Living within all of us is a complex ecosystem of microorganisms - bacteria, fungi, viruses and even parasites. The human microbiome plays a significant role in determining the health status of every individual, and very much so in regards to our reproductive health and fertility. Inflammation is a biproduct of an imbalance between beneficial and harmful bacteria. When we begin to accumulate overgrowth of unhealthy bacteria or fungi, adverse reactions can occur ranging from mild to more serious complications. A yeast infection of bacterial imbalance may seem trivial, but it could be the difference between a successful pregnancy or lack there of. Candida Albicans, is the most common of all digestive and urogenital infections, affecting 40-60% of people. Its the focus of today's seesion because it has been shown to cause issues with sperm quality, egg fertilization, failed implantation and miscarriage.In today's day and age, our lifestyles can take a massive toll on our internal ecosystem. This can be the result of poor diet, overconsumption of alcohol, excess stress, medications and so much more. IVF meds or antibiotics, for example, can upset the balance and lead to more chronic challenges with fertilty, digestion and overall health. My point is not only to point out the problem, but to raise awareness on a lesser known topic and tell you what you can do about it. Abstract: Candida albicans has the remarkable capacity to neutralize acidic growth environments by releasing ammonia derived from the catabolism of amino acids. The molecular components and mechanisms controlling this capacity remain poorly understood. Here, we present an integrative model with the cytosolic NAD+-dependent glutamate dehydrogenase (Gdh2) as the principal component. We show that the alkalization defect of a strain lacking the SPS-sensor regulated transcription factor STP2 is due to the inability to fully derepress GDH2 and the two proline catabolic enzymes, PUT1 and PUT2. Notably, the Stp2-dependent regulation of PUT1 and PUT2 occurs independent of Put3, the proline-dependent activator. Accordingly, a stp2-/- put3-/- strain is unable to derepress the expression of these enzymes resulting in a severe alkalization defect that nearly phenocopies the abrogated alkalization of a gdh2-/- strain. In wildtype cells, alkalization is tightly dependent on mitochondrial activity and occurs as long as conditions permit respiratory growth. As alkalization proceeds, Gdh2 levels decrease and glutamate is transiently extruded from cells. Together these two processes constitute a rudimentary regulatory system enabling cells to prevent the rapid intracellular build-up of ammonia. Similar to C. albicans, Gdh2-dependent alkalization is dispensable for C. glabrata and C. auris virulence as assessed using a wholeblood infection model. Intriguingly, fungal-dependent alkalization does not influence the growth or proliferation of Lactobacillus crispatus, a potent antagonist of C. albicans that normally resides in the acidic vaginal microenvironment. Our data suggest that it is time to reconsider the idea that pH modulation driven by pathogenic fungi plays a crucial role in shaping the architecture and dynamics of (poly)microbial communities. Other factors are likely to be more critical in contributing to dysbiosis and that favor virulent growth. Copy rights belong to original authors. Wer weiß nicht, was gesunde Ernährung ausmacht? Viel Gemüse und Obst, wenig Fertigprodukte, dafür frisch kochen und Vollkorn statt Weissmehl. Ich für meinen Teil versuche gezielt durch meine Ernährung Entzündungen vorzubeugen, die Lebensmittel zu meiden, die Entzündungen sogar verursachen können. Durch schlechte Essgewohnheiten kann man sich krank machen, etwa Bluthochdruck oder Diabetes regelrecht "anfressen". Haben Sie mal Bilder von früher gesehen, wie etwa in Reportagen aus der Zeit vor dem 2. Weltkrieg? Hier sahen fast alle Menschen rank und schlank aus, diese Vielzahl an unglaublich dicken Menschen gab es damals nicht. Woran könnte das liegen? Was hat sich verändert? Sicherlich ist immer mehr Nahrung immer einfacher zu erhalten, an jeder Ecke gibt es heute Fastfood, einen Bäcker oder Kiosk. Aber auch die Qualität des Essens ist völlig anders geworden, unser Essen hat sich komplett verändert. Um die Menschenmassen schnell und billig zu versorgen wurden immer mehr raffinierte Lebensmittel verarbeitet, die erstens länger haltbar und die zweitens billiger in der Herstellung sind. Wer sich damit genauer beschäftigen mag wird feststellen, dass die Einführung von raffiniertem Mehl und raffiniertem Zucker (Industriezucker) der Wendepunkt in der Ernährung war. Diese Produkte kamen 1950 bis 1960 in großer Menge in die Fertigprodukte. Genau zu dieser Zeit konnte man zusehen, wie (zuerst die Amerikaner, später andere Nationen) explosionsartig in die Breite gingen. Dazu kommt, dass übermäßiger Zuckerkonsum Entzündungen wenigstens begünstigt. Weiterhin wurde Alkohol gesellschaftlich immer akzeptierter und bald auch überall erhältlich. Man muß aber wissen, wie Alkohol vom Körper verarbeitet wird, Alkohol wird nämlich in einen zuckerähnlichen Stoff umgewandelt, ist also auch schädlich wie Zucker (abgesehen davon, dass Alkohol dem Körper an sich schadet). 