As a longtime friend of One Life Radio, Maura is with us almost every Friday! Caring For Pets In Freezing Temperatures Bernadette talks about a cat that her daughter found dead where she feeds a colony of stray cats. Bernadette's daughter took the deceased cats to the vet to be identified for gender and and chip. His name was Willie and he was only ten months old. Willie froze to death. The cat's real owners contacted Bernadette's daughter and the owners did not want the ashes. It was very sad, but rather a grim reminder to keep your animals inside. Symptoms of animals suffering from hypothermia is shivering, pale grey gums, lethargy, dilated pupils, and collapsing. If your dog or cat has any of these symptoms, take them to the vet immediately. Please keep your animals inside. Dogs sensitive to cold should be kept inside with quick potty breaks outside. The small dogs and short haired breeds benefit from a doggy jacket that helps to keep their core warm. Booties may also be a good idea as lots of heat escapes through a dogs foot pads. With cats, please keep them inside or if they or indoor outdoor allow them a shelter like the ones here. Trap-Neuter-Return is the humane and effective approach for stray and feral cats. Now in practice for decades in the US after being proven in Europe, scientific studies show that Trap-Neuter-Return improves the lives of feral cats, improves their relationships with the people who live near them, and decreases the size of colonies over time. Faraz Khan Faraz Khan is a former computer scientist turned longevity educator, speaker and biohacker. Seeing his parents age before his eyes prompted Faraz to dive headfirst into the world of anti-aging and longevity. Faraz turned from helping large media, life sciences and pharma companies to helping everyday people beat back the scourge of aging. He launched his podcast "Anti-Aging Hacks" in 2019 where he interviews the top minds in longevity, anti-aging and health. Faraz is also the host of a yearly Anti-Aging Summit. How To Reduce Scarring Faraz Khan suffered a horrible oil incident at 8 years old that made him form a large scar. Faraz discovered that over the past 15 years, all the scientific advancements, studies of older populations and a vastly improved understanding of our biology, have resulted in exciting information on how to slow down the aging process today – and turn back our biological clock by many years, and get back the energy, skin, hormones we had many years ago. Theres different kinds of scars that must all be treated differently. Keloids and hypertrophic scars are caused by cutaneous injury and irritation, including trauma, insect bite, burn, surgery, vaccination, skin piercing, acne, folliculitis, chicken pox, and herpes zoster infection. Notably, superficial injuries that do not reach the reticular dermis never cause keloidal and hypertrophic scarring. Once your wound is healed and a scar is formed, keeping it hydrated is also extremely important, especially if you are attempting to reduce the appearance of your scar. Keeping your wound hydrated will help your body produce a more flexible scar that is able to be reduced in the future. Hydration is very important for healing scars. Drink water and Alsop supplement with silicone gel sheets. It's best to address a scar before they heal completely. But, Faraz says you can heal old scars, even 20-30 years old too! Thank You! Abstract: Purpose To propose an efficient retrospective image-based method for motion correction of multi-contrast acquisitions with a low number of available images (MC-MoCo) and evaluate its use in 3D inhomogeneous Magnetization Transfer (ihMT) experiments in the human brain. Methods A framework for motion correction, including image pre-processing enhancement and rigid registration to an iteratively improved target image, was developed. The proposed method was compared to Motion Correction with FMRIB's Linear Image Registration Tool (MCFLIRT) function in FSL over 13 subjects. Native (pre-correction) and residual (post-correction) motions were evaluated by means of markers positioned at well-defined anatomical regions over each