Fish on treadmills? Lisa Truong, PhD, MBA, Oregon State University, discusses the unique ways that they test exposure effects using zebrafish. Co-hosts Anne Chappelle, PhD, and David Faulkner, PhD, also ask her about the benefits of having an MBA as a scientist.About the GuestThe overall goal of Lisa Truong's research program is to utilize the zebrafish model to help build computational predictive toxicity models. Secondarily, she aims to move the field to be less reliant on animal testing and to conduct toxicity-testing based on toxicity pathways. Dr. Truong has a Bachelor's degree in pre-pharmacy with a minor in chemistry, which provided the foundation to evaluate a structurally diverse class of fluorinated compounds during her Master's studies. Her PhD thesis was focused on developing rapid in vivo assays to investigate structure response relationships using larval and adult zebrafish. Using the methods that Dr. Truong developed, the zebrafish developmental toxicity screen is now fully automated at the Sinnhuber Aquatic Research Laboratory (SARL). During Dr. Truong's postdoctoral training at the US Environmental Protection Agency, she developed systemic toxicity models while working within the then-National Center for Computational Toxicology and L'Oreal. She built numerous models using the high-throughput data generated from over 800 assays and became adept at programming in R and in advanced statistics. More recently, since her recruitment as the Deputy Director of the SARL, Dr. Truong has begun to build robust statistical methods to integrate multi-dimensional phenotypic and expression zebrafish data and has helped to streamline data collection which has further increased the throughput which is critical for this proposal. In summary, for nearly a decade, Dr. Truong has developed advanced zebrafish as a premier model for environmental health sciences research. The research is now at a point to begin mining the data to develop models to prioritize and predict toxicity of chemicals/nanoparticles/mixtures that have insufficient hazard information.Send SOT thoughts on the episodes, ideas for future topics, and more.

Ah, the joys of a post-hunt podcast. The feet were sore, and the schnoz was rosey after spending a few days tromping around in the Montana woods. Much knowledge was dropped in this episode by Dr. Kelsey Martin, who grew up on a nature preserve and cattle ranch that her family manages along the windswept plains of Eastern Colorado. She spent her youth working as a ranch hand and reveling in life on the prairie, a lifestyle that eventually led to her career as a large animal veterinarian. She applies the conservation values that were inspired in her at an early age by her father's work to her professional life in medicine and wildlife conservation. Her work has taken her to many places, including Peru, Australia, the Caribbean, and now Montana. She is currently working as a wildlife disease researcher and bison veterinarian. In her off-hours, she can be found enjoying the mountains around her home with her partner Kevin and their two dogs or working in her “studio” (aka garage) as a stained glass artist (@sagehornglass).Kevin Grunewald (you know the guy; he's a fan favorite) is a Montana-based conservationist, hunter, and angler. His winding career path in conservation biology has seen him teaching students how to trap bats in the outback of Australia, guiding clients to experience bear country in Alaska, dissecting Zebrafish embryos in a university laboratory in Wisconsin, and, most recently, conducting and sharing the benefits of sound, ecological land stewardship practices on nature preserves in California and Colorado. In his current role as a Land Steward with The Nature Conservancy, Kevin works and lives at a remote field station and nature preserve called Phantom Canyon. He spends his days working in, exploring, and sharing the wonder of the natural landscapes that he aspires to conserve. Kevin Grunewald is a Montana-based conservationist, hunter, and angler. His winding career path in conservation biology has seen him teaching students how to trap bats in the outback of Australia, guiding clients to experience bear country in Alaska, dissecting Zebrafish embryos in a university laboratory in Wisconsin, and, most recently, conducting and sharing the benefits of sound, ecological land stewardship practices on nature preserves in California and Colorado. In his former role as a land steward with The Nature Conservancy, Kevin worked and lived at a remote field station and nature preserve called Phantom Canyon. In episode 7, we are joined by Ernesto Gagarin, a Research Assistant in a zebrafish lab at Columbia University. He will be joining the University of Chicago in the fall to start his PhD! With another fish fan on the podcast this week, we wasted no time to start nerding out about fish keeping and how it has shaped our current paths in life. Having a closet and/or a bathroom full of fish tanks is clearly a key to future success in our respective fields in science. We eventually get ourselves onto the topic at hand (no pun intended), and discuss what a day in the lab looks like for Ernesto, and why zebrafish are a great model organism for studying joint regeneration. The use of model organisms in science dates to ancient Greece and represents an important way humans have progressed our collective understanding of biology and disease. We've probably all heard of using mice, rats, or even Drosophila melanogaster (i.e., fruit flies) in the lab, but zebrafish have surged in their use as a model organism. Zebrafish are small, more cost effective to maintain, have a fast generational lifecycle, and have clear embryos that enable direct observation of their development. As is the case with any model system used across the globe, standards matter and help make results transferrable to other studies.  This is where our guests come into the picture.  For this unique episode we're joined by both Corbin Schuster and Zoltan Varga of Zebrafish International Resource Center (ZIRC) at the University of Oregon. In their roles, they help raise and maintain over 12,000 genetically unique zebrafish lines for use in studies across the globe.  We learn about the health monitoring they conduct on a regular basis to maintain their colony and support their own and partner studies using their zebrafish.  The conversation touches on: Environmental DNA (eDNA) methods they employ Assay development and method selection based on their need for sensitivity and specificity The use of qPCR and dPCR in their work Studies and monitoring of parasites and pathogens that affect their colony How zebrafish have helped shift research from forward genetic studies to reverse genetic studies As always, you'll also get to know our guests on a more personal level. We learn about how they each got into this line of work, what they love about it, and how they both value human relationships and helping their communities. Scoliosis is an abnormal curvature of the spine that can cause issues with breathing, back problems and can even lead to noticeable changes such as uneven shoulders and waist. While many cases tend to be diagnosed in adolescence, doctors still have little knowledge of what causes this condition. Now, new research involving zebrafish has shed some light on the origins of scoliosis. Beth Bearce is a post-doctoral research fellow with the University of Oregon's Grime's Lab. They join us to share what they learned and the broader implications of the research.

In Episode 67 of 17 Minutes of Science, we are joined by Dr. Gabriel Bossé. Gabriel Bossé is currently a postdoctoral fellow with Randall Peterson at the University of Utah. Prior to joining the Peterson lab, Gabriel did his graduate studies at the University of Laval in Quebec City with Martin Simard, where he studied the regulations of the micro RNA pathways with the nematode C. elegans. After completing his PhD, he was interested in developing novel behavior based assays to study neuronal function and disease modeling. He chose to join the Peterson lab with the hope of combining such assays with unbiased screening approaches such as small molecule screening. Given the ongoing crisis with opioid abuse, he set out to develop the first self administration model using the zebrafish model. This unique assay allows him to study the impact of different small molecules on opioid seeking and to investigate important biological pathways regulating this behavior.
