In this episode of the Epigenetics Podcast, we talked with Kyle Eagen from Baylor College of Medicine about his work on BET Proteins and their role in chromosome folding and compartmentalization. In the early days of his research career Dr. Eagen made use of genomics and microscopy to study chromosomes, particularly polytene chromosomes in Drosophila. The correlation between the folding patterns detected by Hi-C and polytene bands highlights the similarities between the two, bridging traditional cytology with modern NGS methods. This work formed the basis of Kyle's thesis and sparked his interest in nuclear organization and chromosome 3D structure. In his independent lab Kyle then studied compartments in chromatin structure and focused on the relationship between histone modifications and the 3D structure of chromosomes. The discovery of BRD4-NUT, a fusion oncoprotein that reprograms chromosome 3D structure, is highlighted as a significant step forward in understanding chromatin structure. The conversation then shifts to the use of a tool to test hypotheses about the involvement of BRD4 in a specific process, leading to consistent results and considerations for manipulating chromosome organization for therapeutic purposes. Three decades ago, Esther Gokhale created the Gokhale Method to apply primal posture to modern pain. Gokhale emphasized small improvements in everyday movements to prevent and heal back pain and musculoskeletal damage. Now, more than 20,000 people have learned to walk, sit, bend and sleep more efficiently by adopting Gokhale's primal posture approach. Dr. D.J. Kennedy, professor and chair of the Department of Physical Medicine and Rehabilitation at Vanderbilt University Medical Center, told us how he sees the Gokhale method work for his patients in this episode. “I think she paints a picture that makes a lot of sense intuitively. And she's also going about trying to get the data to prove it,” said Kennedy. The Gokhale method is about to be tested in a randomized clinical trial at Stanford University. In this MBO episode, we learn how Esther Gokhale developed the method in part due to her own suffering. Pregnant with her second child, Gokhale was experiencing severe back pain even after a corrective surgery. Gokhale tried physical therapy, chiropractic, acupuncture, massage, yoga, stretching, strengthening, and “checking her head.” Nothing helped. Thanks to an upbringing in India studying classical dance and observing the posture of local laborers, Gokhale knew that “something was wrong with this picture.” In the quest for relief, Gokhale built on those childhood observations and her own scientific background to create a new theory of movement based on traditional practices. Soon after, she published Eight Steps to Pain Free Back, which has sold over 300,000 copies. Today, the Gokhale Method may be poised for even wider acceptance. In addition to online courses and a nationwide network of teachers, the Method now offers a Posture Tracker app to help students reinforce better posture. Meanwhile, Stanford University researchers are embarking on a long-term study of the system's efficacy in healing and preventing back pain and other musculoskeletal issues. A Supermassive Black Hole The Mass Of 30 Billion Suns This week, astronomers reported that they may have found signs of one of the largest black holes ever detected–a space behemoth the mass of some 30 billion suns. The supermassive black hole, located in part of the Abell 1201 galaxy cluster, was detected using a combination of gravitational lensing and supercomputer simulations. First, the astronomers observed how the images of other more distant objects viewed by the Hubble Space Telescope were warped by the vast gravitational well produced by the black hole. They compared those images to thousands of simulations created via a supercomputer, and found that a simulation containing a supermassive black hole matched the real-world images. The work was reported in the journal Monthly Notices of the Royal Astronomical Society. What's Driving A Rise In Mumps Cases In The United States? In 1971, the United States rolled out a revolutionary new vaccine for measles, mumps, and rubella. The MMR vaccine nearly eradicated all three of those viruses by the start of the 21st century. Over the last several years, there have been numerous measles outbreaks cropping up across the country, especially among unvaccinated kids. What about mumps—that second “m” in the MMR vaccine? Since 2006, there have been mumps outbreaks too. But unlike measles, most of the people getting the mumps are vaccinated. And they're older too, mostly teens and young adults. New research suggests that the efficacy of the mumps vaccine wanes over time, unlike the ones for measles and rubella. Guest host Shahla Farzan talks with Dr. Deven Gokhale, co-author of a recent study on the reemergence of mumps. Gokhale recently completed his PhD from the University of Georgia's Odum School of Ecology, based in Athens