Galan Lithium (ASX: GLN) managing director Juan Pablo Vargas de la Vega joins Small Caps to discuss to recent signing of significant offtake agreement with global mining giant Glencore for its Hombre Muerto West (HMW) project in Argentina. The binding offtake deal is for lithium chloride and/or lithium carbonate from Phase 1 production at HMW. Notably, Glencore has also offered to potential provide Galan with a $107 million to $151 million in support of a financing prepayment facility for the project. Galan considers the Glencore agreement as a validation of the world class quality of the HMW project. The company has already commenced construction of the Phase 1 development at HMW and remains on track for first pond fill in Q1 2024 and to achieve initial production in the first half of 2025. At the same time, Galan is continuing to advance discussions for the development of Phase 2 at HMW. Abstract: Insoluble fibrillar tau, the primary constituent of neurofibrillary tangles, has traditionally been thought to be the biologically active, toxic form of tau mediating neurodegeneration in Alzheimer's disease. More recent studies have implicated soluble oligomeric tau species, referred to as high molecular weight (HMW) due to its properties on size exclusion chromatography, in tau propagation across neural systems. These two forms of tau have never been directly compared. We prepared sarkosyl insoluble and HMW tau from the frontal cortex of Alzheimer patients compared their properties using a variety of biophysical and bioactivity assays. Sarkosyl insoluble fibrillar tau is comprised of paired helical filaments seen by electron microscopy (EM), and is more resistant to proteinase K, compared to HMW tau which presents only sparse, short, filaments. Sarkosyl insoluble and HMW tau are nearly equivalent in potency in a HEK cell bioactivity assay for seeding aggregates. Injection of sonicated fibrils or native HMW tau reveals similar local uptake into hippocampal neurons in PS19 Tau transgenic mice. However, the HMW preparation appears to be far more potent in inducing a glial response, and promotes rapid propagation of misfolded tau to distal, anatomically connected regions, such as entorhinal and perirhinal cortices. These data suggest that soluble HMW tau has similar properties with regard to tau seeding potential but may be equal or even more bioactive with respect to propagation across neural systems and activation of glial responses, both relevant tau-related Alzheimer phenotypes. H., Kins, S., Eggert, S. Abstract: The amyloid precursor protein (APP), a key player in Alzheimer's disease (AD), is part of a larger gene family, including the APP like proteins APLP1 and APLP2. They share similar structures, form homo- and heterotypic dimers and exhibit overlapping functions. We investigated complex formation of the APP family members via two inducible dimerization systems, the FKBP-rapamycin based dimerization as well as cysteine induced dimerization, combined with coimmunoprecipitations and Blue Native (BN) gel analyses. Within the APP family, APLP1 shows the highest degree of dimerization and high molecular weight (HMW) complex formation. Interestingly, about 20% of APP is dimerized in cultured cells while about 50% of APP is dimerized in mouse brains, independent of age and splice forms. Furthermore, we could show that dimerized APP originates mostly from neurons and is enriched in synaptosomes. Finally, BN gel analysis of human cortex samples shows a significant decrease of APP dimers in AD patients compared to controls, suggesting that loss of dimers of full-length APP might correlate with loss of synapses in the process of AD. Hawkmoon Resources CEO Branden Haynes and Vice President of Exploration Thomas Clarke joined Steve Darling from Proactive to share news the company has received assay results for 12 drill holes from the summer drill program on the Wilson gold project in Quebec. Clarke telling Proactive, every hole intersected at least one altered and mineralized zone. The two key holes were HMW 21-24 and HMW 21-25. A near surface intercept was encountered in HMW 21-24 with 10.0 grams per tonne while HNW 21-25 returned 2 meters at 6.3 grams per tonne at 109 meters down hole. Haynes telling Proactive his thoughts on the summer program and what is means for the project in general.