1. Alkohol Was aber besonders schlecht an Alkohol ist, ist der Fakt, dass er Entzündungen im Körper begünstigt. Die Leber bildet ein spezielles Protein bei übermäßigem Genuss (es fragt sich, ob es überhaupt ein Genuss ist, mit Kopf- und Gliederschmerzen aufzuwachen?). 2. Zucker Der Haushaltszucker ist die wirklich billigste Energiequelle, die es gibt, weshalb auch alle Fertignahrung damit angereichert wird (auch schmeckt Zucker gut). Ich für meinen Teil habe beobachtet, dass es einen Zusammenhang zwischen Candida Albicans und Zucker gibt. Der Soorpilz befällt uns Menschen in der selben Weise, wie Rost unsere Autos. Ich glaube, dass Candida von Zucker lebt, und dass Candida eine Vorstufe von Krebs sein kann. Ausserdem sind Menschen, die viel Zucker konsumieren, auch häufiger von Entzündungen betroffen. Nur zur Erinnerung: in einem halben Liter Cola ist ca. 50 Gramm Zucker, selbst in Salami und Senf findet sich zum Teil reichlich Zucker, Zucker, mehr Zucker, und noch mehr Zucker, egal was man im Supermarkt in die Hände nimmt, Zucker ist fast immer drin. My guest today is actually a returning guest, Dr. Alan Dattner. He is a board-certified dermatologist practicing holistic and integrative dermatology for nearly 40 years, helping patients who have failed conventional care. Don’t Feed the Candida !!! Part One Welcome to the podcast Fibromyalgia Real Solutions with Amanda Love. You are listening to episode 6. If you missed my previous episode on Immune health then you may want to listen to it before this one. I do believe that there is a lot of information both good and bad out there about fibromyalgia . As an educator who specializes in this area, I think it is my job to point out to you that beneficial information can be organized in a fashion that will reduce the symptoms of this malady. My name is Amanda Elise Love and I was diagnosed with fibromyalgia 9 years ago which influenced my decision to enter the field of nutrition and now I am a Registered Holistic Nutritionist who works with those who are suffering from fibromyalgia. Today we are going to explore the world of Candida and its impact on your general health. Candida is a yeast, which is a form of fungus. Yeasts are everywhere, on your skin, in your mouth, and in your gut and also in your reproductive parts especially thriving in warm dark moist places. As you probably know, yeasts are present in many of the things we love to eat such as bread, cheese, wine, pastries, and carbohydrates. Yeasts are fungi that usually live in balance within our bodies ecosystem. The Candida Albicans strain is a fungus that only becomes problematic when it starts to grow out of control competing with the good bacteria for resources. When a person has an overgrowth of candida albicans the condition is known as Candidiasis. The Candida albicans strain exists peacefully with bacteria that produce acid in the gut where the pH is kept low which helps keep fungal infections at bay. But what causes it to go haywire? What about the pH of our bodies? Ordinarily, our bodies like to exist around a normal pH of 7.4. However, certain areas tend to be more acid such as the gut and vaigna.If these areas become more alkaline then Candida will grow and begin to process more sugars. This process creates byproducts that are called mycotoxins which have the potential to change body tissues and can make you sick. These mycotoxins produced by an overgrowth of Candida disrupt cell communication and confuse the body. Many scientists believe that this interruption of cell communication is responsible for many autoimmune conditions. One by-product produced by Candida processing sugars is acetaldehyde which can be toxic. This is one of the same byproducts made when your liver breaks down alcohol. Acetaldehyde can create a deficiency in vitamin B- One which contributes to brain fog, irritability, sugar cravings and depression which is why those with Candida overgrowth can feel like they are constantly hungover. C. albicans is one of the most common fungal pathogen of humans, causing local and superficial mucosal infections in immunocompromised individuals. Given that the key structure mediating host-C. albicans interactions is the fungal cell wall, we aimed to identify features of the cell wall inducing epithelial responses and be associated with fungal pathogenesis. We demonstrate here the importance of cell wall protein glycosylation in epithelial immune activation with a predominant role for the highly branched N-glycosylation residues. Moreover, these glycan moieties induce growth arrest and apoptosis of epithelial cells. Using an in vitro model of oral candidosis we demonstrate, that apoptosis induction by C. albicans wild-type occurs in early stage of infection and strongly depends on intact cell wall protein glycosylation. These novel findings demonstrate that glycosylation of the C. albicans cell wall proteins appears essential for modulation of epithelial immunity and apoptosis induction, both of which may promote fungal pathogenesis in vivo.

