D-Wave has been making quantum computers for over a decade, and their annealers excel at optimization. If you look at the use cases the company has been helping customers with, it almost feels like quantum advantage is here. If you're saving time or money, or both, how can you complain? And even after the other type of quantum computers, universal-gate-based systems, become fault tolerant, annealers will likely be around forever due to how powerful they are at specific use cases (sort of like how graphics cards work alongside CPUs). C., Tuma, R., Calabrese, A. N., Borodavka, A. Abstract: Rotavirus genomes are distributed between 11 distinct RNA segments, all of which are essential for virus replication. Stoichiometric genome segment selection and assembly is achieved through a series of sequence-specific, intersegment RNA-RNA interactions that are facilitated by the rotavirus RNA chaperone protein NSP2. The C-terminal region (CTR) of NSP2 has been proposed to play a role in rotavirus replication, although its mechanistic contribution to the RNA chaperone activity of NSP2 remained unknown. Here, we use single-molecule fluorescence assays to directly demonstrate that the CTR is required for promoting RNA-RNA interactions and that it limits the RNA unwinding activity of NSP2. Unexpectedly, hydrogen-deuterium exchange-mass spectrometry and UV-crosslinking data indicate that the CTR does not interact with RNA. However, removal of the CTR reduced the RNA release activity of NSP2, suggesting that the CTR is important for chaperone recycling. To further interrogate the role of the CTR, we determined cryo-EM structures of NSP2 and its ribonucleoprotein complexes. These reveal that although the CTR is ampholytic in nature, it harbours a highly conserved acidic patch that is poised towards bound RNA. Using a reverse genetics approach, we demonstrate that rotavirus mutants harbouring triple alanine mutations within the acidic patch failed to replicate, while mutations that preserve the charge of the CTR successfully restored viral replication. Together, our data suggest that the CTR reduces the accumulation of kinetically trapped NSP2-RNA complexes by accelerating RNA dissociation via charge repulsion, thus promoting efficient intermolecular RNA-RNA interactions during segment assembly. Chicago Quantum have recently been publishing some interesting results, e.g. “ Portfolio Optimization of 40 Stocks Using the D-Wave Quantum Annealer ”. They've investigated the use of quantum computers for building a portfolio out of a universe of US listed, liquid equities that contains an optimal set of stocks. Starting from historical market data, they examined formulations on the D-Wave 2000Q System (DWave) to find optimal risk vs return portfolios; an optimized portfolio based on the Markowitz formulation and the Sharpe ratio, a simplified Chicago Quantum Ratio (CQR), then a new Chicago Quantum Net Score (CQNS). Their results show that practitioners can use a DWave to select attractive portfolios out of up to 60 US liquid equities. This webinar will examine what may well be the earliest actual quantum computing application in finance since Z/Yen first looked at the implications of quantum computing back in 1998, The Potential Impact Of Quantum Computing , and the formation of the Quantum Computing in Europe Pathfinder Project for the European Commission with Charles T Ross. There will an opportunity to get a feel for the quantum computer ‘rig' being used. The webinar will also discuss the performance outcomes of two of the portfolios, and speculate at what point we should expect to see quantum advantage in stock picking. Speaker: Jeffrey Cohen is an executive IT strategy management consultant. Classically educated in finance, economics and management sciences. Jeff has spent his career leading and growing IT infrastructure consulting and project businesses (largest had $447M sales globally). Jeff has changed tack and spent the past two years building a start-up in quantum computing consulting. His industry experience is deepest in financial services, but he is comfortable supporting the intersection of finance and IT infrastructure across industries and geographies. For example, Jeffrey co-led the North American Outsourcing and Offshoring practice in the McKinsey & Company BTO. Jeff's work grows IT consultancies, typically in new areas of market innovation. He started in IT infrastructure, then moved to hybrid cloud, and is now focused on quantum computing. On a personal note, Jeffrey is involved in politics and after holding the office of Precinct Committeeman, is up for election for North Shore Water Reclamation District Trustee in Lake County, Illinois in November 2020. It’s episode #97 and I’ve got another JayBurn Journal for you.  A technical article written by Jay Burnham-Kidwell. He’s a longtime blacksmith residing in Arizona, Since 1974, he has worked in various mediums and exhibited his work throughout the world.  He works as a studio artist, lecturer, and demonstrator in all kinds of metalsmithing including jewelry, copper hollow forms, and blacksmithing.  He’s written more than 40 technical articles for various magazines such as the Anvils Ring and Anvil Magazine.  So a JayBurn Journal titled “Punches, Drifts, Hammers and Top tools”.  