Guests: Dr. Alex Bach, ICREA (Catalan Institution for Research and Advanced Studies), and Dr. Bill Weiss, The Ohio State UniversityIn this journal club episode, Dr. Alex  Bach with the Catalan Institution for Research and Advanced Studies joins Dr. Bill Weiss from the Ohio State University. Dr. Weiss introduces the paper as one that's immediately applicable to the industry and answers a question he received a lot during his Extension career: What's the cost of moving cows? This research gives us some real data to help producers on cow management. (3:49)Dr. Bach states that grouping cows is necessary, and the goal is to feed cows as close to their requirements as possible. But in a practical world, that can be difficult, and producers may resist moving cows due to the increased work and perceived drop in milk production. Dr. Bach gathered data from the field to see if that's the case or not by evaluating three farms with different diets and evaluating income over feed cost. (4:33)Dr. Bach goes on to describe the farms and the methods his team used for estimating individual cow intakes in a group pen setting. Cow pen/group changes coincided with a diet change. Individual farms made their own ration decisions and pen movement decisions. (8:17)In general, cows moved from a high to a medium to a low diet over the course of lactation. Primiparous cows moved from the fresh pen to the medium diet. If diet differences were adequate between groups, the loss in milk was compensated by the lower cost ration, and producers made an additional 20-30 cents per cow per day in income over feed costs. However, if the diets were more similar, lower feed costs did not compensate for the loss in milk production. (15:30)  Dr. Weiss asks Dr. Bach if he could only build two rations, a high and a low, how would he do that? Dr. Bach's approach is to look at a histogram of milk production in the pen and split that into quantiles. His goal is to make a ration that satisfies at least 70% of the animals in the pen for the high diet and around 60% of the animals in the pen for the low diet. (24:36)Dr. Bach also ran a sensitivity analysis evaluating how results would change if milk prices or feed costs (or both) went up or down. He found that the higher the milk price, the more resilient a farm will be to a single diet and that feed cost is the opposite. The most interesting scenario is high feed costs and low milk prices - that's where it's almost mandatory to make groups, if you want to survive on a dairy. (27:23)Dr. Bach evaluated the change in nutrient intake for the diet switch and projected the milk production change from that nutrient change compared to how the cows actually performed. The cows always lost less milk production than predicted. Dr. Bach thinks the main reason is that the cows were overfed before moving. (37:46)Dr. Bach invites the audience to experiment a little bit with grouping cows. Don't be afraid of losing milk, and look beyond milk. Put in place mechanisms on the farm that allow you to measure income over feed costs as the ultimate goal. Cows are flexible, so don't be afraid of making a mistake. If something goes wrong, it will go wrong for a short period of time. You can correct it. You can change the diet right away, and the cows will recover. Abstract: It is well established that neural mass models (NMMs) can effectively simulate the mesoscopic and macroscopic dynamics of electroencephalography (EEG), including epileptic EEG. However NMMs are characterized by a high-dimensional parameter space and a lack of knowledge on what NMM parameters can be reliably estimated, thus limiting their application to clinical EEG data. In this article, we analyze the parameter sensitivity of Jansen and Rit NMM (JR NMM) in order to identify the most sensitive NMM parameters for reliable parameter estimation from EEG data. We also propose a joint estimation method for NMM states and parameters based on expectation--maximization combined with unscented Kalman smoother (UKS-EM). Global sensitivity analysis methods including Morris method and Sobol method are used to perform sensitivity analysis. Results from both the Morris and Sobol method show that the average inhibitory synaptic gain, B and the time constant of the average inhibitory post-synaptic potentials (PSPs), 1/b have significant impact on the JR NMM output along with having the least interaction with other model parameters. The UKS-EM method for estimating the parameters $B$ and b is validated using simulations under varying levels of measurement noise. Finally we apply the UKS-EM algorithm to intracranial EEG data from 16 epileptic patients. Our results, both at individual and group-level show that the parameters B and b change significantly between the pre-seizure and seizure period, and between the seizure and post-seizure period, with transition to seizure characterized by decrease in average B and high frequency activity in seizure characterized by an increase in b. These results establish sensitivity analysis guided Bayesian parameter estimation as a powerful tool for reducing the parameter space of high dimensional NMMs enabling reliable and efficient estimation of the most sensitive NMM parameters, with the potential for online and fast tracking of NMM parameters in applications such as seizure tracking and control. Abstract: Deep learning frameworks utilizing convolutional neural networks (CNNs) have frequently been used for brain age prediction and have achieved outstanding performance. Nevertheless, deep learning remains a black box as it is hard to interpret which brain parts contribute significantly to the predictions. To tackle this challenge, we first trained a lightweight, fully CNN model for brain age estimation on a large sample data set (N = 3054, age range = [8,80 years]) and tested it on an independent data set (N = 555, mean absolute error (MAE) = 4.45 years, r = 0.96). We then developed an interpretable scheme combining network occlusion sensitivity analysis (NOSA) with a fine-grained human brain atlas to uncover the learned invariance of the model. Our findings show that the dorsolateral, dorsomedial frontal cortex, anterior cingulate