lab. Antibodies are an essential part of our bodies' defenses against diseases. They are specialized proteins that attack and kill foreign invaders like infectious bacteria and viruses. Antibodies are normally naturally produced by our immune system in response to an infection. However, in recent years, scientists have learned to produce antibodies in a lab to treat a variety of diseases. Several Antibody treatments have been investigated in ALS, including tegoprubart, which was invented by ALS TDI and recently successfully completed a phase 2a clinical trial. Dr. Silvia Pozzi is Assistant Professor at the Université Laval's Brain Research Center in Quebec City specializing in ALS Research. She recently was an author on a paper reviewing the current state of antibody therapies for ALS.
