K.-G., Hopkins, C. E., Starr, D. A. Abstract: Hereditary spastic paraplegia (HSP) is a group of degenerative neurological disorders. We identified a variant in human kinesin light chain KLC4 that is suspected to be associated with autosomal dominant HSP. How this and other variants relate to pathologies is unknown. We created a humanized C. elegans model where klc-2 was replaced with human KLC4 and assessed the extent to which hKLC4 retained function in the worm. We observed a slight decrease in motility but no nuclear migration defects in the humanized worms, suggesting that hKLC4 retains much of the function of klc-2. Five hKLC4 variants were introduced into the humanized model. The clinical variant led to early lethality with significant defects in nuclear migration when homozygous, and a weak nuclear migration defect when heterozygous, possibly correlating with the clinical finding of late onset HSP when the proband was heterozygous. Thus, we were able to establish humanized C. elegans as an animal model for HSP and use it to test the significance of five variants of uncertain significance in the human gene KLC4. This week on Parent Tell, Kaila had the privilege of talking with Marjorie Aunos, a single mother by choice who became wheelchair bound only 16 months after the birth of her son. Majorie and Kaila have an open and honest discussion about what it was like to become a paraplegic parent, need unanticipated and sometimes unwanted help from her own parents, and how frustrating it can be to live and parent in a world that is rarely adapted to anyone except those with the ability to walk. Parenting looks different for everyone but especially for those who are born or become differently abled. We must be willing to support and advocate for all parenting experiences, for an inclusive parenting community will result in a new generation of accepting children who grow up to be accepting adults. Her expertise is working with parents who have intellectual disabilities. In 2012, she sustained a Spinal Cord Injury in a car accident. Parenting is tough sometimes. But when your child is struggling with a disability or health issues, it can be very overwhelming. This is especially true when you can't get the proper diagnosis, the key to getting effective help is understanding what's going on and advocating for your kids with the right doctors and specialists. Today on the show, I'm joined by Molley Regan. I sat down to chat with Molley about her journey with her daughter Grace's diagnosis of hereditary spastic paraplegia (HSP) 54 and how Molley followed her gut to find the correct diagnosis and treatment for her daughter. We also dive into information and resources for parents who are going through a similar situation. Molley Regan is a proud wife, mom, advocate, and full-time Cyber Project Manager for a large Technology & Financial Institution. She has been married to her college sweetheart for 13 years, Tim. They reside in Alexandria, VA with their beautiful daughters Reese and Grace and their crazy, golden retriever George. Molley and her family were on an incredible high/low journey for answers for their youngest daughter and the mobility limitations and challenges she was facing. In October 2020, during a pandemic, they took a leap of faith and pursued a life-altering neurosurgical procedure (SDR) at John's Hopkins Children's Hospital. Grace overcame the intense physical rehabilitation and is still continuing to get stronger, every day. SDR involves accessing nerves within the spinal cord, determining which of the nerve fibers are causing the spasticity, and severing those nerves.Do you know about my coaching services?
