I desired for years, to do a Kailash Chora…. Was it an Ego trip, or Challenge or spiritual desire?The Kailash Chora is the most famous Pilgrimage for the Hindus and Tibetans. For the Hindus represents Mount Kailash, Mount Meru the place where the God Shiva lives. It is not allowed to climb Mount Kailash, so the pilgrims go around Kailash, which is called Kailash Chora. The stunning altitude is on this pilgrimage trek mainly between 5200m and 5700m and is about 50 km long… Finally, I booked the Kailash Chora, and we drove with the bus from Kathmandu to Dunche, the starting point of the Kailash Chora trek. I repeated always in my mind the famous Shiva mantra and got blissful, sometimes I cried for Shiva,... Awesome experiences...Just before we arrived there was a terrible accident just in front of us… A car passed by our bus and crashed with the speed of 80km/h in another car…. Both cars were destroyed and one car flipped upside down… I just saw a hand hanging out of the car that flipped upside down … Nobody wanted to help. I shouted stop I have to help, and the guide tried to stop me… The driver of this flipped car was mooring, the roof was nearly flat to the level of the crashed front of the car…Is he still alive?How to get him out of the car….We tried many ways… nothing worked out, and we were desperate, finally, I got the idea to remove the plastic from inside the door… Then we got him out and nursed him… His shoulder and some ribs were broken, severe head injuries…. Still, he could breathe… Luckily he had survived and there was no other person inside the car...Then we continued our drive to Dunche…I took my pilgrimage seriously and meditated in the morning and night repeating the name Shiva mantra with every step that I took… The first day we trekked from the altitude of 4600m to 5200m and 20Km length and the last 5 Km was so exhausting… I had to reduce my speed, my 16kg backpack made it even worse. I enjoyed the beautiful mountain area and even more my rest! On this day, was the Tibetan Saga festival and 1000 Tibetan pilgrims were doing the trek… And that was so motivating….On the second day we stayed at the monastery to trek close to the Kailash and touch the Kailash instead of continuing with Chora….Unexpected this day became the Highlight for most Tourists and pilgrims…Just when I started I was so overwhelmed by love, that I cried … IncredibleAnd all people who touched the Kailash had a high spiritual experience, regardless if they had faith or not….A friend of mine took a stone from Kailash and brought that to our Guest house for the people who could not touch Kailash…. One Indian woman cried when he gave her the stone and so me…She decided that we couldn't bring the stone home and should give the stone back.Half of our group, could not continue the Chora because they suffered from symptoms of altitude sickness and went back and one woman rode a horse to the 5700m high pass…. This 5700m high pass got steeper and steeper, and I switched to snail gear… stopped every 5 minutes… the backpack drew me back… Still, it was not as hard as the Thorens la pass, because I trained every morning on the treadmill… Our group and guide supported me a lot, and thought I could do that… So I did… I lost 5 kg body weight because I could not get my raw food, and could not eat much-dried fruits and ground nuts…. The digestion didn't work properly at high altitudes, hard to sleep… Congestion of the nose...I had the fortune to complete the Chora. The Tibetans needed only one day to complete the Chora and we required 4 days, including touching the Kailash.I think everybody should be well-trained before starting the Chora… It will be an unforgettable experience, - a life time experience. For sure we don't get holier, but a glimpse of the Lord will be worth it.And You strengthen your perseverance and stamina … Regardless how difficult it is, you learn to do it in baby steps when there is no other choice.