And you know what that means. When this keeps going on too long you will eventually becoming irrelevant. And fail to achieve your mission. Tricia Wang, our guest this week, has an interesting take on why this is happening... Designers have the tendency to get in their own way. What does this mean? It means that we've adopted anti-patterns that actually prevent us from achieving success on the level we desire. According to Tricia we need to uncenter ourselves and transition from being a guru to being a guide. If this sounds abstract don't worry. Tricia provides some very practical things you can do in your day-to-day work to make this happen. Things like sharing unfinished work, ditching the HWM and much more. So when you feel like you could be making a bigger impact on your organisation, but aren't sure how to do that. This episode might just hold the clue you're looking for. In order to boost the economy of Houston, Houston Center for Literacy strengthens providers of adult education who, in turn, provide necessary skills for underemployed citizens to fulfill their potential and become economically self-sufficient.  Houston Money Week (HMW) is a community wide initiative to empower families and individuals with financial education.  Through the collaboration and coordinated effort of organizations across Houston and surrounding areas, including businesses, financial institutions, schools, libraries, non-profits, government agencies and the media, individuals of all ages will have access to financial resources and ongoing opportunities.  Jackie Aguilera has over 20 years experience in adult education and curriculum development. Native Houstonian Jennifer M. Leighton lived in Washington, DC and San Antonio, TX before returning home to Houston to develop JenMarie Consulting. Jennifer obtained a Bachelor of Arts degree with a major in Political Science from George Washington University and a Master of Public Administration degree from University of Texas at San Antonio. After graduating with her Master's degree, Jennifer began working for Texans Can Academies in San Antonio at San Antonio Can High School in fund development and event planning. She also served as Development Director at Houston Can Academy. She currently works for Texas Capital Bank in community development.  Jennifer has served on many boards including Multicultural Education and Counseling through the Arts, George Washington Alumni Club in Houston, Bexar County Community Plan, and YMCA of Greater San Antonio. In Episode 16 we focus on our IDEO Futures design theme of "Hacking Creativity and Design". Specifically, we took Episode 16 on a mission to explore the future of a broad spectrum of digital media: the media we listen to, the media we write on (blogs and Twitter that is, not so much paper), the media we broadcast with, and even the media we print with. We're very honored to have John Lilly join us as our guest and co-host for the middle portion of Episode 6. The three of us got together to discuss the idea of using social media as a tool for creativity and design. How do we choose to express ourselves in different types of social media? Should we be consistent across everything, or tailor our content to fit? How does Twitter fit into our workflow? Is it an aid to creativity? And how about the recent emergence of broadcasting via Periscope and Meerkat? John is not only a prolific tweeter with a massive Twitter following, but is one of us, a dreamer, a builder, and an operator. He loves to ship stuff. He is a partner at Greylock Partners, where he has invested in a few companies you may have heard of, including Tumblr, Instagram, and Dropbox. Before joining Greylock he was the CEO of Mozilla and a serial entrepreneur. He is also on the faculty of the Stanford d.school, and last year he and Diego taught a class there about hacking refrigerators. We think you'll agree with us when we say that it was educational and fun to have John join us as a co-host. Back in Episode 15 Joe promised us a new "How Might We?" segment on this new-fangled thing called Meerkat, and so went there. Meerkat, Periscope — we find them both fascinating and entrancing, both as users of these apps and as business designers trying to make sense of it all. Today we learned about Houston Money week and visited with HMW partner Money Management International. We were joined by Regional Director of Education and Community relations for MMI Rudy Cavazos Jr. We also discussed working with a non-profit credit counseling agency, financial literacy and credit card fraud   Recognized as a credit authority, Cavazos has represented MMI in numerous television, radio, print and online interviews.  He has hosted a weekly money management segment on a major San Antonio television station.  Also, Cavazos' quotes have appeared in national periodicals such as Wall Street Journal, TIME Washington Post, Chicago Tribune and Newsweek magazine. Locally, you can find his quotes and articles in newspapers such as The Courier and The Galveston Daily News. Novel biodegradable gene carriers based on oligomerized polyamines