image. Results Both motion correction strategies significantly reduced inter-image misalignment, and the MC-MoCo method yielded significantly better results than MCFLIRT. Abstract: Background: A mutation in the C9orf72 gene is the most common genetic mutation of familial and sporadic ALS, as well as familial FTD. While prior studies have focused on elucidating the mechanisms of neuronal dysfunction and neurodegeneration associated with this genetic mutation, the contribution of microglia to disease pathogenesis in the ALS/FTD disease spectrum remains poorly understood. Methods: Here, we generated a new disease model consisting of cultured C9orf72 ALS/FTD patient-derived induced pluripotent stem cells differentiated into microglia (iPSC-MG). We used this model to study the intrinsic cellular and molecular phenotypes of microglia triggered by the C9orf72 gene mutation. Results: We show that C9orf72 ALS/FTD iPSC-MG have a similar transcriptional profile compared to control iPSC-MG, despite the presence of C9orf72-associated phenotypes including reduced C9orf72 protein levels and dipeptide-repeat protein translation. Interestingly, C9orf72 ALS/FTD iPSC-MG exhibit intrinsic dysfunction of phagocytic activity upon exposure to A{beta} or brain synaptoneurosomes and display a heightened inflammatory response. Detailed analysis of the endosomal and lysosomal pathways revealed altered expression of endosomal marker early endosome antigen 1 and lysosomal associated membrane protein 1 in C9orf72 ALS/FTD iPSC-MG, which was confirmed in patient postmortem tissues. Conclusions: These findings demonstrate that unstimulated C9orf72 iPSC-MG mono-cultures share a largely similar transcriptome profile with control microglia, despite the presence of C9orf72 disease phenotypes. The dysfunction of the endosomal-lysosomal pathway as demonstrated by aberrant microglia phagocytosis and engulfment of cellular debris and brain pathogens suggests that disease-related microglia phenotypes are not intrinsic but instead require microglia to be activated. In summary, the C9orf72 iPSC-MG culture system provides a novel human disease model to study the role of microglia in C9orf72 ALS/FTD disease pathogenesis. Copy rights belong to original authors. In this episode, Kribashini chats with Sarah and Richard Bryant from Bryant Alsop, a Melbourne based architecture firm who take a highly personalised approach to delivering carefully considered designs to their clients. They share about their approach to the design process and how placing the client’s needs at the core of every project is absolutely vital. One of our very own BuildHer’s, Suzie, was fortunate to have Bryant Alsop design her forever home. When we popped in to see the site the other day, we were blown away by the level of detailing and the amazing spaces they’ve created. Sarah and Richard go into more detail about the design concept for Suzie’s home and the key areas of the brief that shaped the way they approached the design. Thinking about a building as a living, breathing 3 dimensional shape, Sarah and Richard talk about how they don’t get fixated on floor plans, but rather think in terms of volume which influences their decisions regarding levels, ceiling heights and room sizes. Sarah and Richard take a really personal approach to design and talk about the importance of listening to their clients and prioritising the personalised social aspects of a brief, as well as the functional. It gives an insight into the level of thought and consideration that goes into each detail of their designs, resulting in spaces that flow in harmony with the people who call them home. We can’t agree more with the way Sarah and Richard tackle the design process. The lifestyle, values and desires of the client are always in the forefront of their minds, shaping each decision, no matter how minute the detail. They work to understand their clients and listen really well to come up with a design that they will love for years to come. At the end of the day, building is a fluid process. Things will go wrong but having the right team around you means that those problems don’t have to result in disaster.
