And there's the unmistakable coo of the mourning dove. Later, we hear chirps from an iconic grassland native, but this one is a rodent, not a bird. Prairie dogs, a type of ground squirrel, stand upright like fluffy meerkats to watch over their underground tunnels. We end our tour of Colorado's prairies with birds at Reservoir Ridge … near the city of Fort Collins. Bobolinks fly all the way from South America each year to find a mate here. The McConney family decides to take a Christmas vacation. It was a welcome break after the troubling situation the family found themselves in. Weeks before Christmas, Heather McConney opened her son's phone, stumbling on disturbing conversations.  13-year-old Evan McConney had been communicating with a stranger through the online game Roblox. The two also reportedly communicated through Oculus, a virtual reality device. The parents then made all the right steps. They took away Evans' devices and turned them over to Layton police.  The McConney family returned home from vacation on Monday. When the family woke up, Tuesday morning Evan was gone.  An investigation revealed Evan McConney and his online “friend” had been exchanging  “sexual and inappropriate” messages for about three weeks. While  Evan's mom had taken steps to end the contact, the two reportedly found new ways to talk. Evan McConney joined a Discord chat at his new friend's invitation.  It was through those chats that the two allegedly arranged to have McConney picked up from his home. Evan even took his passport with him, after his new friend encouraged him.  The "friend" is Aaron Zemen...a 26-year-old man, living in Arizona.  Zemen has several social media accounts revealing his admitted dog fetish and that he is a furry. A furry is a person who is an enthusiast for animal characters with human characteristics. Abstract: As social animals, our health depends in part on interactions with other human beings. Yet millions suffer from chronic social isolation, including those in nursing/assisted living facilities and people experiencing chronic loneliness. Perhaps the most egregious form of chronic isolation is seen in criminal justice system, where approximately 80,000 people are housed, on any one day, in solitary confinement. In this study, we developed a model of isolation that starts in adulthood. Mice (C57BL/6J) were born and raised in an enriched environment until 4 months of age and then either maintained in that environment or moved to social isolation for 1 or 3 months. We then examined neuronal structure, catecholamine and brain derived neurotrophic factor (BDNF) levels, and CNS-mediated behaviors, comparing social isolation to enriched environment controls. We found there were significant changes in neuronal volume, dendritic length, neuronal complexity, and spine density that were dependent on brain region, sex, and duration of the isolation. Isolation altered dopamine in the striatum and serotonin levels in the forebrain in a sex-dependent manner, and also reduced levels of BDNF in the motor cortex and hippocampus of male but not female mice. To determine if SI altered a behavior, we tested mice in the open-field (general activity), the resident intruder paradigm (aggression), the tail suspension test (depression), and the Barnes maze (spatial memory). Adult male mice isolated for 1 month exhibited increased locomotor activity, aggression, and enhanced aspects of spatial memory, most of which remained after 3 months of isolation. After 3 months of isolation, mice also exhibited depressive behaviors. Similar (but not exact) results were seen in female mice, with the exception that the females did not show increased aggression. These studies show that isolation enforced in adulthood has significant impact on brain structure, neurochemistry, and behavior Copy rights belong to original authors. Abstract: Sleep apnea (SA) during pregnancy is detrimental to the health of the pregnancy and neonate, but little is known regarding long-lasting consequences of maternal SA during pregnancy on adult offspring. SA is characterized by repeated cessations in breathing during sleep, resulting in intermittent hypoxia (IH). We show that gestational IH (GIH) in rats reprograms the male fetal neuroimmune system toward enhanced inflammation in a region- and sex-specific manner, which persists into adulthood. Male GIH offspring also had deficits in the neural control of breathing, specifically in the ability to mount compensatory responses to central apnea, an effect that was rescued by a localized anti-inflammatory or microglial depletion. Female GIH offspring appeared unaffected. These results indicate that SA during pregnancy sex- and region-dependently skews offspring microglia toward a pro-inflammatory phenotype, which leads to long-lasting deficits in the capacity to elicit important forms of respiratory neuroplasticity in response to breathing instability. These studies contribute to the growing body of recent