Sequester is a word that has important legal and scientific uses, and a long history besides. In fact, it can be traced back to the Latin preposition secus, meaning, well, “beside” or “alongside.” Setting someone or something apart (figuratively “to the side”) from the rest is sequester's raison d'être. We frequently hear it in the context of the courtroom, as juries are sometimes sequestered for the safety of their members or to prevent the influence of outside sources on a verdict. It is also possible, legally speaking, to sequester property—sequester can mean both “to seize” and “to deposit” property by a writ of sequestration. The scientific sense of sequester most often encountered these days has to do with the binding or absorption of carbon. Kelp forests, for example, sequester massive amounts of carbon dioxide during photosynthesis, keeping it “apart” from the atmosphere—by some estimates doing so twenty times as much as terrestrial forests. You might even say kelp's got this sequestering thing locked up.

Cquesta is developing technology to leverage the natural ability of plants to sequester more carbon in the soil.
The idea behind the Icelandic company Carbfix is simple: pack water full of carbon dioxide (literally carbonate it, like a SodaStream) and inject it deep underground into Iceland's porous basaltic rock. Minerals in the rock dissolve in the water, where they react with the CO2 to become calcium carbonates. The carbon effectively becomes rock, which it will remain, for all intents and purposes, permanently. Or at least thousands and thousands of years. It is as long-term as carbon sequestration gets.The idea dates back to 2006, but pilot injections didn't begin until 2013 and it wasn't until 2016 that a study published in Science confirmed that 95 percent of the CO2 in the water was mineralizing within two years — far faster than most had assumed possible. Since it started, Carbfix has sequestered almost 100,000 metric tons of CO2 at its original site, but that is just a drop in the bucket compared to what it believes is possible. It has plans to make Iceland a major international carbon-burial hub and to replicate its technology in other geographies, maybe even in the shallow ocean. When I visited the Carbfix operation in October and saw it in action, I was extremely intrigued and had a million more questions, so last week I got in touch with Ólafur Teitur Guðnason, Carbfix's head of communications, to talk about where the company gets the CO2 it buries, where it plans to get it in the future, whether burial can work in other kinds of rocks and geographies, and exactly how much carbon Iceland can store. Abstract: The loss of nuclear TDP-43 localization and its accumulation in cytoplasmic aggregates are hallmarks of neurodegeneration and major therapeutic targets. We recently demonstrated that TDP-43 binding to endogenous nuclear GU-rich RNAs sequesters TDP-43 in the nucleus and restricts its passive nuclear export. Here, we tested the feasibility of synthetic RNA oligonucleotide-mediated augmentation of TDP-43 nuclear localization. Using biochemical assays, we compared the ability of GU-rich oligonucleotides to engage in multivalent, RRM-dependent binding with TDP-43 and identified (GU)16 as a strong multivalent binder. When transfected into cells, unlike monovalent oligonucleotides that displaced TDP-43 from the nucleus, (GU)16 preserved steady-state TDP-43 nuclear localization and prevented transcriptional blockade-induced TDP-43 mislocalization. RNA pulldowns from (GU)16-transfected cells confirmed that (GU)16 induced high molecular weight RNP complexes, incorporating TDP-43 and possibly other GU-binding proteins. Transfected (GU)16 caused partial failure of TDP-43 cryptic exon repression, likely because the high-affinity oligonucleotides diverted TDP-43 from endogenous RNAs. Thus, while GU-rich oligonucleotides can attenuate TDP-43 mislocalization, optimization is needed to avoid TDP-43 loss of function. Copy rights belong to original authors. Abstract: Altered RNA metabolism is a common pathogenic mechanism linked to familial and sporadic Amyotrophic lateral sclerosis (ALS). ALS is characterized by mislocalization and aggregation of TDP-43, an RNA-binding protein (RBP) with multiple roles in post-transcriptional RNA processing. Recent studies have identified genetic interactions between TDP-43 and Ataxin-2, a polyglutamine (polyQ) RBP in which intermediate length polyQ expansions confer increased ALS risk. Here, we used live-cell confocal imaging, photobleaching and translation reporter assays to study the localization, transport dynamics and mRNA regulatory