Show Notes: To produce a tool of quality the smith should use tool or alloy steels. Tool steels can be bought new or as a drop or discontinued stock or as “road kill” recycled steel scraps. The average blacksmith shop has adequate resources to forge, normalize, anneal and heat treat many tool steels safely and most available tools steels can be forged and heat treated by using modified methods of the manufacturers recommended procedures. Steel is a body-centered cubic crystal at room temperature. When heated to critical temperature, iron and steel undergo a phase change and become a face-centered cubic crystal structure.  The hardening process changes the internal structure of the steel to form austenite.  When quenched, the austenite is transformed into martensite, the hardest constituent in steel. Most tool steels generally trade one quality for another: wear resistance vs. toughness and accuracy vs. red hardness. Wear resistance is preferable when the tool must hold an edge or stand up to continued service.  Toughness is needed for tools that are used under stress that may cause breakage.  Accuracy addresses machining after forging and the ability to retain shape after heat treating.  Red Hardness is the ability to retain shape and hardness when used at high temperatures (punching, chiseling, drifting). Normalizing – most, but not all, tool and alloy steels are normalized after forging by air cooling to remove most of the stress introduced by the forging process. Annealing – heating to critical temperature (nonmagnetic) and slowly cooling will restore varying degrees of softness in tools steels. This requires burying the steel in wood ash, lime, dry dirt or sand.  You can use vermiculite, but know that you should wear a respirator because it contains asbestos. Machining – Most, if not all, tools require some grinding, filing or sanding after forging and before heat treating, as annealed steel is in a softer state and is obviously easier to grind or sand. Hardening – tool steels are heated to a specified critical temperature and quenched in the correct medium for optimum hardening. Tools steels are usually classified as air, water or oil hardening.  Air hardening is accomplished by heating to critical temperature and cooling in still air.  Water hardening is usually done in a 5 – 10% brine solution.  Oil hardening is accomplished in commercial heat treating oils or vegetable oils. Tempering – hardening will produce maximum hardness and must be softened or tempered because the steel is too brittle at this point. The tool should be tempered as soon as possible after hardening.  Tempering temperatures usually run between 300 – 600 f. W-1 tool steel – a straight carbon, water hardening tool steel that tends to chip or break. Old files were made from W-1 O -1 tool steel – a straight carbon, oil hardening steel classified for coldwork, chasing and repousse. S series tool steel – very tough and exhibit very good red hardness. Due to varying amounts of tungsten and chromium, it can be a bit tough to forge. 4140 – a chromium molybdenum alloy steel that has great toughness, resists torque and can be used as punches, hammers and top tools. 4340 tool steel – a close relative to 4140 but with some added nickel. NEVER quench or cool in water, use only oil. The surface of an ordered Pt80Fe20(110) crystal exhibits (1×2) and (1×3) reconstructions depending on the annealing treatment after ion bombardment. The (1×3) structure occurs after annealing in the range 750 to 900 K. Annealing above 1000 K leads to the (1×2) structure, which is, from the present result, unambiguously attributed to the same geometrical reconstruction as Pt(110) but with smaller relaxation amplitudes: a detailed low-energy electron-diffraction analysis concludes to a missing-row structure with row pairing in layers 2 and 4 accompanied by a buckling in layers 3 and 5. The top layer spacing is contracted by 13%, and further relaxations are detectable down to the fifth layer. The specific diffraction spots associated with the bulk chemical ordering along the dense [1¯10] rows are very weak: The I(V) analysis shows that this chemical ordering is absent in the outermost ‘‘visible’’ rows but gradually recovers over five to six layers deep. General Pt enrichment is found in the surface ‘‘visible’’ rows (in layers 1–3), but segregation and order yield a subtle redistribution of Pt and Fe atoms in deeper rows: For example, in layer 2, the visible row is Pt rich, whereas the other row (buried under layer 1) is enriched with Fe. Because of the many parameters considered, a fit procedure was applied to a large data basis to solve the structure; the results were confirmed and illustrated subsequently by a standard I(V) analysis for the most relevant parameters. The final r factors are RDE=0.36, RP=0.34, and RZJ=0.14 for two beam sets at normal and oblique incidence consisting of 26 and 21 beams, respectively.

Positron lifetime measurements have been performed to study vacancy defects in Hg0.78Cd0.22Te. Post-growth annealing under various Hg vapour pressure conditions have been used to create a well-defined number of Hg vacancies. The sensitivity range of the positron annihilation method was found to be 1015 < cHgvac
