Abstract: The median eminence (ME), located at the base of the hypothalamus, is an essential centre of information exchange between the brain and the pituitary. We and others previously showed that mutations and duplications affecting the transcription factor SOX3/Sox3 result in hypopituitarism, and this is likely of hypothalamic origin. We demonstrate here that the absence of Sox3 predominantly affects the ME with phenotypes that first occur in juvenile animals, despite the embryonic onset of SOX3 expression. In the pituitary, reduction in hormone levels correlates with a lack of endocrine cell maturation. In parallel, ME NG2-glia renewal and oligodendrocytic differentiation potential are affected. We further show that low-dose aspirin treatment, which is known to affect NG2-glia, or changes in gut microbiota, rescue both proliferative defects and hypopituitarism in Sox3 mutants. Our study highlights a central role of NG2-glia for ME function during a transitional period of post-natal development, and indicates their sensitivity to environmental signals. Abstract: Oligodendrocytes are generated via a two-step mechanism from pluripotent neural stem cells (NSCs): after differentiation of NSCs to oligodendrocyte precursor/NG2 cells (OPCs), they further develop into mature oligodendrocytes. The first step of this differentiation process is only incompletely understood. In this study, we utilized the neurosphere assay to investigate NSC to OPC differentiation in a time course-dependent manner by mass spectrometry-based (phospho-) proteomics. We identify double cortin like kinase 1 (Dclk1) as one of the most prominently regulated proteins in both datasets, and show that it undergoes a gradual transition between its short/long isoform during NSC to OPC differentiation. This is regulated by phosphorylation of its SP-rich region, resulting in inhibition of proteolytic Dclk1 long cleavage, and therefore Dclk1 short generation. Through interactome analyses of different Dclk1 isoforms by proximity biotinylation, we characterize their individual putative interaction partners and substrates. Abstract: Oligodendrocyte-lineage cells of patients and animal models undergo prominent changes in various neurodegenerative disorders. This raises the question of how myelinating cells interact with neurodegenerative processes. Here, we investigated the role of oligodendrocyte precursor cells (NG2 glia) in prion infections. We found that NG2 glia were activated in prion-infected cerebellar organotypic cultured slices (COCS) and in brains of prion-inoculated mice. In both model systems, depletion of NG2 glia exacerbated prion-induced neurodegeneration and accelerated prion pathology, suggesting a protective effect for NG2 glia. Loss of NG2 glia unleashed a microglial reaction promoting the biosynthesis of prostaglandin E2 (PGE2), which augmented prion toxicity in HovS cells and COCS through binding to the EP1 and EP4 receptors. Pharmacological or genetic inhibition of PGE2 biosynthesis attenuated prion-induced neurodegeneration in COCS and mice, antagonized the enhanced neurodegeneration in NG2 glia-depleted COCS and brains after prion infection, and dampened the acceleration of prion disease in NG2 glia-depleted mice. These data unveil a non-cell-autonomous interaction involving NG2 glia and microglia in prion disease and suggest that PGE2 signaling may represent an actionable target against prion diseases. Abstract: Glia cells expressing neuron-glial antigen 2 (NG2) play a critical role as oligodendrocyte precursor cells (OPCs) in the healthy brain; however, their differentiation potential after ischemic injury remains an unresolved question. Here, we aimed to elucidate the heterogeneity and role of NG2 glia in the ischemic brain. We used transgenic mice to label NG2-expressing cells and their progeny with red fluorescent protein tdTomato in the healthy brains and those after focal cerebral ischemia (FCI). Based on single-cell RNA sequencing, the labeled glial cells were divided into five distinct subpopulations. The identity of these subpopulations was determined based on gene expression patterns. In addition, membrane properties were further analyzed using the patch-clamp technique. Three of the observed subpopulations represented OPCs, whereas the fourth group exhibited characteristics of cells destined for oligodendrocyte fate. The fifth subpopulation of NG2 glia carried astrocytic markers. Importantly, we detected features of neural progenitors in these cells. This subpopulation was present in both healthy and post-ischemic tissue; however, its gene expression changed after ischemia, with genes related to neurogenesis being more abundant. Neurogenic gene expression was monitored over time and