The Joyful Mourning - A Podcast for Women Who Have Experienced Pregnancy or Infant Loss
Listen in to an honest conversation about pregnancy after loss with my dear friend Katelyn James Alsop. We talk very candidly about what it’s like to be pregnant again after losing a baby. There are moments of tears and of laughter in this episode, but isn’t that the reality of this grief journey; a dance of joy and sorrow. We talk about how Katelyn made the decision to try again for another baby after her son James died from complications from Down Syndrome. We talk about when or if there ever is the right time to announce another pregnancy, what it’s like to keep our baby’s memories alive when a new baby is born and I love when Katelyn talks to me about battling the fear that plagues so many of us in our pregnancies after loss. We even talk about parenting after loss and parenting while pregnant after loss; what it looks like to invite our living children into our grief and our fear. And in doing so, hopefully teach them about what it looks like to trust God even when it’s scary and even when the worst happens and also show them what finding healing can look like. I am so grateful for Katelyn and her story and her honesty in this episode. I pray that it reminds you that you are not alone. And that there is joy to be had, even in the scary, fearful places like pregnancy after loss. February, 1838 As night fell upon the dimly lit streets of London, Jane Alsop hears a knocking on her door. Curious, she opens the door to reveal a man draped in a large coat who claims to be a police officer. The man convinces Alsop to follow him towards the street, this is when the man attacks. Tossing off his coat the man reveals his horrific image: Fiery red eyes, large metal claws, and blue flame pouring from his nostrils, the creature begins to slash at Alsop, clawing at her neck. Luckily, one of Jane’s sisters was nearby and assisted her in fending off the assailant. The superstitions surrounding the ideas of ghosts, death, and what happened to your soul as your body approached it’s demise were all inspiring topics for the plays of William Shakespeare. It turns out that these superstitions were not only fuel for dramatic presentations on the stage, but formed the foundation of pretty creepy tales about the real life of one of England’s most famous monarchs, Queen Elizabeth I. According to tales from her handmaiden, Elizabeth was paranoid to a fault, scared of dying, and left specific orders in her will for how her body was to be handled that defied the traditions of the land, and left a set of at least 6 individuals doing work that was scarier than grave digger in Shakespeare’s Hamlet. Here this week to share with us the real life history of the three weeks after the death of Elizabeth I and the surprising, often macabre, realities of what those weeks entailed by order of the Queen is the man who wrote his PhD thesis on the Living Death of Elizabeth I, Dr. James Alsop.

Seth Horner, tubist of the North Carolina Symphony on talking to himself, studying with David Fedderly and his box of mouthpieces. Seth Horner on trusting your gut, turning down a military band gig and phrasing. From his website: A native of Eugene, Oregon, Seth Horner is the newly-appointed principal tuba of the North Carolina Symphony in Raleigh, NC. Horner was the acting-principal tuba of the Baltimore Symphony Orchestra for three seasons beginning in 2014, served as part-time instrumental faculty teaching tuba and euphonium at Towson University for three semesters, and in winter of 2017 Horner was selected as the tuba instructor at the Peabody Conservatory before relocating to North Carolina. At the age of 19, Seth served a one-year position as principal tuba of the Colorado Symphony Orchestra under Marin Alsop and has held positions as the principal tuba of the West Virginia Symphony and the Oregon Ballet Theatre Orchestra. Seth studied primarily with Richard Frazier, Mike Grose, Paul Krzywicki, Warren Deck, and David Fedderly. Horner attended the Peabody Conservatory, was an honors graduate of the University of Oregon and was valedictorian of his class at the Curtis Institute of Music. Seth had performed frequently with the Baltimore Symphony prior to assuming a full-time position in 2014 and performed during the BSO’s west coast tour and its 2010, 2014, and 2016 Carnegie Hall appearances with Marin Alsop. He can be heard on the BSO’s two most recent albums featuring the symphonies of Leonard Bernstein conducted by Marin Alsop (Naxos), as well as Ms. Alsop's recording with the Colorado Symphony of Roy Harris Symphonies 3 and 4 (Naxos). Horner has performed with the Philadelphia Orchestra, National Symphony, Kennedy Center Opera, New Jersey Symphony, Louisville Symphony, Oregon Symphony, and New World Symphony among others. He has made chamber music appearances with the Washington Symphonic Brass, the Bay Street Brassworks, and the Clipper City Brass. Horner was a featured soloist with the Capital Wind Symphony in Vienna, VA, and was a finalist at the 2010 International Tuba and Euphonium Conference. Seth Horner has served as an audition panel member and coach for Carnegie Hall’s National Youth Orchestra of the United States of America as well as Carnegie’s NYO2. In the summer, he has been on the faculty of Wyoming Seminary’s Performing Arts Institute in Kingston, PA as the instructor of tuba and euphonium as well as performing with the Oregon Bach Festival and Britt Festival orchestras. Mr. Horner has given masterclasses at the University of Oregon, Ithaca University, and Towson University. Seth resides in Durham, NC.