Das Krankheitsbild der rezidivierenden Candidose ist sowohl diagnostisch als auch therapeutisch nach wie vor eine Herausforderung für den behandelnden Gynäkologen. Ziel der Untersuchung war mehr über die lokale Immunreaktion der Scheide herauszufinden, um somit eine effektive und gezielte Behandlung der Patientinnen zu erlangen. Wir untersuchten 184 Patientinnen die klinisch Symptome wie Erythem, Pruritus und weißlichen Fluor aufwiesen und bei mindestens 4 Episoden pro Jahr die Diagnose einer CRVVC gestellt wurde. Zum Nachweis von Candida und der Spezifizierung wurden die Methoden der kulturelle Anzüchtung sowie die der PCR verwendet. Die candidaspezifischen Immunglobuline IgA und IgG wurden mittels ELISA bestimmt. Ergebnisse: In der Gruppe der symptomatischen Patientinnen konnte nur bei 17% mittels Kultur, in 28% mittels PCR eine Pilzinfektion nachgewiesen werden. Ein negativer ELISA von CIgA und CIgG spricht gegen eine Infektion. Ein positiver CIgG-Nachweis spricht für eine Infektion, CIgG könnte somit als diagnostisches Zusatzkriterium und der Verifizierung der Beschwerden dienen. So könnte auch die Anwendung neuer therapeutischer Möglichkeiten, wie Gabe von Immunstimulanzien, bei Patientinnen mit lokaler Immunsuppression Vorteile zeigen. Das diagnostische Ziel sollte eine genaue Keim-Identifikation und Feststellung des lokalen "Immunstatus" sein. Somit ist der Arzt in der Lage ein individuelles Therapieschema zu erstellen, und so mit einer effektiven Behandlung auch Kosten einzusparen.