complemented by immunohistochemical staining, which showed increased numbers of Purkinje cell protein 4-positive NG2 cells at the edge of the ischemic lesion 12 days after FCI, and NeuN-positive NG2 cells 28 days after injury, indicating the existence of neuron-like cells that develop from NG2 glia in the ischemic tissue. Our results provide further insight into the differentiation plasticity and neurogenic potential of NG2 glia after stroke. Copy rights belong to original authors. Abstract: Adult mammalian spinal cord stops re-entering sensory axons, resulting in incurable sensory loss after dorsal root injury. Challenging the prevailing view, we previously showed that axons rapidly stop by forming synaptic, not dystrophic endings, and that myelin inhibitors and CSPGs are not the primary mechanism arresting axons. Here, we report that in adult mice dystrophic endings are formed by occasional axons embedded in fibrous collagens, exclusively at the CNS:PNS boundary where oligodendrocyte precursor cells (OPCs, or NG2 glia) are sparse. Most axons stop beyond the boundary in extensive contacts with OPC processes. Live imaging, immuno-EM and OPC-DRG co-culture show that axonal synapses form on OPCs. Genetic OPC ablation, although incomplete, enables many axons to continue regenerating deep into the spinal cord. These data, together with earlier findings, conclusively demonstrate that axons stop largely because OPCs stimulate presynaptic differentiation. Tito started his career as a Salsa Dancer by his parent's initiative, both musicians.  A top level dancer of many genres such as: Jazz, Merengue, "Bomba y Plena", Rumba, Modern Dance and Tap among others. He has visited more than 100 cities worldwide giving performances and classes, with his wife and dance partner Tamara Livolsi.Having participated in various television commercials, dance videos and movies such as, "Dirty Dancing: Havana Nights", Yellow, El Cantante: Hector Lavoe and American Gangster as dancers, he is no stranger to the stage.Today, Tito is not only a Salsa Dancer, he also is the Director of the San Juan City Salsa Dance Program and has made the choreographic works for concerts of artists such as: Gilberto Santa Rosa, Víctor Manuelle, El Gran Combo, Michael Stuart, Tito Nieves, Rubén Blades, Richie Rey y Bobby Cruz, Domingo Quiñones, Ismael Miranda, Elvis Crespo, Cheo Feliciano, Modesto Cepeda, Andy Montañez, NG2, Luis "Perico" Ortiz, Tommy Olivencia, Julio Voltio and Tego Calderón. He was also the choreographer for the "Bomba" and "Cha-Cha-Cha" routine in Ricky Martin's appearance in Miss Universe 2001 and the Celia Cruz "Azúcar" Special in Miami in 2003. Even after all this stardom, Tito remains humble, kind and readily shares his knowledge with so many!Don't forget to subscribe and leave a review!

Abstract: NG2 and A2B5 are important biological markers of human oligodendrocyte progenitor cells. To study their functional differences during the development of human oligodendrocyte progenitor cells to oligodendrocytes, we used cell sorting technology and obtained a large number of sterile, high-purity NG2+/- and A2B5+/- cells with high viability. Further research was then conducted via in vitro cell proliferation and migration assays, single-cell sequencing, mRNA sequencing, and cell transplantation into shiverer mice. The results showed that the migration ability of the cells was inversely proportional to the myelination ability. NG2 may be a marker of early oligodendrocyte progenitor cells and is conducive to cell migration and proliferation, while A2B5 may be a marker of slightly mature oligodendrocyte progenitor cells and is conducive to cell differentiation. Further, cell migration, proliferation, and myelination capacity of the negative cell population were stronger than those of the positive cell population. In summary, these results suggest that oligodendrocyte progenitor cells in the mid-stage may be more suitable for clinical cell transplantation to treat demyelinating diseases. M. Abstract: Oligodendrocyte progenitor cells (OPCs) are responsible for generating oligodendrocytes, the myelinating cells of the CNS. Life-long myelination is promoted by neuronal activity and is essential for neural network plasticity and learning. OPCs are known to contact synapses and it is proposed that neuronal synaptic activity in turn regulates OPC proliferation and differentiation. To examine this in the adult, we performed unilateral injection of the synaptic blocker botulinum neurotoxin A (BoNT/A) into the hippocampus of adult mice. We confirm BoNT/A cleaves SNAP-25 in the CA1 are of the hippocampus, which has been proven to block neurotransmission. Notably, synaptic silencing by BoNT/A significantly decreased OPC