cortex, and thalamus had the highest contributions to age prediction across the lifespan. More interestingly, we observed that different regions showed divergent patterns in their predictions for specific age groups and that the bilateral hemispheres contributed differently to the predictions. Regions in the frontal lobe were essential predictors in both the developmental and aging stages with the thalamus remaining relatively stable and saliently correlated with other regional changes throughout the lifespan. The lateral and medial temporal brain regions gradually became involved during the aging phase. At the network level, the frontoparietal and the default mode networks show an inverted U-shape contribution from the developmental to the aging stages. The framework could identify regional contributions to the brain age prediction model, which could help increase the model interpretability when serving as an aging biomarker. Summary Problem. Pesticides play an important role in the agricultural production but their misuse affect the health of farmers and workers who manipulate such toxic substances. In the field of occupational hygiene, researches have been working in finding out the most appropriate method to estimate the human exposure in order to assess the risk and therefore to take the due decisions to improve the processes in the pesticide management and to reduce the health risk. Goals. The goal of this research was focused in creating a model for human exposure assessment specially for farming systems in developing countries by (i) evaluating the available models for human exposure assessment developed in industrialized countries, (ii) measuring the exposure in the study areas of potato and flower farming systems in Colombia, and (iii) proposing a pesticide flow model to estimate quantitatively the human exposure. Methods. The research was organized in three phases by using different methods, namely (i) evaluation of previous models of human exposure assessment (by means of a Multi-Criteria and Sensitivity Analysis); (ii) quantification of dermal exposures in Vereda La Hoya (by applying the Whole Body Dosimetry, Luminiscence Spectrometry and Tracer Method); and (iii) the development of a pesticide flow model for the human exposure assessment (by applying the Material Flow Analysis method). This model was built with dermal exposure measurements obtained in the study area of greenhouse flower crops in Sabana de Bogotá, Colombia. Results. DERM, DREAM, PHED and RISKOFDERM were selected as the most appropriate models to be applied in farming systems in developing countries as their determinants are relevant for the assessment of pesticide use and all the processes involved during the pesticide management. Afterwards these four models were applied to assess the dermal exposure in the case study of Vereda La Hoya and their determinants were compared with the characteristics of the study area, DREAM and DERM were found as the most appropriate models to assess the dermal exposure in these study areas. However, because some relevant determinants are still absent, the accuracy of these models could be improved if these are included. When comparing the final model assessment of dermal exposure in the study area, DREAM was found as the model that assesses more accurately the dermal exposure in this study area. In the study area of Vereda La Hoya, Colombia was found that the application was the activity with the highest PDE. Even though lower body parts (thighs and legs) were the most exposed, these body parts also showed the highest level of protection because of the work clothing. The ADE was high for arms and upper back due to the lack of adequate work clothing covering the complete arm and the direct contact of the upper back with the spills on the sprayer tank. Furthermore, it was found that Metamidophos is the most toxic pesticide used in Vereda La Hoya. Farmers may reduce significantly the health risk by using adequate work clothing made of appropriate fabrics that covers the whole body including the arms, cleaning properly all the pesticide residues left on the sprayer before each application, and avoiding the modification of nozzles using only nozzles with the standard discharge. The proposed pesticide flow model helps to identify the patterns of pesticide distribution on the body, the level of protection given by personal protective equipment and the estimates of potential and actual dermal and inhalation exposure. This information can be used to determine the health risk level by comparing the model estimates with the AEOL reference values for each pesticide. In addition, the model makes it possible to easily identify the activities or body parts that have high levels of exposure. This is useful in identifying improvements that will decrease the exposure during pesticide management. Because it is not feasible to measure directly the dermal exposure in all study areas, this model might help to obtain a quick estimation which could help stakeholders and authorities to make further decisions. Conclusions. This research evaluated in depth the available models for human exposure assessment, so assessors can decide which model is the most appropriate according to the characteristics of the study area in which the model is going to be applied and furthermore this research suggested improvements in the models in order to increase the estimation accuracy. This research also contributes in the proposal of a new model for human exposure based on the material flow analysis methodology studying the pesticide fractioning during the pesticide management in a certain interval of time. With this model quantitative estimations of human exposure are obtained which facilitate the risk assessment and the implementation of measures to improve the safety during the pesticide management and to decrease the risk. The proposed model also demonstrates the feasibility of applying the material flow analysis methodology in the field of human exposure, obtaining a tool that helps to understand the mechanisms of distribution of the pesticide in the farming system based on the processes involved and the flows between these processes.
