Mapping the Core Senescence Phenotype of Human Colon Fibroblasts

BUFFALO, NY- February 29, 2024 – A new #research paper was #published on the #cover of Aging (listed by MEDLINE/PubMed as "Aging (Albany NY)" and "Aging-US" by Web of Science) Volume 16, Issue 4, entitled, “Mapping the core senescence phenotype of primary human colon fibroblasts.” Advanced age is the largest risk factor for many diseases and several types of cancer, including colorectal cancer (CRC). Senescent cells are known to accumulate with age in various tissues, where they can modulate the surrounding tissue microenvironment through their senescence associated secretory phenotype (SASP). Recently, researchers showed that there is an increased number of senescent cells in the colons of CRC patients and demonstrated that senescent fibroblasts and their SASP create microniches in the colon that are conducive to CRC onset and progression. However, the composition of the SASP is heterogenous and cell-specific, and the precise senescence profile of colon fibroblasts has not been well-defined. In this new study, to generate a SASP atlas of human colon fibroblasts, researchers Namita Ganesh Hattangady, Kelly Carter, Brett Maroni-Rana, Ting Wang, Jessica Lee Ayers, Ming Yu, and William M. Grady from Fred Hutchinson Cancer Center and the University of Washington School of Medicine induced senescence in primary human colon fibroblasts using various in vitro methods and assessed the resulting transcriptome. “[...] we utilized various relevant stressors to induce senescence in primary cultures of colon fibroblasts and perform RNA sequencing (RNASeq) to define an atlas of stressor-specific senescent profiles and a core senescent profile that is commonly regulated by all senescence inducers.” Using RNA Sequencing and further validation by quantitative RT-PCR and Luminex assays, the team define and validate a ‘core senescent profile' that might play a significant role in shaping the colon microenvironment. They also performed KEGG analysis and GO analyses to identify key pathways and biological processes that are differentially regulated in colon fibroblast senescence. These studies provide insights into potential driver proteins involved in senescence-associated diseases, like CRC, which may lead to therapies to improve overall health in the elderly and to prevent CRC. “Further studies will be needed to address the limitations of our study and to translate our understanding of the SASP and disease into clinical care.” DOI - https://doi.org/10.18632/aging.205577 Corresponding authors - William M. Our guest, Kelly Carter, is a recently elected official on her school board and she's here to tell us how God has strategically fit the puzzle pieces of her life together to prepare her for this great assignment. Ready to hear the spiritual and practical steps Kelly took to become an elected official on her school board? Let's go! KELLY CARTER and her husband, Joe, have been married 23 years and have 8 children. After high school, Kelly attended Messiah University for two years as an Elementary Education Major. She transferred to Rowan University to finish her degree in Elementary and Early Childhood Education with a specialization in Math and Science. While attending high school and college, Kelly worked in daycare and as an aide in an elementary classroom.  When she graduated from college, she began working in her hometown as a Kindergarten teacher. She had two children and then moved to Pennsylvania for her husband's job.  In Pennsylvania, Kelly worked for a local church in several leadership roles within their kidMinistry. Kelly and Joe have also been involved in the foster care system for the last ten years and have adopted five of their eight children through foster care. Kelly is a part-time substitute teacher as she cares for her four year old at home and manages the many appointments, schedules and laundry.  Kelly's life experiences have uniquely prepared her to step into the role of a School Board director. The reality is that most everyone has some sort of worms . Yes, even healthy people with no symptoms. However be mindful that viruses & bacterias are common co-infection, so its imperative that we have a comprehensive process to fight pathogens! Children who don't wash their hands often or adults who eat raw fish and under cooked meat or people who drink from contaminated water resources are most at risk for contracting parasites, but it can happen to anyone. Being in poor health is basically opening the door for these invaders to establish themselves in your body, and slowly feeding off your inner resources. This can cause nutrient deficiencies; a suppressed immune system, which can further perpetuate disease; This is a vicious cycle - poor health weakens the immune system, thus attracting more pathogens and bacteria.In conventional medicine, anti-parasitic drugs, such as praziquantel and mebendazole, are the first line of defense. The problem is that these drugs can cause many uncomfortable side effects that can be almost as bad as the parasitic infection itself, including nausea, vomiting, headaches, weight loss, and insomnia. After finding that Nexvax2 did not provide, "meaningful protection from gluten exposure when compared with placebo," ImmusanT pulled the plug on their trial. Mike and Cam share their thoughts and also discuss a highly informative article written by Kelly Carter, a participant in the study. In the end, we are optimistic that more good will come out of this down the line, and we talk about why trials like these are so important to help us hopefully find a cure for celiac disease.
