Joey Henderson returns to the podcast to talk about heat pumps and inverters. The reversing valve, defrost cycles, and auxiliary heat can cause confusion for people who have primarily worked with furnaces or straight-cool A/C systems. Heat pumps use defrost cycles and bring on the auxiliary heat when the coil is ice-bound, which can present a challenge; we need to maintain cold coils without going into defrost all the time. Even though heat pumps were significantly less effective in years past, we will still see reduced performance in very cold conditions with the newer inverter-driven systems. Proper design, installation, and commissioning will also help occupants get the best performance out of their heat pumps. Inverters offer plenty of advantages for the cooling aspect of heat pumps, too, especially when it comes to achieving longer runtimes for dehumidification. They can also float their coil temperature, much like how refrigeration systems can use floating suction or head pressure. Condensate assemblies absolutely must be run properly to prevent backed-up drains and other related problems. Liquid line sizing and proper commissioning are also especially crucial for ductless inverter-driven systems. Abstract: Biomolecular condensates regulate a wide range of cellular functions from signaling to RNA metabolism1,2, yet, the physiologic conditions regulating their formation remain largely unexplored. Biomolecular condensate assembly is tightly regulated by the intracellular environment. Changes in the chemical or physical conditions inside cells can stimulate or inhibit condensate formation3,5. However, whether and how the external environment of cells can also regulate biomolecular condensation remain poorly understood. Increasing our understanding of these mechanisms is paramount as failure to control condensate formation and dynamics can lead to many diseases6,7. Here, we provide evidence that matrix stiffening promotes biomolecular condensation in vivo. We demonstrate that the extracellular matrix links mechanical cues with the control of glucose metabolism to sorbitol. In turn, sorbitol acts as a natural crowding agent to promote biomolecular condensation. Using in silico simulations and in vitro assays, we establish that variations in the physiological range of sorbitol, but not glucose, concentrations, are sufficient to regulate biomolecular condensates. Accordingly, pharmacologic and genetic manipulation of intracellular sorbitol concentration modulates biomolecular condensates in breast cancer, a mechano-dependent disease. We propose that sorbitol is a mechanosensitive metabolite enabling protein condensation to control mechano-regulated cellular functions. Altogether, we uncover molecular driving forces underlying protein phase transition and provide critical insights to understand the biological function and dysfunction of protein phase separation. Abstract: Stress conditions can cause the relocalization of proteasomes to condensates in yeast and mammalian cells. The interactions that facilitate the formation of proteasome condensates, however, are unclear. Here, we show that the formation of proteasome condensates in yeast depends on long K48-linked ubiquitin chains together with the proteasome shuttle factors Rad23 and Dsk2. These shuttle factors colocalize to these condensates. Strains deleted for the third shuttle factor gene, DDI1, show proteasome condensates in the absence of cellular stress, consistent with the accumulation of substrates with long K48-linked ubiquitin chains that accumulate in this mutant. We propose a model where the long K48-linked ubiquitin chains function as a scaffold for the ubiquitin binding domains of the shuttle factors and the proteasome, allowing for the multivalent interactions that further drive condensate formation. Indeed, we determined different intrinsic ubiquitin receptors of the proteasome (Rpn1, Rpn10, and Rpn13) are critical under different condensate inducing conditions. In all, our data support a model where the cellular accumulation of substrates with long ubiquitin chains, potentially due to reduced cellular energy, allows for proteasome condensate formation. This suggests that proteasome condensates are not simply for proteasome storage, but function to sequester soluble ubiquitinated substrates together with inactive proteasomes. Abstract: Altered RNA metabolism is a common pathogenic mechanism linked to familial and sporadic Amyotrophic lateral sclerosis (ALS). ALS is characterized by mislocalization and aggregation of TDP-43, an RNA-binding protein (RBP) with multiple roles in post-transcriptional RNA processing. Recent studies have identified genetic interactions between TDP-43 and Ataxin-2, a polyglutamine (polyQ) RBP in which intermediate length polyQ expansions confer increased ALS risk. Here, we used live-cell confocal imaging, photobleaching and translation reporter assays to study the localization, transport dynamics and mRNA regulatory functions of TDP-43/Ataxin-2 in rodent primary cortical neurons. We show that Ataxin-2 polyQ expansions aberrantly sequester TDP-43 within ribonucleoprotein (RNP) condensates, and disrupt both its motility along the axon and liquid-like properties. Our data suggest that Ataxin-2 governs motility and translation of neuronal RNP condensates and that Ataxin-2 polyQ expansions fundamentally perturb spatial localization of mRNA and suppress local translation. Overall, these results indicate Ataxin-2 polyQ expansions have detrimental effects on stability, localization, and translation of transcripts critical for axonal and cytoskeletal integrity, particularly important for motor neurons. Gavrilik et al. on Wednesday 23 November By improving the Bose-Einstein condensate model of dark matter through the repulsive three-particle interaction to better reproduce observables such as rotation curves, both different thermodynamic phases and few-particle correlations are revealed. Using the numerically found solutions of the Gross-Pitaevskii equation for averaging the products of local densities and for calculating thermodynamic functions at zero temperature, it is shown that the few-particle correlations imply a first-order phase transition and are reduced to the product of single-particle averages with a simultaneous increase in pressure, density, and quantum fluctuations. Under given conditions, dark matter exhibits rather the properties of an ideal gas with an effective temperature determined by quantum fluctuations. Characteristics of oscillations between bound and unbound states of three particles are estimated within a simple random walk approach to qualitatively models the instability of particle complexes. On the other hand, the density-dependent conditions for the formation of composites are analyzed using chemical kinetics without specifying the bonds formed. Finite temperature effects on magnetized Bose-Einstein condensate stars by Gretel Quintero Angulo et al. on Monday 12 September We study the role of temperature and magnetic field on the equation of state and macroscopic properties of Bose-Einstein condensate stars. These compact objects are composed of a condensed gas of interacting neutral vector bosons coupled to a uniform and constant magnetic field. We found that the main consequence of a finite temperature in the magnetized equations of state is to increase the inner pressure of the star. As a consequence, magnetized hot Bose-Einstein condensate stars are larger and heavier than their zero-temperature counterparts. However, the maximum masses obtained by the model remain almost unchanged, and the magnetic deformation of the star increases with the temperature. Besides, augmenting the temperature reduces the number of stable stars, an effect that the magnetic field enhances. Eric Kaiser returns to the podcast to discuss high-voltage wiring, low-voltage wiring, and condensate assemblies as they relate to systems thinking. On the high-voltage side, the disconnect should be in a secure location, and it should be able to keep water out. The wires should be appropriately sized, have an appropriate level of tension, and should not be vulnerable to chafing or abrasion. Overall, best practices include using proper grommets and ensuring that you have a solid connection. Do not run high voltage wiring in parallel with low-voltage or control wiring. It's also worth noting that double-lugging is a poor practice that is against code. On the low-voltage side, you also need to be careful of where you route your wires to avoid induction, contact with hot surfaces, or abrasion. The insulation ratings also need to be appropriate. We can think of the condensate assembly as its own system. Condensate drains have uphills and downhills, and they may have traps, vents, and cleanouts throughout. Cleanouts and vents may be confused for each other, but cleanouts allow the technician to access and clean the drain. Cleanouts are also capped when in use, but vents are not. The location of a vent can help equalize the siphoning effects of pressurization. Condensate systems also consist of pans and switches. In those cases, redundancy is desirable to prevent overflowing. Secondary drain pans should be large enough to overlap with the primary pan, especially in horizontal air handlers.