Der opportunistisch humanpathogene Hefepilz Candida albicans gehört bei vielen gesunden Menschen zur mikrobiellen Schleimhautflora, kann jedoch bei abwehrgeschwächten Patienten oberflächliche Infektionskrankheiten sowie lebensbedrohliche Organmykosen verursachen. Obwohl der Immunstatus des Wirtes für eine Infektion mit diesem Erreger von entscheidender Bedeutung ist, trägt auch eine Reihe von Virulenzfaktoren, insbesondere die sekretorischen Aspartatproteasen (Saps), zur Pathogenität von C. albicans bei. Die angeborene Immunität ist in der Lage, derartige Pathogene schon beim Erstkontakt zu erkennen und zu bekämpfen. Haupteffektoren dieser schnellen, angeborenen Immunantwort sind Makrophagen und neutrophile Granulozyten. Mitglieder der Toll-Proteinfamilie, sogenannte Toll-like Rezeptoren (TLRs), wurden kürzlich als Rezeptoren auf diesen Immunzellen in Säugern identifiziert. Sie erkennen unterschiedliche Erreger anhand von in der Evolution hoch konservierten Strukturen, den Pathogen-assoziierten molekularen Mustern (PAMPs), was zur Aktivierung des Transkriptionsfaktors NF-кB und zur Freisetzung von Zytokinen führt. Sowohl TLR2 als auch TLR 4 wurden kürzlich für die Erkennung von C. albicans diskutiert. Zielsetzung dieser Arbeit war es, die Interaktion von Makrophagen mit C. albicans im Hinblick auf die Aktivierung von Toll-like Rezeptoren und die nukleäre Translokation von NF-кB zu untersuchen. Neben dem lebenden C. albicans-Isolat wurden zudem drei weitere Präparationen untersucht: Mit den Antimykotika (AM) Amphotericin B, Nystatin und Itraconazol vorbehandelte Keime, durch Hitze inaktivierte Keime sowie Sap-inaktivierte Keime. Die Zellstimulationsexperimente wurden mit murinen Wildtyp-Makrophagen, TLR2- bzw. TLR4- defizienten Einzelknockoutmutanten und mit TLR2/4-Doppelknockoutmutanten durchgeführt. Die TLR-vermittelte Aktivierung von NF-кB wurde mit Gelshifts (EMSA) nachgewiesen. Mit Western Blots wurden die intrazellulären Signaltransduktionswege untersucht. Der Hitze-inaktivierte Stamm bewirkte keine Translokation von NF-кB in Wildtyp-Makrophagen. Eine Inhibition der Saps bewirkte keine Abschwächung der NF-кB Induktion, so dass im Umkehrschluss dieser bedeutende Virulenzfaktor die TLR-vermittelte NF-кB Aktivierung nicht beeinflusst. Der lebende Stamm benutzte sowohl TLR2 als auch TLR4 für die Induktion von NF-кB. Nach Vorstimulation der Makrophagen mit Interferon-γ ließ sich jedoch eine klare TLR2-Abhängigkeit – unabhängig von TLR4 – in der Aktivierung von NF-кB und in der Induktion von TNF-α zeigen. In beiden Fällen wurden die Makrophagen erst ab einer Candida-Dichte von 106 Zellen pro 100 µl PBS stimuliert. Für den AM-vorbehandelten Stamm ergab sich eine deutliche TLR2-Abhängigkeit in der Regulation von NF-кB, welche durch die Präinkubation der Makrophagen mit IFN-γ nicht beeinflusst wurde. AM-vorbehandelte Keime konnten NF-кB in den Makrophagen erst ab einer Dichte von 107 Zellen pro 100 µl PBS aktivieren. Zusammenfassend zeigen diese Ergebnisse, dass lebende und AM-vorbehandelte Keime im Gegensatz zur Hitze-inaktivierten Präparation und zu den Saps relevante PAMP-Strukturen für eine TLR-vermittelte NF-кB Hochregulation besitzen. Die Beteiligung beider Rezeptoren, TLR2 und TLR4, belegt beim lebenden Stamm das Konzept, dass immunkompetente Zellen sich mehrerer TLRs bedienen, um die Immunantwort möglichst spezifisch und fein zu regulieren. Beim AM-vorbehandelten Stamm scheint den Antimykotika Amphotericin B, Nystatin und Itraconazol eine besondere Rolle zuzukommen, da diese die Integrität der Pilzmembran stören und somit TLR2-aktivierende PAMPs aus Zellwand und/oder Zytosol freisetzen. Neben dem direkten Effekt auf die Pilzmembran kommt es somit zusätzlich zu einer indirekten, TLR2 vermittelten Stimulation der Makrophagen. Untersuchungen der Signaltransduktion (Stimulation von Wildtyp-Makrophagen mit dem AM-vorbehandelten C. albicans-Isolat) ergaben eine vorübergehende, zeitlich eng begrenzte Induktion von NF-кB, die durch den Inhibitor IкB-α reguliert wird. Gleichzeitig wurden im zeitlichen Verlauf der Stimulation auch MAP Kinasen (ERK, p38, JNK) und c-Jun, eine Subeinheit des Transkriptionsfaktors AP-1, phosphoryliert. Diese simultane Aktivierung beider Transkriptionsfaktoren weist auf eine feinregulierte Immunantwort der Makrophagen gegenüber C. albicans hin und legt zudem einen Cross-Talk zwischen NF-кB und AP-1 nahe.
