The 2023 LaoisToday QuizCast came to a conclusion in the Square Bar in Portlaoise on Friday night when the four semi-finalists - Brian Hogan, Murt Norton, Colin Delaney and Jack Scully - battled it for supremacy in front of a live audience. With a full house and a cracking atmosphere, quizmaster Mark Comerford put the contestants through their paces. And though there was a high standard of questions, there was also a very high standard of answers with both semi-finals and final being well contested affairs. The final was only decided with the very last question. And as well as a thrilling decider, we also had an entertaining curtain raiser - where Team Alan did battle with Team Steven in the 'Senior B'. Team Alan was comprised of LaoisToday's Alan Hartnett as well as Timahoe father and son duo Pauric and Liam 'Sheriff' Ramsbottom while Steven Miller from LaoisToday was joined by Courtwood's Mick Hogan and Portarlington's Darren Bracken. J., Clegg, P. D. Abstract: Variation in Mesenchymal Stromal Cell (MSC) function depending on their origin is problematic, as it may confound clinical outcomes of MSC therapy. Current evidence suggests that the therapeutic benefits of MSCs is primarily attributed to secretion of various biologically active factors (secretome). However, the effect of donor characteristics on the MSC secretome composition remains largely unknown. Here, we examined the influence of donor age, sex and tissue source, on the protein profile of the equine MSC secretome. Initially, we used dynamic metabolic labelling with stable isotopes combined with liquid chromatography-tandem mass spectrometry (LC-MS/MS) to identify secreted proteins in MSC conditioned media (CM). Seventy proteins were classified as classically-secreted based on the rate of isotope label incorporation into newly synthesised proteins released into the extracellular space. Next, we analysed CM of bone marrow- (n = 14) and adipose-derived MSCs (n = 16) with label-free LC-MS/MS proteomics. Clustering analysis of 314 proteins detected across all samples identified tissue source as the main factor driving variability in MSC CM proteomes. Linear modelling applied to the subset of 70 secreted proteins identified tissue-related difference in the abundance of 23 proteins. There was an age-related decrease in the abundance of two proteins (CTHRC1, LOX), which has been validated with western blot and enzymatic activity assay. There was limited evidence of sex-related differences in protein abundance. In conclusion, this study provides evidence that tissue source and donor age contribute to heterogeneity in the protein composition of MSC secretomes which may influence the effects of MSC-based cell therapy. In this episode, the second in our Mudlark series, Anne Marie talks to Mudlark Lynn Pew. Lynn mudlarks along the River Irwell in Lancashire, England, where she scours its banks for beautiful pieces of broken pottery, little pieces of history tumbled by the river for over 100 years. Lynn tells us about the river's history and where many of the pieces she finds originated.Lynn is fascinated with how these tiny fragments made their way into the river and the journey they have been on. Cracked and faded, their edges are worn smooth from their watery passage, a mere remnant of what once was. Who last held them in their former, utilitarian life before she plucked them from the banks of the river.Years after she began collecting these lovely miniature pieces of history, she incorporated them into handcrafted one-of-a-kind mosaic works of art, which she entered and won a place in Country Living Magazine's 'Kitchen Table Talent' competition. This Thursday evening, ladies football great Sue Ramsbottom from Timahoe will become the first Laois person to feature on Laochra Gael, the iconic TG4 documentary series that profiles the top GAA personalities from around the country. The one-hour programme will air on TG4 at 9.30pm on Thursday and chronicles Sue's career, from her early days playing with the Timahoe school, the Stradbally parish boys team and The Heath ladies senior side. The documentary features incredible footage, including an interview when she was only 13 after helping The Heath win a senior All Ireland club title in 1987. It also includes interviews with her brother James and long-time team mate Connie Conway. And it journeys her career with Laois throughout the 1980s and 1990s - when Sue lost All Ireland finals in 1988, 1990, 1991, 1992, 1993 and 1996 before famously enjoying success in 2001. Ahead of the TV show, we caught up with Sue on this week's Talking Sport Podcast where Sue chatted about why she agreed to go on the programme, what it was like looking back and some of the great people she encountered along the way. Paul Ramsbottom OBE joins Purposely Podcast to share his leadership story. We also touched on how he got his name and what led him to be awarded an Officer of the British Empire. Paul is CEO of the Wolfson Foundation and has been volunteer founder Chairman of Savannah Education Trust for almost two decades. We discuss the incredible contrast between the two organisations, one a large charitable foundation donating millions of pounds to good causes annually and the other a small effective charity saving young lives in West Africa on a limited annual budget of just over £200k. Paul is passionate about both organisations and wearing two hats , as a funder and fundraiser, has added to his knowledge and ability to understand the struggle smaller charities have fund their services. Established in 1955 Wolfson Foundation has now passed over £1 billion milestone in terms of grants awarded (over £2 billion in today's money). The funds have gone to support and promote thousands of remarkable projects across virtually every community in the UK – and within education, science, medicine, heritage, humanities, the arts and health and disability. Paul also shares the fascinating story of the Wolfson family where their success in commerce led them to an impressive philanthropy journey. Paul attended Corpus Christi College, Oxford where he gained a First in modern history. Paul Ramsbottom is Chief Executive of the Wolfson Foundation and its sister charity, the Wolfson Family Charitable Trust. Paul takes a wider interest in issues relating to philanthropy in the UK as a speaker and writer. He has undergraduate and postgraduate degrees in History from the University of Oxford. Outside of philanthropy, he has an interest in international development which includes founding the Savannah Education Trust - a charity which works in West Africa – and sitting on the Board of Mercy Ships UK. His hometown university, the University of Bedfordshire, awarded him an honorary doctorate in 2013. Teagasc Dairy Specialist, George Ramsbottom, joins Emma-Louise Coffey on this week's Dairy Edge podcast to provide context on the appropriate time to finishing the breeding season in order to facilitate compact calving next spring. George quantifies the average 6 week calving rate being achieved nationally at 65%, owing the 10% increase in the last decade to a combination of improved management, genetics and reduced disease burden. He acknowledges that despite gains being made, there is significant progress required to meet the target 6 week calving rate of 90%. In spring calving herds where all cows are dried in December, later calvers have lower milk production (April 900 litres less, May 1,200 litres less and June 1,800 less) compared with a February calved cow producing 6,500 litres. George recommends a 10 week breeding season to facilitate a 10-11 week calving spread, allowing farmers a 2-3 week break between calving 2022 and mating start date. Although many Christians are keen to come to fully-fledged services, many remain anxious about the possibility of being infected or infecting others. And simply meeting once a week may simply be papering over the cracks of the loneliness people feel. How can the Church love lonely and fearful people? What is a Biblical and pastoral response to these issues? And what innovative ways are Christians sharing the hope of Christ, who is 'a very present help in trouble' (Ps 46:1).
