Nous sommes 30 décembre 1893, à Bruxelles. Ce jour-là, le Conseil d'administration de l'ULB ajourne une invitation envoyée au géographe français Elisée Reclus de venir donner cours au sein de l'université. La grande majorité du Conseil, mené par son président, le bourgmestre de Bruxelles, Charles Buls, estime que le moment est mal choisi : on pourrait soupçonner la jeune institution de sympathies pour l'anarchie, Reclus en étant un partisan. On craint de possibles débordements durant les leçons. Cette décision va entraîner, quelques jours plus tard, la démission du recteur, Hector Denis, qui avait fait pression pour lancer l'invitation, elle provoque aussi une agitation dans les rangs des étudiants ainsi qu'une une réaction du monde politique libéral et des loges maçonniques. Cet événement, ajouté à la question de justice sociale, celle du suffrage universel et du rôle de l'Etat en cette fin du XIXe siècle, va favoriser l'éclosion d'une école de pensée, que l'on appellera l'Ecole de Bruxelles. Une approche philosophique et pragmatique cultivée sur le terreau de la montée en puissance des sciences expérimentales du vivant et par l'avènement de la sociologie et des sciences sociales. Le tout engendrant une crise spirituelle et idéologique opposant les tenants du libre-arbitre aux défenseurs du dogme religieux. Quelles sont les idées promues par l'Ecole de Bruxelles ? De quelle manière se sont-elles développées ? Quelles en sont les grands porteurs ? A-t-elle rayonné au-delà de nos frontières ? Qu'en reste-t-il aujourd'hui ? In episode 222 of The Just Checking In Podcast we checked in with Eric Frydman. Eric works in law and has previously been President and Vice-President of the Jewish Law Student Association at Osgoode Hall Law School at York University, where he studied in Toronto in Canada. As part of his role there, Eric dealt with issues of antisemitism on campus and did a joint statement with the Muslim Law Student Association at the university to build bridges when the Israel-Palestine last ignited in 2021, prior to the October 7th massacre by Hamas. As part of this work, Eric also led two trips to Israel for Jewish and non-Jewish students alike for a cultural programme called iTrek. iTrek introduces tomorrow's leaders in business, law, policy and STEM to Israel, helping them experience Israel first-hand through peer-led, week-long trips. With over 22,000 graduate student participants across 103 nationalities since its launch in 2012, iTrek brings the richness and complexity of Israel to life through trips that combine education, culture, and fun. After we interviewed British-Palestinian journalist Hamza Ali Shah about the Israel-Palestine conflict from the Palestinian side, we wanted to cover the conflict from the Israeli or Jewish side as well. This desire was strengthened after the horrific events of October 7th 2023 where Hamas invaded Israel by car, foot and even hang-glider, murdered 1,200 Israeli citizens and kidnapped an estimated 240 people. During a six-day ceasefire at the end of November, 105 hostages were released. However, tragically, of the 135 hostages still unaccounted for, Israel says that 19 are believed to be dead at time of recording. In this episode we discuss the events of October 7th, the rise in antisemitism that has exploded since the conflict reignited across the world and the West and the impact that has had on Jewish people in those communities. We also discuss his role as president of the Jewish law student association, iTrek and how we build bridges across what has now become one of, if not, THE most controversial and divisive political issue in the world right now. We discuss the politicisation of empathy and compassion through the lens of this debate and what the future holds for Israelis and Palestinians. For Eric's mental health, we discuss why he is ‘culturally Jewish' but not practicing and how he came to that conclusion, the mental health impact of the antisemitism he's faced in life and the importance of finding your purpose and path forward. Abstract: The role of proteostasis and organelle homeostasis dysfunction in human aging and Alzheimers disease (AD) remains unclear. Analyzing proteome-wide changes in human donor fibroblasts and their corresponding transdifferentiated neurons (tNeurons), we find aging and AD synergistically impair multiple proteostasis pathways, most notably lysosomal quality control (LQC). In particular, we show that ESCRT-mediated lysosomal repair defects are associated with both