Gene therapy is a very promising approach to treat or to prevent diseases. However, progress in this field is hindered by lack of suitable vectors. Current research focuses on the development of novel nonviral biodegradable gene carriers with improved gene transfer activity and low toxicity. In the course of this thesis, a library of degradable DNA compacting domains based on oligomerized polyamines was synthesized and analyzed. Degradation of the originated polymers was either based on site-specific reductive cleavage of disulfide bonds or on time-dependent ester/amide hydrolysis. DNA binding activity, polyplex stability, transfection efficiency, toxicity, and hemocompatibility studies were performed in order to identify promising candidates. Some of the novel gene carriers, especially the degradable oligoethylenimine (OEI) derivatives were successfully applied for in vitro transfection and could easily compete with the current ‘golden standard’ linear polyethylenimine with an average Mw of 22 kDa (PEI22lin). Furthermore, screening results revealed critical structure activity relationships which were very helpful for improving the polymer design. According to transfection and biocompatibility results, efficiency and toxicity correlated to some degree. Polymers with an overall high charge density and a high molecular weight like OEI-HD-1 provided polyplex stability and formed small uniform particles. On the other hand these polymers tended to induce erythrocyte aggregation and exhibited a pronounced cytotoxicity when applied at high concentrations. Polycation with a lower molecular weight (~ 10 kDa) like e.g. OEI-IP-1 were essentially nontoxic, but had to be applied at high concentrations in order to achieve efficient gene transfer. Intrinsic membrane activity of certain polymers could damage cellular membranes but may also trigger endosomal release and therefore boost transfection activity. Crosslinking of OEI 800 with 1,6-hexanedioldiacrylate resulted in highly efficient degradable polycations. Different reaction temperatures during OEI-HD-1 synthesis had a strong impact on molecular weight and the ester/amide ratio. Despite structural differences, both OEI-HD-1 (synthesized at 60°C) and lt-OEI-HD-1 (synthesized at 20°C) possessed equal gene transfer activity as the ‘golden standard’ PEI22lin when applied at their optimal polymer/DNA-ratio (w/w). It was important to note that lt-OEI-HD-1, the LMW-derivative which is predominantly based on ester linkages, was significantly less toxic than its HMW amide-linked counterpart. OEI-HD displayed a very promising basis for the development of further powerful gene carriers. A two-step synthesis protocol was established in order to generate OEI-HD cores bearing excessive linker which could be subsequently modified with various functionalities like spermine. OEI-HD-Sper pseudo-dendrimers were characterized by a pronounced intrinsic membrane activity and possess high transfection efficiency. Since current nonviral vectors are still very inefficient as compared to their viral competitors, natural viruses present an ideal example educating us how to further optimize polycationic gene carriers in terms of specific cell-targeting and improved endosomal release. Modification of polyplexes towards a “smart” virus-like system was achieved in the following way. Degradable DNA compacting domains (OEI-HD-1) were utilized for complex formation. Furthermore, epidermal growth factor (EGF) was incorporated as targeting ligand into OEI-HD-1 polyplexes and thus allowed cell-specific cellular uptake via the EGF receptor (EGFR). Gene transfer potential of EGFR-targeted degradable polyplexes was further improved by applying technologies which promoted the endosomal release of endocytosed particles. Photochemical intracellular release (PCI) is based on accumulation of amphiphilic photosensitizers (PS) in endosomal membranes. Illumination of PS pre-treated transfected cells results in activation of the PS and subsequent light-induced rupture of endocytic vesicles. Combination of biological (EGFR) and physical (PCI) targeting greatly enhanced reporter gene delivery mediated by OEI-HD-1 polyplexes. Finally, the incorporation of membrane active melittin derivatives into EGF/OEI-HD-1 polyplexes was the first example of a biodegradable synthetic virus for gene delivery.
