BUFFALO, NY- October 23, 2024 – A new #editorial was #published in Oncotarget's Volume 15 on October 11, 2024, entitled, “Gene regulatory network and signalling pathway rewiring: How blood cancer cells shift their shapes to evade drug treatment.” As highlighted in the paper, Acute Myeloid Leukemia (AML) is a complex and diverse disease caused by multiple mutations in genes that regulate transcription and growth. These mutations lead to extensive rewiring of the gene regulatory network (GRN), which alters the identity of hematopoietic stem and progenitor cells, ultimately blocking normal myeloid differentiation. A key feature of AML is the presence of mutations in growth factor receptor and signaling genes, such as FLT3, KIT, and RAS. Notably, FLT3 is one of the most commonly mutated genes in AML, with around 25% of cases showing an internal tandem duplication (ITD) that causes the receptor to remain constantly active. In their paper, researchers Constanze Bonifer and Peter N. Cockerill from the Institute of Cancer and Genomic Sciences at the University of Birmingham, UK, and the Murdoch Children's Research Institute, Royal Children's Hospital in Melbourne, Australia, discuss recent publications from their group addressing this issue through a multi-omics study. The authors investigated how gene regulatory networks (GRNs) in FLT3-ITD patients were rewired compared to normal cells and in response to FLT3 inhibitor treatment. The Institute on Theology and Disability (ITD) is a yearly conference bringing together academics, religious professionals, and lay people who are interested in the intersections of disability, ministry, and theology. Justin and Bekah have attended several Institutes over the years, and found them useful and enriching, but also deeply challenging. In this episode, Bekah shares her experiences from the 2024 ITD, which she found to be a true joy, and improved in many ways. She and Justin discuss the need to center disabled voices in disability theology spaces, and consider the present and future of ITD.

BUFFALO, NY- November 8, 2023 – A new editorial #paper was #published in Oncotarget's Volume 14 on March 31, 2023, entitled, “Impact of SHP2 tyrosine phosphorylation on the development of acquired resistance to allosteric SHP2 inhibitors.” The SH2 domain-containing tyrosine phosphatase 2 (SHP2) is a ubiquitously expressed non-receptor protein tyrosine phosphatase, encoded by the PTPN11 gene. It is positively regulated by upstream receptor tyrosine kinases (RTKs) to activate downstream the RAS-ERK pathway. In this new editorial, researchers Giulia Franciosa and Jesper V. Olsen from the University of Copenhagen discuss potential leukemia therapies that effectively prevent adaptive resistance. Acute myeloid leukemia (AML) is a bone marrow malignancy characterized by a blockage of differentiation and an uncontrolled proliferation of myeloid hematopoietic progenitor cells. The internal tandem duplication (ITD) in the juxtamembrane domain of the RTK FLT3 is an oncogenic driver mutation that leads to constitutive activation of its tyrosine kinase activity. Consequently, FLT3 inhibitors that block its tyrosine kinase activity represent the targeted treatment option for patients with FLT3-ITD AML, often administrated in combination with induction chemotherapy. “Nevertheless, the short duration of remission urges the development of novel combinatorial therapies for FLT3-ITD AML.” Since 2016, several potent and selective allosteric, noncovalent SHP2 inhibitors have been developed and tested in clinical trials for solid tumors. A recent study reported the effectiveness of short-term treatment with the allosteric SHP2 inhibitor SHP099 as a single agent in clinically relevant mouse models of Flt3-ITD AML. This observation was in contrast with published data showing that allosteric SHP2 inhibition is only effective as combination treatment with inhibitors of other nodes of the RAS-ERK pathway. I'm not exactly sure why baby naming is headline news here in Utah. Maybe because we have more babies to name… but whenever the list of top baby names of the year comes out… we pour over the list looking for the names of our own kids and grandkids… first to see if they made the list… and second check whether our offspring were given a name that's unique enough. It's a cutthroat business… and here's a perfect example. A family member of mine had selected a name for her first baby. There was no baby. Just a name and a gleam in her eye. But if she was ever bestowed with a baby girl… she landed on the name "Kiara with a “K.” She told everybody the good news. There's a name! For whenever a baby might show up. Itd's Kiara. You know? The next week… one of her friends got a new dog and named it Kiara. That's ice cold. But in the cutthroat game of baby naming… no harm no foul… because the dog was Kiarra with 2 “r's.” Their friendship continues. And years later when a baby did come… they named it Ava. Which is usually in the top 20… but this year… boys and girls… there was only one new name added to the top 10 lists... it's Luna. A name that laid dormant for 82 years… til Harry Potter featured a rather looney girl named “Luna Lovegood.” Which sounds like it belongs to one of those websites blocked by the state - but the name really took off when model Chrissy Teigan named her baby Luna in 2016and now the name's in the top 10. This week Mike is doing another spoiler-free book review from the High Republic era, continuing with the second wave (of the first phase) with Race To Crashpoint Tower by Daniel José Older – the junior novel which runs parallel to the “adult” novel, The Rising Storm, with a bunch of new and familiar characters! In more detail, Mike start by giving information on the other releases for The High Republic (the first two waves within Phase 1), plus some background/plot information on the book and then delves into his spoiler-free review; providing information from the blurb & the themes, he then delves into the central characters in this book (as well as some who have appeared previously). Mike proceeds to talk about the way RTCT connects to The Rising Storm (with some mild TRS spoilers) and give some plot details from the first chapter or two of this book. Nearer the end Mike then explains RTCT's plot (in brief) before giving his final thoughts and confirming what's next for SWCIC! Race To Crashpoint Tower was released June 29th 2021 and is by Daniel José Older, who has also written the Han & Lando book Last Shot and all of IDW Publishing's High Republic Adventures comics plus the Trail Of Shadows mini-series. Race To Crashpoint Tower is the junior novel of the second wave of the High Republic's first phase along with the “adult” novel The Rising Storm and the YA book Out Of The Shadows, all set in 231BBY. This week Mike is doing another spoiler-free book review from the High Republic era, now venturing into the second wave, starting with The Rising Storm by Cavan Scott – the “main adult” novel with a huge amount of characters, major plot points and shocking moments! In more detail, for the first half of the episode, Mike gives a bit of information on the other releases for The High Republic (the first two waves within Phase 1), gives some background/plot information on the book and then delves into his spoiler-free review; providing information from the blurb, the themes, how it continues from Light Of The Jedi, what he enjoyed about it, which characters have returned and details found within the first few pages. From around the halfway mark, Mike reads out the blurb & crawl before providing more details on the characters and their general journeys/thoughts, the state of the Republic at the start of this book, the wider themes and how they impact story elements, as well as how it fits in with other High Republic material, which all falls into light-spoiler territory. He then talks about “connected content”, primarily the tie-in High Republic comics and gives a mini-review of the Tempest Runner audio drama before giving plenty of warning and delving into his more detailed review, which still doesn't fully spoil the book but it does mention some plot details right at the very end, so you can listen to as little or as much info about The Rising Storm before picking it up yourself. The Rising Storm was released June 29th 2021 by Cavan Scott. He has also written the Dooku Jedi Lost audio drama, The Tempest Runner audio drama (set just after this book), as well as one story from each A Certain Point Of View book, all of Marvel's standard The High Republic comics plus several titles under Star Wars Adventures for IDW publishing including the Monster Of Temple Peak mini-series, all 3 sets of Vader's Castle stories and Adventures In Wild Space.
