For this week's Talkhouse Podcast we've got a special two-part episode featuring both members of one of my favorite bands, Wye Oak, in conversation with some other fantastic musicians, so be sure to stick around for both halves! First up it's Wye Oak's Jenn Wasner and singer-songwriter Johanna Samuels, both of whom have new music coming out, coincidentally, on June 23. Samuels has been making beautiful, deeply personal music for the last decade or so, and her new one is a doozy. The album is called Bystander, and it was recorded with Josh Kaufman of Bonny Light Horseman—he's the Josh you'll hear referenced in this chat. Check out “Ugly on the Inside” from Bystander right here. Samuels is joined here by Talkhouse pal Jenn Wasner, who's probably best known as half of Wye Oak, but has also recorded music recently under the name Flock of Dimes. It's no wonder that Wasner and Samuels get along, as both are incredibly thoughtful and sincere in their musical pursuits: They're looking for something much bigger than a hooky song to sing, they're out to connect on a deeper level. Wye Oak has sort of morphed into a different kind of band in recent years, choosing to stay away from the make-an-album-then-tour-forever cycle and instead release singles and one-off songs as Wasner and Andy Stack are able. A bunch of those songs have just been compiled on an album called Every Day Like the Last. They sound as good as anything the duo has done, which is to say they sound great. Check out “I Learned it From You” right here. In this conversation, Wasner and Samuels talk about the magic of the Newport Folk Festival, how great it is to work with Josh Kaufman, and how to express heavy thoughts within joyful music. You'll find out whether these are women of mystery, and you'll hear Wasner refer to herself as “a big golden retriever of a person.” Enjoy. In the second half of this epic Talkhouse two-parter, we've actually got three speakers: Andy Stack, who's best known as half of Wye Oak, but who's also made some solo records under the name Joyero. Along with him is Jay Hammond, with whom Stack recently made a great instrumental record, and Joe Westerlund. I'll start with Westerlund, and I'll try to do his bio justice: He started out in the Wisconsin band Deyarmond Edison, which has a massive retrospective box set coming out this year, and which is best known as the band that started Justin Vernon of Bon Iver down his musical path. Westerlund has also contributed to lots of other song-based music over the years, drumming for the likes of Califone and his project Megafaun. But it definitely feels like Westerlund has found his true calling in his solo instrumental work, including the recent Elegies for the Drift, whose compositions were largely inspired by important people in his life who'd passed away recently. It's a gorgeous, emotional ride. Check out “The Circle,” which is dedicated to Akron/Family's Miles Cooper Seaton, a friend of Westerlund's who tragically died in a car crash in 2021. It makes sense that Westerlund got together for a chat with Andy Stack and Jay Hammond, because they all have North Carolina in common: It's become something of a refuge for a bunch of bands over the past few years, with their friends in Sylvan Esso building a studio there, and lots of folks—including both members of Wye Oak—taking up residence there. Stack and Hammond got together recently and struck gold with a series of electro-acoustic improvisations that they winnowed down to album length and titled Inter Personal—a nod to the connection that they deepened by making this music together. Abstract: Introduction: In the progressive phase of multiple sclerosis (MS), the hampered differentiation capacity of oligodendrocyte precursor cells (OPCs) eventually results in remyelination failure. We have previously shown that DNA methylation of Id2/Id4 is highly involved in OPC differentiation and remyelination. In this study, we took an unbiased approach by determining genome-wide DNA methylation patterns within chronically demyelinated MS lesions and investigated how certain epigenetic signatures relate to OPC differentiation capacity. Methods: We compared genome-wide DNA methylation and transcriptional profiles between chronically demyelinated MS lesions and matched normal-appearing white matter (NAWM), making use of post-mortem brain tissue (n=9/group). DNA methylation differences that inversely correlated with mRNA expression of their corresponding genes were validated for their cell-type specificity in laser-captured OPCs using pyrosequencing. The CRISPR-dCas9-DNMT3a/TET1 system was used to epigenetically edit human-iPSC-derived oligodendrocytes to assess the effect on cellular differentiation. Results: Our data show hypermethylation of CpGs within genes that cluster in gene ontologies related to myelination and axon ensheathment. Cell type-specific validation indicates a region-dependent hypermethylation of MBP, encoding for myelin basic protein, in OPCs obtained from white matter lesions compared to NAWM-derived OPCs. By altering the DNA methylation state of specific CpGs within the promotor region of MBP, using epigenetic editing, we show that cellular differentiation can be bidirectionally manipulated using the CRISPR-dCas9-DNMT3a/TET1 system in vitro. Conclusion: Our data indicate that OPCs within chronically demyelinated MS lesions acquire an inhibitory phenotype, which translates into hypermethylation of crucial myelination related genes. Altering the epigenetic status of MBP can restore the differentiation capacity of OPCs and possibly boost (re)myelination. Copy rights belong to original authors. The result is a communal set — filmed at Sylvan Esso's new studio in Durham, N.C., called Betty's — that doesn't merely re-create three of the songs from this year's Head of Roses. These acoustic performances actually shed new light, thanks to radiant and radically different arrangements, while fully capturing the warmth we look for from Tiny Desk concerts. Clemens Wasner ist CEO und Gründer des AI-Unternehmens enliteAI, Mitinitiator des Vereins „AI Austria“ und einer der führenden Köpfe im DACH-Raum im Bereich der angewandten künstlichen Intelligenz. Im Gespräch mit Sinan Tankaz, Head of Artificial Intelligence bei der Kapsch BusinessCom, grenzt Wasner die künstliche Intelligenz von der menschlichen ab: „KI-Systeme sind wie zehntausende Praktikanten, die für einen arbeiten. Sie können aber keine Unternehmensstrategie erstellen oder Picassos malen“. Tankaz ergänzt: „Und diese Praktikanten sind zudem extrem fleissig, diszipliniert und machen kaum Fehler. Aber sie erledigten Tasks, keine Jobs.