Il patriarca François osserva da una delle finestre del primo piano il giardino sottostante: un ragazzino elegantemente vestito sta duellando con un adulto. Il ragazzo è suo figlio, si chiama François, come lui (lo hanno battezzato così in onore di san Francesco d'Assisi); l'altro duellante è il suo maestro di armi. Il patriarca è combattuto fra la gioia di vedere la bravura del figlio nel maneggiare le armi e la tristezza per la notizia che il giovane desidera ardentemente diventare prete.Ma lui ha ben altri progetti per il figlio. Lo manderà a Parigi, a studiare il diritto, così diventerà un ottimo magistrato. Il giovane Francesco (1567-1622) si piega alla decisione del padre e prosegue gli studi presso il collegio parigino di Clermont, tenuto dai gesuiti. Studia retorica, latino, greco, ebraico, filosofia e teologia, conoscenze che gli permettono di "imparare gli esercizi della nobiltà". Impara bene anche il francese, che comincia ad utilizzare in sostituzione del dialetto natale. Ma ciò che lo attira di più è lo studio della teologia.Francesco mostrava un forte interesse per la teologia dei santi Agostino di Ippona e Tommaso d'Aquino, concentrandosi in particolare sulla grazia e la predestinazione, che era molto discussa allora a causa del protestantesimo. Qualche tempo prima della nascita del Sales, Calvino aveva teorizzato una sua teologia della predestinazione, in modo molto diverso dalla dottrina cattolica. Questo approccio suscitò grande ansia in Francesco, che per alcune settimane - tra il dicembre 1586 e il gennaio 1587 - si immaginò di essere predestinato all'Inferno. Sconvolto, pregava davanti a una statua della Vergine Maria in una chiesa domenicana, la chiesa di Saint-Étienne-des-Grès. Con l'aiuto della preghiera arrivò alla liberazione dalle sue paure. Fece anche voto di castità, accrescendo preghiere e penitenze.STRAORDINARIE QUALITÀ SACERDOTALINel 1588, decise di continuare gli studi in Italia, arrivando in una delle più notevoli università europee del tempo: Padova. Cercando consiglio e aiuto, si pose sotto la direzione spirituale del gesuita Antonio Possevino, che gli fece fare gli esercizi spirituali. Come confidò una volta a un amico: "Ho studiato diritto per piacere a mio padre e teologia per piacere a me stesso". A 25 anni, viaggiò tra Loreto, Roma, Venezia e poi tornò in Savoia. Qui il padre gli offrì la signoria di Villaroger e gli presentò una fidanzata. Ma Francesco ribadì il suo desiderio di diventare prete.Qualche tempo dopo, il vescovo Claude de Granier gli offrì l'incarico di prevosto del capitolo di Ginevra. Così, Francesco indossò la tonaca il 10 maggio 1593 e il giorno successivo divenne canonico di Annecy; il 13 maggio rinunciò alla primogenitura e al titolo di signore di Villaroger. Si ritirò quindi nel castello di Sales fino al 7 giugno 1593, lottando contro i suoi dubbi e le sue tentazioni. Ricevette il diaconato l'11 giugno 1593; il 18 dicembre dello stesso anno fu ordinato sacerdote e quindi divenne prevosto di Ginevra.Le sue straordinarie qualità sacerdotali (calma, dolcezza nelle parole e nell'approccio ai fedeli, un'eloquenza rara) fecero di lui un ottimo strumento per la conversione delle anime. L'8 dicembre 1602 Francesco di Sales fu consacrato, a Thorens, vescovo di Ginevra. Servì la sede episcopale in esilio ad Annecy, poiché Ginevra era divenuta una roccaforte protestante. Come nuovo vescovo, decise di istituire il catechismo per diffondere, far conoscere e far comprendere la fede cattolica ai credenti della sua diocesi. I suoi seguaci lo chiamavano "l'amabile Cristo di Ginevra".L'INCONTRO FECONDO CON SANTA GIOVANNA DI CHANTAL Nel marzo 1604, a Francesco fu chiesto di tenere i sermoni quaresimali a Digione. Lì conobbe la baronessa Giovanna Francesca di Chantal (1572-1641). Vedendola, Francesco credette di riconoscere colei che gli era apparsa durante una visione e che avrebbe dovuto fondare un nuovo ordine religioso. Qualche tempo dopo, divenne direttore spirituale della stessa Giovanna di Chantal, oggi santa, alla quale disse: "Tutto si deve fare per amore, e niente per forza, bisogna amare l'obbedienza più che temere la disobbedienza". Insieme a lei, avrebbe poi fondato l'Ordine della Visitazione.San Francesco di Sales è stato anche uno scrittore notevole, uno dei primi ad usare il francese contemporaneo nei suoi scritti per avvicinarsi ai lettori. Nel 1608, scrisse la sua opera più famosa, l'Introduzione alla vita devota (o Filotea). Inizialmente, il santo aveva scritto a Madame de Charmoisy, la moglie di un cugino che desiderava praticare una vita di preghiera. Per due anni, le diede consigli spirituali, finché la guida spirituale di Francesco lo esortò a pensare a una pubblicazione. L'autore, dopo una serie di opportune modifiche, pubblicò quindi quei consigli sotto il titolo di Introduzione alla vita devota. Il linguaggio e lo stile utilizzati in quest'opera erano molto semplici per l'epoca, senza citazioni latine o greche. Offrendo devoti consigli a uomini e donne di ogni stato di vita, si rivolgeva a un pubblico molto più vasto rispetto a molti altri trattati spirituali dell'epoca.Riguardo al protestantesimo, si distinse per la carità con cui si proponeva di riportare le anime alla Chiesa cattolica: "È con la carità che le mura di Ginevra devono essere scosse, con la carità che deve essere invasa, con la carità che deve essere recuperata [...]. Non ti offro ferro, né questa polvere il cui odore e sapore ricorda la fornace infernale [...]. È da noi stessi che dobbiamo respingere il nemico [...], con l'esempio e la santità della nostra vita [...]. Dobbiamo abbattere le mura di Ginevra con preghiere ardenti e lanciare l'assalto con fraterna carità".Francesco di Sales è stato canonizzato nel 1665 da Alessandro VII. Nel 1877 il beato Pio IX lo ha proclamato Dottore della Chiesa. E nel 1923, Pio XI lo ha dichiarato patrono dei giornalisti e degli scrittori cattolici.

