Today's guest is screenwriter Brian Herskowitz. He wrote the book called Process to Product: From Concept to Script: A Practical Guide for the Screenwriter. Like millions before him, Brian Herskowitz moved out to Los Angeles to pursue an acting career. He soon learned that there was a dearth of roles for shorter leading men and began searching for other outlets for his creative passions. In 1980 he wrote his first screenplay, an action thriller about a young man who moves to Tokyo to train for the Olympics in Judo and gets caught up in an intrigue with the Yakuza. Through that script, based on his true-life experience as an international judo competitor, he uncovered a knack and passion for writing. As a writer, Brian has completed well over a dozen feature films. His first produced feature was a low budget slasher titled DARKROOM. He currently has several projects in active development including, THE ABDUCTION, THE EMPRESS OF ELSEWHERE (co-written with Theresa Nelson based on her best selling novel), TAKE A RIDE, and FAIRIES (co-written with Pam Dawber and Andy Tobey).His TV credits include a staff writing position on the NBC sit-com BLOSSOM, multiple episodes of the syndicated series HERCULES: THE LEGENDARY JOURNEYS, and ACAPULCO HEAT, the HBO hit comedy DREAM ON, CBS's RENEGADE, THE EXILE, MURDER, SHE WROTE, the FOX Network's YOUNG HERCULES, and CBS's critically acclaimed was series TOUR OF DUTY (associate producer). Abstract: Neuroimaging is commonly used to infer human brain connectivity, but those measurements are far-removed from the molecular underpinnings at synapses. To uncover the molecular basis of human brain connectivity, we analyzed a unique cohort of 98 individuals who provided neuroimaging and genetic data contemporaneous with dendritic spine morphometric, proteomic, and gene expression data from the superior frontal and inferior temporal gyri. Through cellular contextualization of the molecular data with dendritic spine morphology, we identified hundreds of proteins related to synapses, energy metabolism, and RNA processing that explain between-individual differences in functional connectivity and structural covariation. By integrating data at the genetic, molecular, subcellular, and tissue levels, we bridged the divergent fields of molecular biology and neuroimaging to identify a molecular basis of brain connectivity. Abstract: Brain-derived neurotrophic factor (Bdnf) plays a critical role in brain development, dendritic growth, synaptic plasticity, as well as learning and memory. The rodent Bdnf gene contains nine 5' non-coding exons (I-IXa), which are spliced to a common 3' coding exon (IX). Transcription of individual Bdnf variants, which all encode the same BDNF protein, is initiated at unique promoters upstream of each non-coding exon, enabling precise spatiotemporal and activity-dependent regulation of Bdnf expression. Although prior evidence suggests that Bdnf transcripts containing exon I (Bdnf I) or exon IV (Bdnf IV) are uniquely regulated by neuronal activity, the functional significance of different Bdnf transcript variants remains unclear. To investigate functional roles of activity-dependent Bdnf I and IV transcripts, we used a CRISPR activation (CRISPRa) system in which catalytically-dead Cas9 (dCas9) fused to a transcriptional activator (VPR) is targeted to individual Bdnf promoters with single guide RNAs (sgRNAs), resulting in transcript-specific Bdnf upregulation. Bdnf I upregulation is associated with gene expression changes linked to dendritic growth, while Bdnf IV upregulation is associated with genes that regulate protein catabolism. Upregulation of Bdnf I, but not Bdnf IV, increased mushroom spine density, volume, length, and head diameter, and also produced more complex dendritic arbors in cultured rat hippocampal neurons. In contrast, upregulation of Bdnf IV, but not Bdnf I, in the rat hippocampus attenuated contextual fear expression. Our data suggest that while Bdnf I and IV are both activity-dependent, BDNF produced from these promoters may serve unique cellular, synaptic, and behavioral functions. As an English major, Orin Herskowitz was an unlikely candidate for the science heavy world of technology transfer. However, after overcoming the initial imposter syndrome he felt when he became part of the technology transfer team at Columbia University, he realized that the field requires people with a wide range of skill sets, and he quickly found his feet. As Orin explains today, universities have four main purposes; training the next generation of world leaders, pushing boundaries of knowledge through basic research, engaging with local communities, and driving change in the outside world. Columbia Technology Ventures (CTV) is able to create a very meaningful impact outside its walls through the approximately 200 patents, 120 commercial license agreements, and 15 to 30 new companies that it brings to fruition every year. In addition to these incredible metrics, they also have a Lab to Market Accelerator Network, programs with a strong focus on diversity and inclusion, a podcast hosted by Orin, and close relationships with numerous external organizations.