Georgia. Foundational Food Sources In The Gulf Of Maine Are Failing At the Bigelow Laboratory for Ocean Sciences, researchers Barney Balch and Catherine Mitchell are looking at a map affixed to a large table. “We're looking at a chart of the Gulf of Maine, and right across the middle there's a line that's drawn from Portland, in Maine, to Yarmouth, in Nova Scotia,” Mitchell says. That line is the route along which Bigelow researchers have been taking regular measurements for the last 25 years. They've analyzed chemical and temperature data that help describe how the waters of the gulf are changing. One tool they use is a six-foot long cylinder with wings. “This is an autonomous underwater vehicle, or a glider,” Mitchell says. “So it's a big robot that moves up and down in a yoyo-like pattern, from the top of the ocean to the bottom of the ocean right across the middle of the Gulf of Maine. 2022 has been a year of geopolitical conflict and tensions. If we were expecting a quieter end to the year, then we were apparently mistaken. On December 9, Chinese and Indian troops had a face-off along the Line of Actual Control (LAC) in the Tawang sector of Arunachal Pradesh. While we don't have many details yet, it appears that a few hundred soldiers were involved in a physical scuffle, and some thirty to forty, on both sides, sustained injuries. The Indian defence minister, Rajnath Singh, told the Parliament that the Chinese People's Liberation Army troops had tried to transgress the LAC in this area and were prevented from doing so. Two days after this incident, the local commanders of the Indian Army and the People's Liberation Army met to discuss the issue. Though it is unclear what, if anything, has been agreed upon to restore tranquility. The relations between India and China have been in deep freeze since May 2020, when troops of the two sides had clashed along the LAC in Ladakh. Both sides have since enhanced their military deployment and upgraded their logistical infrastructure along the LAC. Arunachal Pradesh has several points where the two sides have different perceptions of where the LAC runs, and both sides patrol up to the line they claim. Apropos the recent standoff, the Indian Ministry of Defence has noted that this has been the case in the Tawang sector since 2006. How do we understand China's posture and actions along the LAC in recent years? Is a purely bilateral framework adequate to grasp Beijing's motivations, or are larger considerations at work? And, what are India's options in dealing with Chinese activism along the LAC? In an interesting coincidence, just as the news of the recent face-off hit the headlines a couple of days ago, Carnegie India published an important paper by Mr. Vijay Gokhale titled “A Historical Evolution of China's India Policy: Lessons for India-China Relations”. While much has been written about India-China relations, most of it tends to be from the Indian perspective. We have few assessments of how Beijing has seen India and sought to deal with it. What was Girish's impact on the national theatre scene? Did his plays influence or inspire other playwrights and theatre makers? In Part 2 of this episode, Shanta Gokhale and Sunil Shanbag talk about how the 60s and 70s were such a critical moment for theatre in different parts of India. They talk about the unique sensibilities that Girish brought to the stage and how Girish set himself apart from his contemporaries such as Vijay Tendulkar. About the Series: In June 2019, a few days before his death, Girish Karnad and Arshia Sattar had extensive conversations about his work, his life, and the concerns that had occupied him over almost sixty years as a writer and a public intellectual. This podcast is a distillation of those freewheeling conversations, a short intellectual biography to celebrate the life and work of one of India's finest playwrights and thinkers. In this series, we hear Girish talk about Kannada literature, existentialism, making of modern Indian theatre, the art and craft of playwriting, and his life as a public intellectual. The series is hosted by Arshia Sattar and Anmol Tikoo, with special guests such as Vivek Shanbhag, Shanta Gokhale, and Sunil Shanbag, who provide the context for Girish's comments. Each episode also contains scenes from his plays read by members of Bangalore's theatre community. The readings show us how closely his philosophical and political ideas were to what he wrote. They also provide an opportunity for audiences, particularly those who might not be already familiar with Girish's works, to experience the power of his work. The title for the series is taken from the song in Hayavadana (ಹಯವದನ), a song which has been musically recreated for us by Pallavi MD and Konarak Reddy. Apart from the fact that Neerina Mele Chitra remains one of Kannada's most beloved ranga geethe, we found it to be particularly evocative as we remember a man who profoundly impacted India's cultural arena in the last half of the 20th century.
