There's a strange contradiction in God's promises to the Jewish people. In Genesis 12, God makes a covenant with Abraham to give him the land of Israel. In Genesis 17:7, God told Abraham that His covenant with him and his descendants would be an everlasting covenant. But then in Leviticus 26, God warns that if the Jewish people break His commandments, He will kick them out of their land. And then we know from our studies in the book of Ezekiel that that's exactly what happened. In fact, in Ezekiel 16, God even tells us that the covenant was broken. This was in the 500s BC. But God had said He was making an everlasting covenant around 2000 BC. So since when does “everlasting” have an expiration date after 1500 years? How can God say that a covenant can ever be broken if God had already promised it was everlasting?And does this mean that the Jewish people actually have no right to the claim of the land of Israel today?Now, I actually believe there are NO contradictions in God's Word, and that when we think we've found one, we actually just need to rightly divide the Word of Truth to figure out how it all fits together. Chris Smaje is a social scientist by training and a small-scale farmer by occupation. For the past 19 years, he has co-worked a small farm in Somerset, in southwest England. Previously, he was a university-based social scientist, working in the Department of Sociology at the University of Surry and the Dept of Anthropology at Goldsmith's College. HIs focus was aspects of social policy, social identities and the environment. Since switching focus to the practice and politics of agro-ecology, he's written for various publications, such as The Land, Dark Mountain and Permaculture Magazine, as well as academic journals such as Agroecology and Sustainable Food systems. He blogs at Small Farm Futures and has previously been a director of the Ecological Land Co-op. His latest book, A Small Farm Future, forms the basis of this conversation - in it, he lays out Ten Crises of our times, which, put together, create the Wicked Problem of this moment in history. From there, the remaining three parts of the book explore the ways in which rural localism can offer a way for humanity to see itself through the numerous crises we currently face both in the richer and poorer countries. In the podcast, we take the book as our starting point (really, you should read it) and look less at the why, of rural localism and more at the ways it might happen and how it might work. Abstract: Microtubule regulation is highly controlled in nerve regeneration. Caltubin, a novel Lymnaea stagnalis protein, contains putative EF-hand calcium-binding motifs and promotes neuronal outgrowth in Lymnaea and mouse. Here, we generated cell-permeable caltubin proteins to investigate mechanisms underlying this effect. We observed increased neurite extension and outgrowth following injury in caltubin-treated mouse neurons compared to vehicle controls. Purified caltubin bound -tubulin between its L391-V405 amino acids and promoted microtubule assembly. Caltubin competitively inhibited binding of tubulin tyrosine ligase, which catalyzes tubulin retyrosination, and increased the ratio of detyrosinated to tyrosinated tubulin. Our crystal structure analysis confirmed that caltubin has four Ca2+-binding EF-hand motifs, like calmodulin but has distinct peptide binding domains. Our work suggests a unique Ca2+-dependent regulatory mechanism of microtubule assembly by caltubin. This may represent an essential mechanism of axonal regulation, which may optimize its activity in response to various calcium states, both physiological and following injury. The SDNEDC focuses on marketing economic development opportunities and amplifying positive news about the North County economy, helping North County employers grow by solving specific business challenges, and connecting K-16 educators and employers through events and individual meetings to strengthen workforce programs throughout North County. The San Diego North Economic Development Council recently released the results from a study on the COVID-19 impacts along the 78 Corridor. The study found that nearly 36,000 jobs were lost. These numbers were extremely high in the hospitality, restaurant and retail sectors.
