When Florida native Jake Nolan was 14 years old, he started showing signs of mental illness and depression. By the age of 17, he was diagnosed with Bipolar Disorder and had tried to kill himself. Jonathan Butler is a singer-songwriter and guitarist who leads a life few can imagine. Born in South Africa under the shadow of apartheid and raised in poverty, Butler was the first non-white artist to be played on South African radio and appear on national television. Though his musical abilities would take him away from the world he grew up in, Jonathan would neither forget the plight of his fellow South Africans, nor the man that led them to freedom. It is for this reason Nelson Mandela credits Butler's music as having inspired him during his imprisonment. In more ways than one, Jonathan Butler is representative of South Africa. Jonathan's story begins at a very young age when his father presented him with a homemade one-string guitar. Little did his father realize that he was giving way to a musical phenomenon. From then on, Jonathan's musical gifts grew by leaps and bounds. In his early teens Butler won a local talent contest, affording him the opportunity to perform with a touring musical company throughout South Africa. On occasion he would perform at lavish concert halls for whites only, where he would not be allowed to use the bathroom, and the very next night he would perform in a dilapidated local township. For this episode, the second of July 2023, I spoke with Puppet Film Director and Creature Effects Artist Kevin McTurk. Kevin and I first connected on Facebook a couple of years ago, maybe it was more than that. We've been trying to find a suitable time to have him on the podcast since then. Well now is the time as Kevin has a project on Kickstarter as we speak. Kevin directs these amazing short films using puppetry and wonderfully atmospheric effects. He's a veteran of creature effects in movies like Batman Returns, Jurassic Park, Galaxy Quest and King Kong. However, he is, as I said also a director of these amazing short films. He started with The Narrative of Victor Karloch a victorian ghost story. Then there was The Mill at Caulder's End in a similar vein. The one that I first spotted was his third film “The Haunted Swordsman”. It's quite extraordinary. Abstract: Sporadic Creutzfeldt-Jakob disease (sCJD) is the most common human prion disease, which occurs when the cellular prion protein (PrPC) spontaneously misfolds into disease-associated forms, culminating in fatal neurodegeneration. In a genome-wide association study of sCJD we recently identified risk variants in and around the gene STX6, with evidence to suggest a causal increase of STX6 expression in disease-relevant brain regions. STX6 encodes syntaxin-6, a SNARE protein primarily involved in early endosome to trans-Golgi network retrograde transport. Here we investigated a causal role of Stx6 expression in mouse prion disease through a classical prion transmission study assessing the impact of homozygous and heterozygous syntaxin-6 knockout on disease incubation time and prion-related neuropathology. Homozygous (Stx6-/-) and heterozygous (Stx6+/-) knockout of Stx6 expression extended survival by 12 days following inoculation with RML prions relative to wildtype controls. Similarly, in Stx6-/- mice, disease incubation time following inoculation with ME7 prions was extended by 12 days. Histopathological analysis revealed a modest increase in astrogliosis in ME7-inoculated Stx6-/- animals and a variable effect of Stx6 expression on microglia activation, however no differences in neuronal loss, spongiform change or PrP deposition were observed at endpoint. Importantly, Stx6-/- mice are viable and fertile with no gross impairments on a range of neurological, biochemical, histological and skeletal structure tests. Our results provide confirmatory evidence for a pathological role of Stx6 expression in prion disease and support further exploration of syntaxin-6 lowering as a potential therapeutic strategy. This week I am talking to Tamara Caulder Richardson. Tamara has had six Near-Death Experiences and is a Multiple Spiritually Transformative Experiencer, Christ Channeler, Christian Minister, Mystic Seer, and an International Evidential Medium. Tamara was born in Hickory, North Carolina. She was born with the gift of “knowing.” Tamara believes that her six NDEs have enhanced her ability to see into hidden realms.