density and caused their shrinkage, as determined by immunolabelling for the OPC marker NG2. Inhibition of synaptic activity resulted in an overall decrease in the number of OPC processes, as well as a decrease in their lengths and branching frequency. These data indicate that synaptic activity is important for maintaining adult OPC numbers and cellular integrity, which is relevant to pathological scenarios characterized by decreased synaptic activity. M., Butt, A. M. Abstract: Myelin disruption is a feature of natural aging and of Alzheimer's disease (AD). In the CNS, myelin is produced by oligodendrocytes, which are generated throughout life by oligodendrocyte progenitor cells (OPCs). Here, we examined age-related changes in OPCs in APP/PS1 mice, a model for AD-like pathology, compared with non-transgenic (Tg) age-matched controls. Analysis was performed in the CA1 area of the hippocampus following immunolabelling for NG2 with the nuclear dye Hoescht, to identify OPC and OPC sister cells, a measure of OPC replication, together with Gpr17 and Olig2 for oligodendrocytes and myelin basic protein (MBP) immunostaining as a measure of myelination. The results indicate a decrease in the number of OPCs between 9 and 14 months in natural ageing and this occurred earlier at 9 months in APP/PS1 mice, without further decline at 14 months. The number of OPC sister cells was unaltered in natural aging, but declined significantly at 14-months in APP/PS1 mice. The number of GPR17+ and Olig2+ oligodendrocytes was not altered in APP/PS1, whereas MBP immunostaining increased between 9 and 14 months in natural ageing, but not in APP/PS1 mice. Notably, OPCs displayed marked morphological changes at 14 months in APP/PS1 mice, characterized by an overall shrinkage of OPC process domains and increased process branching, characteristic of reactive pathological changes. While studies have identified myelination processes and oligodendrocytes as targets of early adversity across species, knowledge about the precise molecular mechanisms and cell populations involved is still lacking. NG2+ cells are oligodendrocyte precursor cells with unique properties, as they form synapses with neurons, engage in cell-cell communication and respond to stress hormones. Methods: Using an established mouse model of early life stress (ELS) we performed cell-type specific molecular profiling in hippocampal NG2 cells at early postnatal and adult stages. To further dissect the impact of glucocorticoids on ELS-induced transcriptional changes, we integrated our data with available Chip-seq data on genomic binding sites of the glucocorticoid receptor (GR). The functional relevance of one candidate gene, Scn7a, was confirmed by electrophysiological recordings in hippocampal NG2 cells. Results: ELS specifically targeted the hippocampal NG2 cell transcriptome, and the molecular changes correlated with the ELS-induced increase of corticosterone. The overlap analysis with Chip-seq data on genomic binding sites of the GR revealed nine overlapping genes. Among those Scn7a, coding for a subunit of sodium channels, remained upregulated until adulthood in ELS animals. Upregulation of Scn7a was accompanied by an increase in the density of voltage-gated sodium channel activated currents in hippocampal NG2 cells. Conclusions: Our findings indicate that ELS specifically targets the transcriptional profiles and electrophysiological properties of hippocampal NG2 glia. Considering that voltage-gated sodium channels are important for the NG2 cell-to-neuron communication, our findings may provide novel insights into the pathophysiological processes underlying stress related mental disorders. Abstract: Pericytes are a unique class of mural cells essential for angiogenesis, maintenance of the vasculature and are key players in microvascular pathology. However, their diversity and specific roles are poorly understood, limiting our insight into vascular physiology and the ability to develop effective therapies. Here, in the mouse retina, a tractable model of the CNS, we evaluated distinct classes of mural cells along the vascular tree for both structural characterization and physiological manipulation of blood flow. To accomplish this, we first tested three inducible mural cell-specific mouse lines using a sensitive Ai14 reporter and tamoxifen application either by a systemic injection, or by local administration in the form of eye drops. Across three lines, under either the PDGFRb or NG2 promoter, the specificity and pattern of Cre activation varied significantly. In particular, a mouse line with Cre under the NG2 promoter resulted in sparse TdTomato labeling of mural cells, allowing for an unambiguous characterization of anatomical features of individual