Abstract: The ependyma lining the third ventricle (3V) in the mediobasal hypothalamus is recognized as a critical player in controlling energy balance and glucose homeostasis. Its molecularly distinct cell types, including diverse tanycyte subpopulations and typical ependymal cells, confer a high functional heterogeneity. The study of gene expression profiles and dynamics of ependymal cells has the potential to uncover fundamental mechanisms and pathways involved in metabolic regulation. Here, we cataloged 5481 hypothalamic ependymocytes using FACS-assisted single-cell RNA sequencing from fed, 12h-fasted, and 24h-fasted adult male mice. First, standard clustering analysis revealed the limitation of the current characterization regarding the different ependymal cell subpopulations along the 3V. Indeed, while typical ependymal cells and {beta}2-tanycytes are sharply defined at the molecular level, other subpopulations (i.e., {beta}1-, 2-, and 1 tanycytes) display fuzzy boundaries and very few specific markers. Moreover, we observed that 12h- and 24h-fasting dynamically modulate gene expression, increasing tanycyte subgroup heterogeneity. Secondly, pseudospatial trajectory analysis based on peculiar UMAP neuroanatomical distribution improved the identification of tanycyte markers, distinguishing specific versus overlapping features and better segregating tanycyte specific versus standard functions. Intriguingly, we discovered numerous functions related to tanycyte-neuron and tanycyte-synapse interactions with modulation by energy balance. Finally, combining pseudospatial analysis and gene regulatory network inference, we observed that fasting dynamically shifts patterns in gene expression and transcription activity along the 3v, creating a metabolic and functional switch for some subpopulations. Altogether, this data provides a mechanism through which energy status leads to distinct cell type-specific responses along the 3V and gives new insights into molecular diversity underlying tanycyte classification. Walking with the Saints | January 24 - Feast of St. Francis de Sales, Patron of Writers and Journalists Have you heard about the Gentle Saint? Most saints possess gentleness as a virtue, but there is one who seemed to exceed all the others, that is why Louise Stacpoole-Kenny, an author of the 19th and 20th centuries wrote a book about him entitled: THE GENTLE SAINT. Friends, our saint for today is St. Francis de Sales, a Frenchman who was born on August 21, 1567 in the Castle of Sales, to a noble family in Thorens, Savoy, France. His family expected nothing from him, as he was born two months premature, but God destined him to be a priest, a Bishop, and finally, a great saint. Francis‘ father sent him to the best school to prepare him to become a lawyer, so even at a young age, he enjoyed a privileged education under the Capuchin Fathers. In 1578, he went to study at the Jesuit's College de Clermont, and took up rhetoric and humanities. Being a nobleman, he was accompanied by a servant and a tutor. To please his father, he also took lessons in horse riding, dancing, and fencing. He was handsome, intelligent, tall, with blue-grey eyes, reserved and quiet. In 1586, after attending a theological discussion on predestination, he began to doubt his eternal salvation. This made him sick as he feared damnation in hell. After a few months, he visited a certain church and prayed to the Black Madonna, and consecrated himself to her. He decided to dedicate his life to God and made a vow of chastity. Soon his doubts vanished and he continued his studies. From College de Clermont, he enrolled at the University of Padua and studied law and theology. Meanwhile, Francis' father tried to find him a prestigious position in Savoy and a bride in the person of a beautiful and noble heiress, but Francis opposed his father's wishes and decided to become a priest. He shaved his lovely hair and when his father consented, Francis ceded to his brother the right of succession to his father's position as Lord of Sales. He was ordained in 1593. As a priest, he preached at the Cathedral of Annecy, in parish churches, and before Catholic confraternities. His sermons were simple and short. He avoided controversy and people loved to listen to his deep and rich voice. He was recommended at age twenty-seven to be named Grand Penitentiary of the diocese, one to whom people go to make Confession. In 1594, Fr. Francis was sent to Chamblais, where most people have become Calvinists, but he met many oppositions and had little success. In 1599, he was appointed co-adjutor Bishop of Geneva. In 1602 he was sent to Henry IV of France to negotiate the restoration of Catholic worship in Gex and he was successful. In 1602, the Bishop of Geneva died and he was consecrated Bishop of Geneva. However, he resided in Annecy because Geneva was then still controlled by the Calvinist. Due to his guidance, his diocese became famous throughout Europe as a model for efficiency in organization, devoted clergy, and well-instructed laity. He worked closely with the Capuchins that they made him an associate member of the Order, the highest honor given to a non-member. He was loved for his patience, kindness, and mildness. Francis was also the author of many books, the most famous of which is Introduction to the Devout Life. He was also co-founder of St. Jane Frances de Chantal of the Order of Visitation of Holy Mary. In December 1622, he was obliged to travel to Lyons with the Duke of Savoy. While there, he suffered a stroke and died on December 28. He was buried in the church of the Visitation Monastery in Annecy. He was beatified in 1616 and was canonized four years later. He was declared Doctor of the Church by Pope Pius IX in 1877. In 1923, Pope Pius XI proclaimed him patron of writers and journalists. He is named The “Gentle Saint” due to his kindness, patience, gentleness, and charity, characteristics not usually inherent to a guy.

