Abstract: Background: Parkinson's disease is a progressive neurodegenerative disorder mainly distinguished by sporadic aetiology, although a genetic component is also well established. Variants in the LRRK2 gene are associated with both familiar and sporadic disease. We have previously shown that PAK6 and 14-3-3gamma protein interact with and regulate the activity of LRRK2. Objectives: The aim of this study is to quantify PAK6 and 14-3-3gamma in plasma as a reliable biomarker strategy for the diagnosis of both sporadic and LRRK2-linked Parkinson's disease. Methods: After an initial quantification of PAK6 and 14-3-3gamma expression by means of Western blot in post-mortem human brains, we verified the presence of the two proteins in plasma by using quantitative ELISA tests. We analysed samples obtained from 39 healthy subjects, 40 patients with sporadic Parkinson's disease, 50 LRRK2-G2019S non-manifesting carriers and 31 patients with LRRK2-G2019S Parkinson's disease. Results: The amount of PAK6 and 14-3-3gamma is significantly different in patients with Parkinson's disease compared to healthy subjects. Moreover, the amount of PAK6 also varies with the presence of the G2019S mutation in the LRRK2 gene. Although the generalized linear models show a low association between the presence of PD and PAK6, the kinase can be added in a broader panel of biomarkers for the diagnosis of Parkinson's disease. Conclusions: Changes of PAK6 and 14-3-3gamma amount in plasma represent a shared readout for patients affected by sporadic and LRRK2-linked Parkinson's disease. Overall, they can contribute to the establishment of an extended panel of biomarkers for the diagnosis of Parkinson's disease. Copy rights belong to original authors. Recording engineer, mixer and producer, Joe Barresi explains how he got the nickname 'evil' while working on a Judas Priest record, and how he went from studying classical guitar and music theory to working with bands including Tool, Queens of the Stone Age, and Slipknot. Barresi reflects on producing Queens of the Stone Age’s debut album (who at the time did not have a record deal), engineering and mixing Tool’s 10,000 Days, and more recently their 2019 album Fear Inoculum, and why Celestion kit plays a huge part in all of his guitar tracking.

Abstract: Basal ganglia (BG) inhibit movement through two independent pathways, the indirect- and the hyperdirect-pathways. The globus pallidus (GP) has always been viewed as a simple relay within these two pathways, but its importance has changed drastically with the discovery of two functionally-distinct cell types, namely the prototypic and the arkypallidal neurons. Classic BG models suggest that all GP neurons receive GABAergic inputs from striato-pallidal indirect spiny projection neurons and glutamatergic inputs from subthalamic neurons. However, whether this synaptic connectivity scheme applies to both GP cell-types is currently unknown. Here, we optogenetically dissect the input organization of prototypic and arkypallidal neurons and further define the circuit mechanism underlying action inhibition in BG. Our results highlight that an increased activity of arkypallidal neurons is required to inhibit locomotion. Finally, this work supports the view that arkypallidal neurons are part of a novel disynaptic feedback loop that broadcast inhibitory control on movement execution. We are proud to have Lance Barresi from Permanent Records as our special guest editor in this episode and he has put together a great tracklist for us with a lot of rare hard rock and heavy psych bands. The majority of the cuts are taken from the Brown Acid compilations, a project that he runs together with Daniel Hall of RidingEasy Records, and in April the fourth trip in the series will be released. Since a young age, Lance has been interested in music, especially unusual and independent artists – “I've never been one to be satisfied with the status quo or mainstream culture. When I discovered punk, independent, and underground music in junior high school, I never turned back”. His interest in records led to the opening of his own record store in Chicago in 2006, which he started together with Liz Tooley, and in 2011 Permanent Records expanded to Los Angeles, where they live today. The need to stock records for the stores and his own interest in discovering new music has led to an never-ending hunt for records, and like many serious collectors he has also spent a lot of time tracking bands down that are of particular interest to him. Through the process of contacting the bands, he also got the idea to compile and license the music and this developed into the Brown Acid compilation series. In this episode we’re going to hear a few of the artists that Lance has been in contact with over the years. Six years ago, a dedicated group of vinyl aficionados and independent record store owners got together and launched an event called 'Record Store Day' - just about the time that corporate record stores were going bankrupt. Heavy metal juggernaut, Metallica, was one of the first bands to get on board, signing autographs and giving away t-shirts to 500 fans at a Bay Area record store for the event in 2008. By 2013, Record Store Day - celebrated annually on the third Saturday in April - has exploded into an international event celebrated at thousands of independent music retailers worldwide. Here in Los Angeles' Eagle Rock neighborhood, Permanent Records co-owners Lance Barresi and Liz Tooley run the sister store to their Chicago-based flagship used record store of the same name. Like many independent record stores, Permanent Records' Record Store Day events will feature in-store performances, meet and greets with artists, and a place where music fans can hang out and talk about their favorite bands and buy exclusive releases. Artists ranging from neighborhood garage bands to industry heavyweights like Bob Dylan and Bruce Springsteen issue special releases for the event every year.