sphincter cells and capillary pericytes. Furthermore, in one PDGFRb line, we found that focal eye drop application of tamoxifen led to an exclusive Cre-activation in pericytes, without affecting arterial mural cells. We then used this approach to boost capillary blood flow by selective expression of Halorhodopsin, a highly precise hyperpolarizing optogenetic actuator. The ability to exclusively target capillary pericytes may prove a precise and potentially powerful tool to treat microcirculation deficit, a common pathology in numerous diseases. We assessed whether Angiopoietin-2 (Ang2), a Tie2 ligand and partial antagonist of Angiopoietin-1 (Ang1), is required for early vessel destabilization during postischemic angiogenesis, when combined with vascular growth factors. In vitro, matrigel co-cultures assessed endothelial-cell tube formation and pericyte recruitment after stimulation of VEGF-A, Apelin (APLN), Ang1 with or without Ang2. In a murine hindlimb ischemia model, adeno-associated virus (rAAV, 3×10(12) virusparticles) transduction of VEGF-A, APLN and Ang1 with or without Ang2 (continuous or early expression d0-3) was performed intramuscularly (d-14). Femoral artery ligation was performed at d0, followed by laser doppler perfusion meassurements (LDI) 7 and 14. At d7 (early timepoint) and d14 (late timepoint), histological analysis of capillary/muscle fiber ratio (CMF-R, PECAM-1) and pericyte/capillary ratio (PC-R, NG2) was performed. In vitro, VEGF-A, APLN and Ang1 induced ring formation, but only APLN and Ang1 recruited pericytes. Ang2 did not affect tube formation by APLN, but reduced pericyte recruitment after APLN or Ang1 overexpression. In vivo, rAAV.VEGF-A did not alter LDI-perfusion at d14, consistent with an impaired PC-R despite a rise in CMF-R. rAAV.APLN improved perfusion at d14, with or without continuous Ang2, increasing CMF-R and PC-R. rAAV.Ang1 improved perfusion at d14, when combined with rAAV.Ang2 (d0-3), accompanied by an increased CMF-R and PC-R. The combination of early vessel destabilization (Ang2 d0-3) and continuous Ang1 overexpression improves hindlimb perfusion, pointing to the importance of early vessel destabilization and subsequent vessel maturation for enhanced therapeutic neovascularization.

Skeletal muscle pericytes as signal mediators in the vessel wall

In this thesis a novel method to isolate and cultivate pericytes from hamster skeletal muscle is presented. By choosing a two-step approach with magnetic-bead cell sorting for the 3G5 antigen and culture in a selective growth medium a high percentage of pericytes could be reached and their nature could be verified in subsequent characterization steps. Morphologically the cells had a typical phenotype with flat and large cell bodies and long protruding processes. Immunocytochemistry and western blotting could confirm the expression of the previously reported pericyte markers NG2, PDGFR-β, and αSMA. However, in accordance with previous reports, even the cells isolated from the same muscle and tissue, were not a uniform population. A certain heterogeneity was also present in the functional studies investigating pericyte membrane channels by electrophysiology. In these experiments in a subset of the pericyte population the presence of the voltage sensitive potassium channel KV1.5 could be demonstrated. Upon pharmacological stimulation of these channels by FFA they were able to elicit a membrane hyperpolarization and conduct it to neighboring endothelial cells via gap junctions formed by connexin 43. The expression of the KV1.5 channel could also be demonstrated by immunohistochemistry in paraffin sections from hamster skeletal muscle. These findings are consistent with the hypothesis that pericytes could act as signal generators for hyperpolarization and vasodilatation in the vessel wall. The coupling with endothelial cells might relay an electric signal further along the vessel wall and lead to “EDHF mediated” dilations [48]. Interestingly in tissue sections KV1.5 positive pericytes were located predominantly at vascular branchings. We additionally studied whether gap junctions may also form a pathway for the exchange of miRNA between cells in a model co-culture system using HeLa. For the detection of the miRNA a detector system consisting of a luciferase reporter protein under control of a miR124 sensitive 3’ UTR was established. Our experiments could not conclusively confirm transfer of miRNA in this system. Similar results were obtained with human miR15b. Taken together these findings open up new possibilities for studying skeletal muscle pericyte physiology and to test novel concepts integrating the pericyte into the vascular signaling network under conditions of health and disease.
