She has served in numerous leadership and staff roles across Army Medicine, including Commander of MEDDAC-HAAF and Director of Tuttle Army Health Clinic and Richmond Hill Medical Home; Chief of Future Medical Operations for ARNORTH; Brigade S3 and Chief of Plans for the 44th MED BDE; Battalion XO and S3 of the 261st MMB; and Commander of the 550th ASMC. Her strategic-level experience includes serving as EA/XO to the OTSG/MEDCOM Chief of Staff and XO to the MEDCOM Director of Communications, as well as a liaison officer to the Army's Office of the Chief of Public Affairs. LTC Sinclair holds a B.S. in Psychology from Appalachian State University and a Master of Public Service and Administration from the Bush School of Government at Texas A&M University. She is a graduate of Intermediate Level Education at the Command and General Staff College. Her operational experience includes a deployment to Iraq (2008–2009) and key roles in multiple homeland and defense support missions, including the 2010 Winter Olympics in Canada, COVID-19 response, Operation Allies Refuge and Allies Welcome, and hurricane and border response efforts. Her awards include the Bronze Star, Meritorious Service Medal (4 OLC), Expert Field Medical Badge, Parachutist Badge, and the Army Surgeon General's “A” Proficiency Designator. The American Society of Military Comptrollers has been surveying the DoD financial management workforce for their views about the planning, programming, budgeting and execution process as part of a task force on PPBE reform. Among other things, ASMC found 71 percent of the workforce thinks PPBE keeps the department from quickly responding to its mission needs, and strong majorities say the information technology tools they use to plan hundreds of billions of dollars in spending each year are woefully inadequate.On this episode of On DoD, we get an update from ASMC on the broader PPBE reform conversation -- including some new insights from the practitioners ASMC surveyed. No one particularly likes the way the Defense Department andCongress handle the DoD budgeting process. It's rigid, inflexible, and veryslow. But the PPBE process, as it's called, has been encoded into the DNA ofthe Defense bureaucracy for more than 60 years, and turning the ship around isgoing to be very tough. That's why there are at least two panels of experts who'llbe working on the problem over the next year or more: one chartered by Congress, andone organized by the American Society of Military Comptrollers. ABOUT KETA & DANI Marquita “Keta” Price (she/her/Goddess), aka “The Hood Planner,” is a third-generation East Oakland native serving as the Director of Urban and Regional Planning for The East Oakland Collective. Keta's formal passion for urbanism came about during recreational research on how gentrification has impacted low-income Black “hoods” across the nation. As director, Keta is the lead on several East Oakland neighborhood & transportation planning projects, participates in the development of local urban and regional planning, and holds the city of Oakland accountable to equitable zoning and land use in East Oakland. Her early activism and care for community is rooted at Merritt College, where she served as the Vice President of The Black Student Union, President of the Kem(istry) Club, President of the Associated Students of Merritt College (ASMC), and served as a Student Trustee of the Peralta Community College School District. Keta's initial goals were to restore the intellect and militancy of Merritt's student government. Seeded by Chairman Bobby Seale and other former Black Panthers, ASMC focused on leveraging the institution's resources to address the socioeconomic issues from the flatlands prohibiting students from thriving academically. Keta's political and social goals pulled her away from studying chemistry to deeply exploring how societies, communities, and cities are planned, designed and constructed. As a chemistry major, Keta received an achievement award for the completion of The Center for Educational Partnerships NIH-UCB internship. During this project Keta synthesized pro-fluorophores used for live-cell RNA imaging. Keta also received recognition and commendation from the California Legislature Assembly and State of California Senate and the Alameda County Board of Supervisors for her student advocacy work with the Tobacco-less Club at Merritt College. Keta has an Associates of Science in Mathematics and Natural Science. When the opportunity presents itself, Keta plans to further her education in Urban Studies, Data Analysis and Design. Danielle "Dani" Dynes is a dedicated community planner who was born in East Oakland and raised in West Oakland. As a planner, she focuses on the present needs of residents and while creating strategies to deal with the challenges of the future. She works to bring more resources and infrastructure to Oakland and ensure we have safer, healthier, and well-connected communities moving forward. Danielle has a B.A. in Urban Studies and Planning from San Francisco State University. She has previously worked at the Oakland Department of Transportation (OakDOT) in their Planning and Project Development section. Several of Danielle's projects at OakDOT prioritized equitable and culturally relevant change in the East Oakland community. She facilitated events around the 90th Avenue Scraper Bike Way and conducted community outreach for the East Oakland Mobility Action Plan. She also helped manage Sustainable Transportation Planning grants while interning at the California Department of Transportation. Before she studied planning Danielle taught website design and visual communication to students in Oakland and Richmond. In 2016 she was honored as a Youth Development Fellow by Coro, a civic leadership training organization. In her leisure, Danielle enjoys photography, crafting, hiking, and gardening. We often discuss the impact that procurement talent has on current results and the role it will play in determining how our function evolves in the future. I’m joined today by Louis Bastone, Indirect Category Manager for ASMC, a manufacturer of chip-making machines in the semiconductor industry. 