sporadic and PSEN1 familial AD. Aging- and AD-linked defects are detected in fibroblasts but highly exacerbated in tNeurons, leading to enhanced neuronal vulnerability, unrepaired lysosomal damage, inflammatory factor secretion and cytotoxicity. Surprisingly, tNeurons from aged and AD donors spontaneously develop amyloid-{beta} inclusions co-localizing with LQC markers, LAMP1/2-positive lysosomes and proteostasis factors; we observe similar inclusions in brain tissue from AD patients and APP-transgenic mice. Importantly, compounds enhancing lysosomal function broadly ameliorate these AD-associated pathologies. Our findings establish cell-autonomous LQC dysfunction in neurons as a central vulnerability in aging and AD pathogenesis. Copy rights belong to original authors. Abstract: Huntingtons disease (HD) results from a CAG repeat expansion in the gene for Huntington (HTT) resulting in expansion of the polyglutamine (Q) tract in the mutant protein (mHTT). Synaptic changes are early manifestations of neuronal dysfunction in HD. However, the mechanism(s) by which mHTT impacts synapse formation and function is not well defined. Herein we explored HD pathogenesis in the BACHD and the deltaN17-BACHD mouse models of HD by examining cortical synapse formation and function in primary cultures maintained for up to 35 days (DIV35). We identified synapses by immunostaining with antibodies against pre-synaptic (Synapsin 1) and a post-synaptic (PSD95) marker. Consistent with earlier studies, cortical neurons from both WT and the HD models began to form synapses at DIV14; at this age there were no genotypic differences in synapse numbers. However, from DIV21 through DIV35 BACHD neurons showed progressively smaller numbers of synapses relative to WT neurons. Remarkably, BACHD synaptic deficits were completely rescued by treating cultures with BDNF. Building on earlier studies using reagents inspired by the chaperonin TRiC, we found that addition of the recombinant apical domain of CCT1 partially rescued synapse number. Unexpectedly, unlike BACHD cultures, synapses in deltaN17-BACHD cultures showed a progressive increase in number as compared to WT neurons, thus distinguishing synaptic changes in these HD models. Using multielectrode arrays, we discovered age-related functional deficits in BACHD cortical cultures with significant differences present by DIV28. As for synapse number, BDNF treatment prevented most synaptic deficits, including mean firing rate, spikes per burst, inter-burst interval, and synchrony. The apical domain of CCT1 showed similar, albeit less potent effects. These data are evidence that deficits in HD synapse number and function can be replicated in vitro and that treatment with either BDNF or a TRiC-inspired reagent can prevent them. Our findings support the use of cellular models to further explicate HD pathogenesis and its treatments. Copy rights belong to original authors. Frydman, PhD, LCSW is the Executive Director of Shalom Task Force. A national organization with a mission to combats domestic violence and foster safe and healthy relationships. Shana has worked in the field of intimate partner violence with a specialty in the Jewish community for 20 years. Carola Frydman is a Professor of Finance at the Kellogg School of Management. Professor Frydman's research focuses on American business history. Recent research projects examine the evolution of financial markets during the early twentieth century, with special emphasis on the role of financial intermediaries for firm growth, and on financial crises. She has also studied the long-run trends in executive compensation, the market for managers, and corporate governance. Her work has been published at journals such as the American Economic Review, the Journal of Political Economy, the Journal of Financial Economics, and the Journal of Economic History, and featured in The Wall Street Journal, the New York Times, and The Economist. She is on the Editorial Board of Explorations in Economic History, the Journal of Economic History, and of the Review Studies in Corporate Finance. Prior to joining Kellogg, Professor Frydman was an Assistant Professor of Finance at the MIT Sloan School of Management (2006-2011) and an Assistant Professor of Economics at Boston University (2011-2016). She holds a Ph.D. in Economics from Harvard University, and B.A. and M.A. degrees in Economics from Universidad de San Andres, Argentina, and is a Research Associate at the National Bureau of Economic Research.