evidence indicating that SA during pregnancy may lead to sex-specific neurological deficits in offspring that persist into adulthood. E., Lin, Q. Abstract: Perturbations during critical time windows in the normal series of development can lead to adverse functional consequences that manifest later in life. Here, we report that prenatal stress (PNS) during the last week of gestation (E14-delivery) dramatically increased the number of proliferating neural stem/progenitor cells (NSC/NPCs) in the ependymal-subventricular zone (E-SVZ) of male mouse offspring. PNS did not cause significant cell death or a deficit in neuroblast migration to the olfactory bulb (OB). Olfactory behavioral tests showed that while prenatally stressed male mice displayed normal olfactory function in differentiating nonsocial odors, these mice showed impairment in discriminating different social smells. Bulk and single nucleus transcriptomic analyses combined with rescue assays using mitogen-activated protein kinases (MAPK) phosphorylation inhibitors revealed that PNS exposure during the critical period of neurogenesis promoted NSC/NPC proliferation in adult male offspring by sustaining MAPK3/1 activity. Compared to prenatally stressed males, their female littermates showed less change in the number of proliferating cells in the E-SVZ. Copy rights belong to original authors. Abstract: Rationale: The endocannabinoid system makes critical contributions to reward processing, motivation, and behavioral control. Repeated exposure to THC or other cannabinoid drugs can cause persistent adaptions in the endocannabinoid system and associated neural circuitry. It remains unclear how such treatments affect the way rewards are processed and pursued. Objective and methods: We examined if repeated THC exposure (5 mg/kg/day for 14 days) during adolescence or adulthood led to long-term changes in rats' capacity to flexibly encode and use action-outcome associations for goal-directed decision making. Effects on hedonic feeding and progressive ratio responding were also assessed. Results: THC exposure had no effect on rats' ability to flexibly select actions following reward devaluation. However, instrumental contingency degradation learning, which involves avoiding an action that is unnecessary for reward delivery, was augmented in rats with a history of adult but not adolescent THC exposure. THC-exposed rats also displayed more vigorous instrumental behavior in this study, suggesting a motivational enhancement. A separate experiment found that while THC exposure had no effect on hedonic feeding behavior, it increased rats' willingness to work for food on a progressive ratio schedule, an effect that was more pronounced when THC was administered to adults. Adolescent and adult THC exposure had opposing effects on the CB1-receptor dependence of progressive ratio performance, decreasing and increasing sensitivity to rimonabant-induced behavioral suppression, respectively. Conclusions: Our findings reveal that exposure to a translationally relevant THC exposure regimen induces long-lasting, age-dependent alterations in cognitive and motivational processes that regulate the pursuit of rewards. Abstract: Chronic stress during the developmental period of adolescence increases susceptibility to many neuropsychiatric diseases in adulthood, including anxiety, affective, and alcohol/substance use disorders. Preclinical rodent models of adolescent stress have produced varying results that are species, strain, sex, and laboratory-dependent. However, adolescent social isolation is a potent stressor in humans that has been reliably modeled in male rats, increasing adult anxiety-like and alcohol drinking behaviors, among others. In this study, we examined the generalizability and sex-dependence of this model in C57BL/6J mice, the most commonly used rodent strain in neuroscience research. We also performed a parallel study using social isolation in adulthood to understand the impact of adult social isolation on basal behavioral phenotypes. We found that six weeks of social isolation in adolescence beginning at postnatal day (PD) 28 produced a hypersocial phenotype in both male and female adults in multiple assays and a female-specific anxiolytic phenotype in the elevated plus maze, but it had no effects in other assays for avoidance behavior, fear conditioning, alcohol drinking, reward or aversion sensitivity, novel object exploration, or forced swim behavior in either sex. In contrast, social isolation in adulthood beginning at PD77 produced an anxiogenic phenotype in the light/dark box but had no effects on any other assays. Altogether, our results suggest that 1) adolescence is a critical period for social stress in C57BL/6J mice, producing aberrant social behavior in a sex-independent manner and 2) chronic individual housing in adulthood does not alter basal behavioral phenotypes that may confound interpretation of behavior following other laboratory manipulations.