9 out of 10 times when you ask someone how they ended up in procurement, their answer involves some an unexpected career twist. Not Louis. He took procurement courses in college while pursuing a degree in Integrated Supply Chain Management. In this interview, Louis shares the role that creativity, enthusiasm and influence have on performance and job satisfaction as well as our elevation from tactical to strategic work. Background: Asthma is characterized by airway remodeling, altered mucus production and airway smooth muscle cell (ASMC) contraction causing extensive airway narrowing. In particular, alterations of ASMC contractility seem to be of crucial importance. The elevation of the cytoplasmic Ca(2+) concentration is a key event leading to ASMC contraction and changes in the agonist- induced Ca(2+) increase in ASMC have been reported in asthma. Objective: The aim of this study was to investigate mechanisms underlying these changes. Methods: Murine tracheal smooth muscle cells (MTSMC) from T- bet KO mice and human bronchial smooth muscle cells (HBSMC) incubated with IL-13 and IL-4 served as asthma models. Acetylcholine- induced changes in the cytoplasmic Ca(2+) concentration were recorded using fluorescence microscopy and the expression of Ca(2+) homeostasis regulating proteins was investigated with Western blot analysis. Results: Acetylcholine- induced Ca(2+) transients were elevated in both asthma models. This correlated with an increased Ca(2+) content of the sarcoplasmic reticulum (SR). In MTSMC from T-bet KO mice, the expression of the SR Ca(2+) buffers calreticulin and calsequestrin was higher compared to wild- type mice. In HBSMC incubated with IL-13 or IL-4, the expression of ryanodine receptors, inositol-3-phosphate receptors and sarcoplasmic/ endoplasmic reticulum Ca 2+ ATPases 2 was increased compared to HBSMC without incubation with interleukins. The enlarged acetylcholine- induced Ca(2+) transients could be reversed by blocking inositol-3- phosphate receptors. Conclusions: We conclude that in the murine asthma model the SR Ca(2+) buffer capacity is increased, while in the human asthma model the expression of SR Ca(2+) channels is altered. The investigation of the Ca(2+) homeostasis of ASMC has the potential to provide new therapeutical options in asthma. Copyright (C) 2008 S. Karger AG, Basel.
Background: The interleukins IL-4 and IL-13 play a key role in the pathophysiology of asthma. The interleukin receptor IL-13R alpha 2 is believed to act as a decoy receptor, but until now, the functional significance of IL-13R alpha 2 remains vague. Methods: Bronchial reactivity was quantified in murine lung slices by digital video microscopy and acetylcholine (ACH)-induced Ca2+ signaling was measured in human airway smooth muscle cells (ASMC) using fluorescence microscopy. Results: IL-4 or IL-13 up to 50 ng/ml induced bronchial hyperreactivity. But after incubation with 100 ng/ml this effect was lost and bronchial responsiveness was again comparable to the control level. The effects of IL-4 and IL-13 on bronchial reactivity were paralleled by the effects on ASMC proliferation. Fifty nanograms per milliliter of IL-4 and IL-13 increased the Ca2+ response of human ASMC to ACH. At 100 ng/ml, however, the effects of the cytokines on the Ca2+ response were no longer evident. The expression of IL-13R alpha 2 increased with increasing concentrations of IL-4 or IL-13, reaching its maximum at 100 ng/ml. Blocking IL-13R alpha 2, the loss of the effect of IL-4 and IL-13 at 100 ng/ml on human ASMC proliferation and the ACH-induced Ca2+ response were no longer present. Conclusions: IL-4 and IL-13 induce bronchial hyperreactivity by changing the Ca2+ homeostasis of ASMC. These effects are counteracted by IL-13R alpha 2. The biological significance of IL-13R alpha 2 might be a protective function by regulating IL-13- and IL-4-mediated signal transduction and thereby limiting pathological alterations in Th2-mediated inflammatory diseases. Background: Airway smooth muscle cells (ASMC) play a key role in bronchial hyperresponsiveness (BHR). A major component of the signaling cascade leading to ASMC contraction is calcium. So far, agonist-induced Ca2+-signaling in asthma has been studied by comparing innate properties of inbred rat or mouse strains, or by using selected mediators known to be involved in asthma. T-bet knock-out (KO) mice show key features of allergic asthma such as a shift towards T(H)2-lymphocytes and display a broad spectrum of asthma-like histological and functional characteristics. In this study, we aimed at investigating whether Ca2+-homeostasis of ASMC is altered in T-bet KO-mice as an experimental model of asthma. Methods: Lung slices of 100 to 200 mu m thickness were obtained from T-bet KO- and wild-type mice. Airway contraction in response to acetylcholine (ACH) was measured by video-microscopy and Ca2+-signaling in single ASMC of lung slices was assessed using two-photon-microscopy. Results: Airways from T-bet KO-mice showed increased baseline airway tone (BAT) and BHR compared to wild-type mice. This could be mimicked by incubation of lung slices from wild-type mice with IL-13. The increased BAT was correlated with an increased incidence of spontaneous changes in intracellular Ca2+-concentrations, whereas BHR correlated with higher ACH-induced Ca2+-transients and an increased proportion of ASMC showing Ca2+-oscillations. Emptying intracellular Ca2+-stores using caffeine or cyclopiazonic acid induced higher Ca2+-elevations in ASMC from T-bet KO- compared to wild-type mice. Conclusion: Altered Ca2+-homeostasis of ASMC contributes to increased BAT and BHR in lung slices from T-bet KO-mice as a murine asthma model. We propose that a higher Ca2+-content of the intracellular Ca2+-stores is involved in the pathophysiology of these changes.