The future St. Francis de Sales was born in a chateau on the shores of Lake Geneva on August 21, 1567. His family was part of the House of Savoie, a family of Nobles in Europe at the time. He was baptised the next day in the Parish church of Thorens, and given the name Francis Bonaventure. The Beatification of St. Francis de Sales in 1662 was the first solemn beatification to take place in St. Peter’s Basilica in Rome. De niño Francisco fue muy delicado de salud ya que nació prematuro; pero gracias al cuidado que recibía, se pudo recuperar y fortalecerse con los año. La madre de Fransciso fue Francisca de Boisy una mujer sumamente amable y profundamente piadosa. San Francisco fue beatificado por el Papa Alejandro VII en el 1661, y el mismo Papa lo canonizó en el 1665, a los 43 años de su muerte. En el 1878 el Papa Pío IX, considerando que los tres libros famosos del santo: "Las controversias"(contra los protestantes); La Introducción a la Vida Devota" (o Filotea) y El Tratado del Amor de Dios (o Teótimo), tanto como la colección de sus sermones, son verdaderos tesoros de sabiduría, declaró a San Francisco de Sales "Doctor de la Iglesia", siendo llamado "El Doctor de la amabilidad". Werkschau im Kontext zeitgenössischer Kunst | Werkvorstellung De holländische Künstler Zeger Reyers spricht in der Ausstellung über die Arbeit Zeger Reyers / Lee Ranaldo: »Träge Thorens« (2001-2007) Der amerikanische Künstler Paul Thek (1933-1988) genießt unter den zeitgenössischen Künstlern Kultstatus – durch sein spannungsreiches Œuvre, in dem er Themen und Symbole aus Religion, Kunst, Theater und Literatur aufgreift, nimmt sein Werk bis heute bedeutenden Einfluss auf die Gegenwartskunst. Zeichnung, Malerei, Skulptur, Installation und raumgreifende Environments gehören gleichermaßen zu Theks Ausdrucksmitteln. Seine Kunstwerke zeigen sich mystisch, religiös und kritisch sozialen Ungerechtigkeiten wie auch der Kunstwelt gegenüber, wobei er mitunter widersprüchlich zwischen nomadieserendem Hippietum und verklärendem Charismatiker pendelt, aber auch nicht selten eine Portion Melancholie und Humor in seine Werke mit hinein spielt. Bekannt wurde Paul Thek mit der Serie Technological Reliquaries (Technologische Reliquienschreine) – Plexiglaskästen mit realistisch anmutenden Fleischstücken aus Wachs, die er zum ersten Mal 1964 in New York ausstellte. The hitherto most comprehensive retrospective of his oeuvre focused on the phenomenal effect of his work on contemporary art and established Thek's historical significance, from legendary outsider to the founder and center of an art movement. It has been possible to bring together more than 300 of Thek’s works, which are largely in private ownership and therefore only seldom publicly shown. In their anti-heroic diversity and multimediality, and with their references to art, literature, and religion, his works (painting, photography, video, sculpture, and extensive environments) are among the central sources for the revolt and eruption of art in the 1960s. It is mainly for this reason that one of the early theoretical masterpieces of this epoch, Susan Sontag's Against Interpretation (1962), was dedicated to him. The mold castings, also those of his own body parts, wax replicas of human tissue, hair, teeth, and bones in Plexiglas cases, which he produced between 1964 and 1967 as Technological Reliquaries, in their mixture of desire and repulsion, decay and pathos, held up the truth of the body to the world of commodities and the transfiguration of the everyday, as well as the idealization and dramatization of corporative minimal art.