“ Beide heben die gute Ausgangssituation von Österreich im Bereich der AI hervor. Österreich boxe hier über seiner Gewichtsklasse, analysiert Wasner. Und Tankaz ergänzt: „Im KI-Bereich ist die Open Source Community sehr vital, global und offen. Dadurch besteht in der Verfügbarkeit zwischen Shenzhen, dem Silicon Valley und Standorten in Österreich kein Gap.“ AI-Experte Wasner geht davon aus, dass vor allem die G20-Staaten, die in Weiterbildung investieren und Fördermittel zum Einsatz von KI in etablierten Unternehmen bereitstellen, von der Entwicklung im AI-Bereich profitieren werden. Dabei ortet er einen Unterschied zwischen Europa und dem angelsächsischen Raum: „In der DACH-Region wird KI vor allem zur Erreichung der jährlichen Effizienzziele eingesetzt, während im angelsächsischen Raum eher neue Business-Modelle oder Gewinne im Vordergrund stehen.“ Tankaz bewertet dies durchaus positiv, denn eine mit KI ausgestattete, überlebensfähige Industrie bedeute eine gute Ausgangslage für die europäische Wirtschaft. Im Ausblick sieht Wasner in den nächsten fünf Jahren einen Wandel von der KI-unterstützten Analyse zur KI-unterstützten Erzeugung von Inhalten. Mackenzie Wasner is an up-and-coming country music artist who is turning heads with powerhouse vocals that are both fresh and somehow classic. A "homegrown" singer/songwriter, Mackenzie is from historic Leiper's Fork, Tennessee, just south of Nashville. She was born and raised in the world of country music. At just 14, Mackenzie sang alongside Vince Gill at The Mother Church of Country Music, the Ryman Auditorium. At age 17, the judges on Season 12 of American Idol lauded her "star power" and "gorgeous" voice as they unanimously sent her to Hollywood. A seasoned live performer, Mackenzie has opened for Amy Grant, Michael McDonald, Jodee Messina, and Vince Gill, among others. She is currently crafting new songs, playing shows in the southeast, and pursuing her dream to become a country music recording artist. She released her second single this year titled “Love Brings You Home."

Flock of Dimes - "Price of Blue" from the 2021 album Head of Roses on Sub Pop. Wye Oak's Jenn Wasner returned to her solo project Flock Of Dimes for an album about heartbreak, titled Head of Roses. In a press release, she explains, "Part of the journey for me has been learning to take responsibility for the parts of things that are mine, even when I’m in a lot of pain through some behavior or action of someone else. If I’m expecting to be forgiven for the things I’ve done and the choices I’ve made and the mistakes that I’ve made, it would be incredibly cowardly and hypocritical to not also do the work that’s required to forgive others the pain they caused me.” On today's Song of the Day, Wasner examines one of the biggest ways a relationship dissolves: lack of communication. “This song is about trying, and failing, to connect," she continues. “It’s about the ways in which, despite our best efforts, we misunderstand each other, and become so attached to stories that we’re unable to see the truth that’s right in front of us. And it’s about the invisible mark that another person can leave on your body, heart and mind long after their absence. She had planned on spending a fair amount of 2020 on the road playing guitar, keys, and singing with Bon Iver, but instead she’s spending it in a manner probably familiar to readers: watching TV and drinking coffee, thinking about the potential end of the world. But that doesn’t mean she hasn’t kept busy: this week, her duo with Andy Stack, Wye Oak, releases its new EP No Horizon, a collaboration with the Brooklyn Youth Chorus. And she’s got another EP out too, the recently released Like So Much Desirefrom her solo project, Flock of Dimes. Both projects are great showcases for her progressive songcraft, which pairs oblique and exploratory lyrics with swooning avant-pop. Wasner has never settled comfortable into just one mode—scanning through her discography reveals folk, synth-driven art rock, and guitar epics—but her inquisitive, intricate lyrics serve as a throughline.  She joined us to discuss the role of imagination in creating the future, staying sane, what’s keeping her company on the turntable, working with Justin Vernon’s Bon Iver collective and what she’s learned from producing singer/songwriter Madeline Kenney.

This week’s podcast brings you a variety of Detroit poets reading at Poets and Pies, a regular event hosted by the Detroit Writers’ Guild. This set of readings was recorded in August at the St. Clair Shores library. Joyce Jenkins is a Detroit native who has spent much of her life in Berkeley, California. Joyce has long been a noted Bay Area poet and the editor of “Poetry Flash,” as well as the producer and sponsor of the Northern California Book Reviewers and the Northern California Book Awards. Caroline Maun teaches creative writing and American literature at Wayne State University, and she is the Chair of the English Department. She is also the editor of The Collected Poetry of Evelyn Scott, and author of Mosaic of Fire: The Work of Lola Ridge, Evelyn Scott, Charlotte Wilder, and Kay Boyle. Her poetry publications include the volumes The Sleeping, and What Remains.Pat Barnes is a longtime Metro Detroit poet who is a favorite with poetry audiences. Her work is witty, funny and always leaving audiences with a good vibe and food for thought.Alinda Dickinson Wasner the author of 4 collections of poetry, the most recent being, When You Don't Know Who You Are published by Crisis Chronicles Press. Winner of several U.S. poetry prizes and an international prize, her work can be found in numerous print and online journals. Called "one of Detroit's best poets" by poet laureate, M.L.