functions of TDP-43/Ataxin-2 in rodent primary cortical neurons. We show that Ataxin-2 polyQ expansions aberrantly sequester TDP-43 within ribonucleoprotein (RNP) condensates, and disrupt both its motility along the axon and liquid-like properties. Our data suggest that Ataxin-2 governs motility and translation of neuronal RNP condensates and that Ataxin-2 polyQ expansions fundamentally perturb spatial localization of mRNA and suppress local translation. Overall, these results indicate Ataxin-2 polyQ expansions have detrimental effects on stability, localization, and translation of transcripts critical for axonal and cytoskeletal integrity, particularly important for motor neurons. Discover how our food production and even our yards and gardens can play a role in reducing CO2 levels. We're joined by Ben Cloud, CEO of Biodel AG Inc., a Maricopa, Arizona company that recently introduced Sequester, a soil treatment that restores the soil's ability to capture and hold CO2 that also promotes improved plant growth. Sequester is available for farming applications and for home use. A $12.81 8 oz bottle of Sequester can be used to improve the carbon sequestion capability of up to 2,500 square feet of yard and gardens. It is produced using a blend of cyanobacteria — the source of much of the breathable oxygen on the planet. In addition to helping capture carbon, Sequester reputedly reduces water consumption, salinity and nitrogen use by 30%.Carbon dioxide is humanity's most pressing problem. Since the dawn of the Industrial era about 250 years ago, humans have pumped more than 1.5 trillion tonnes of CO2 into the atmosphere, warming the planet by 1.8 degree Fahrenheit and bringing on the Climate Crisis. In addition to reducing emissions to zero, we need to remove this CO2 to start to cool the planet back to the temperatures in which we and the rest of the species on the planet evolved. One of many discussed solutions to the climate crisis, besides of course emission reduction, is the potential of storing carbon in the ocean, for example using marine biomass such as seaweed or more complicated chemical mechanisms. In order to really understand how those solutions might be working, though, today's episode will start out with actually explaining how the ocean takes up and processes carbon in the first place. Not only are we talking about that, however, but we are also discussing what is behind that huge promise of blue carbon, as it is often termed. And to make it even more exciting, on this episode I don't have one guest, but five! The groundwork and basis for this long conversation is a paper published by seven phenomenal female scientists who have a very multidisciplinary background within the marine science and policy sector. The paper is called The Promise of Blue Carbon Climate Solutions: Where the Science Supports Ocean-Climate Policy written by Anne B. Christianson, Anna Cabre, Blanca Bernal, Stacy K. Baez, Shirley Leung, Alicia Perez-Porro and Elvira Poloczanska. With five of these authors I dissected their paper, talking about the different mechanisms of the oceanic carbon cycle, the ways in which climate solutions can enhance or support these mechanisms, and to what degree we already have policy supporting that. With a group of such interdisciplinary guests we really get a broad perspective on the issue or potential of blue carbon and you will see, also throughout the entire series of next episodes, that opinions are sometimes quite far apart. Nonetheless, in this episode we sort of set the ground rules for understanding the carbon cycle in the first place and getting a sense of how incredibly difficult it is to create policy supporting that science, and vice versa. Although a tax cut may have been a nice Christmas gift, it needs some follow-on spending cuts to work. At best, Republicans have eaten their dessert first, waiting to eat their spending cut vegetables later; at worst, they will have increased the federal deficit by another trillion dollars or more. By most estimates, even stimulating economic growth will not fully pay for the tax cut. Republicans will now have to undertake the politically courageous step of cutting federal spending. It will be difficult to make spending cuts without touching Medicare or Social Security, which President Trump has said are off limits. Meanwhile there is pressure to undo the sequester, automatic cuts on spending no one liked, but which have at least kept spending growth down. Ideally, Republicans would have disciplined themselves to do tax and spending cuts at the same time. Tax cuts may come and go, but the federal debt remains forever, it seems.
