After an unplanned summer break, Scoreography returns to cover the 2024-2025 season, starting with the Challenger Series. We take a look at all the major competitors we've seen so far, specifically from the Nebelhorn and Lombardia Trophy competitions, as well as Cranberry Cup and other small events. Welcome back, Scoreogra-fans! (too much?) It's a new dawn, it's a new day, it's a new SEASON of figure skating — and we're so happy to be back! It was a long summer of far too much adult-ing, and we've missed so much in the world of figure skating. From the ISU's legalization of the backflip (or somersault) in competition to the 2022 Olympic teams from the USA and Japan FINALLY getting their medals at the 2024 games in Paris, we're definitely a bit behind. However, with a new competitive season already in progress, we're moving forward and talking about what we've already seen at early season Challenger Series events including Lombardia Trophy, Nebelhorn Trophy, and Cranberry Cup. Since its still quite early and most skater's programs aren't fully “baked” yet, in this episode we break down which skaters have competed so far by discipline — and dig into who's already looking strong. Women's Division The women of Team USA have come out hot already with some big moments for reigning U.S. Champion, Amber Glenn, and well as two young skaters who are starting to make their mark internationally: Elyce Lin-Gracey and Sarah Everhardt. Amber threw down a personal best short program at Lombardia Trophy — which included a gorgeous triple axel — and skated well enough in the free to earn her first gold at a Challenger Series event. Elyse Lin-Gracey was consistently gaining ground last season, but she looks like she's found her flow and looked incredible at Nebelhorn Trophy. She scored a personal best of 213.33 overall and made the skating world sit up and take notice. Also bringing the heat is Sarah Everhart, who looked fantastic at last year's U.S. Nationals where she landed in 4th overall. She started her season with a victory at Cranberry Cup over the likes of reigning World Silver medalist, Isabeau Levito and Elyse Lin-Gracey. Isabeau Levito, for her part, has perhaps not had the flawless programs and scores she would have liked so far — with a bronze at Cranberry Cup and a silver at Nebelhorn Trophy. However, we are not too concerned, as she's already shown a new short program that taps into an elegant, Audrey Hepburn vibe that looks like a winner to us. Plus, her free skate at Nebelhorn showed significant improvement from Cranberry Cup. We think these are just early competitions and that she looks to be right on track. Team Japan is already on the board with medals as well, with reigning 3-time world champion, Kaori Sakamoto, taking the bronze at Lombardia Trophy with a safe but solid competition. Again, her programs look stellar and we're chalking up the lower-than-usual difficulty and scores to the fact that it's a brand new season. At Nebelhorn Trophy, we saw her Japanese teammate and last season's rookie of the year, Hana Yoshida, take bronze and include triple axels in both her short and long programs. The short program saw a fall on that triple axel, but she scored a nice one in her excellent free skate. Pairs Division Nebelhorn Trophy provided us a fantastic early-season rematch between two of last season's top teams: World Champions Deanna Stelatto-Dudek & Maxime Deschamps and World Bronze medalists, Minerva Hase & Nikita Volodin. Canada's Deanna & Maxime had a great new short program, but drew the short straw, going first at the competition, so were scored lower than they would have been had they skated later. Their new free skate has incredible potential to be one of their best, but a few issues on their side-by-side jumps held them solidly in the silver medal position. Germany's Hase & Volodin looked nearly ready for Worlds in terms of their technical. After having to make some relatively last-minute changes to their music in the offseason, they looked poised and polished — claiming their first victory of this new season. If this represents the promise of what's to come, they will make it an exciting Grand Prix series and beyond. Also looking strong already are Sarah Conti & Niccolo Macii from Italy, who took the gold at Lombardia Trophy with two much better skates than most of what we saw from them last season. It was great to see a solid comeback for this team — and will hopefully lead to more positive outcomes. Reigning world silver medalists, Japan's Riku Miura & Ryuchi Kihara, had a stunning short program at Lombardia Trophy. However, their new long program—while looking like it has the potential to be a gem—saw a scary fall in a lift resulting in what appeared to be at least a minor injury for Ryuchi. Their coach, Bruno Marcotte, later said Ryuchi would recover in a week or two and that no major damage was done, thankfully. Also looking strong so far are Hungary's Maria Pavlova & Alexei Sviatchenko, who were third at Lombardia Trophy, and the U.S.'s Ellie Kam & Danny O'Shea, who had a stunning personal best short program at Nebelhorn Trophy and earned the bronze medal there. Men's Division We were NOT prepared for the Lombardia Trophy rematch of the two top men in the world from last season, Ilia Malinin and Yuma Kagiyama. In what had to be the most exciting moments of the season so far, these two men lit up the arena in Italy — showing programs that look both like they have tons of room to improve and also like they've been skating them for years. Reigning world champ and quad god, Ilia Malinin, has brought some of his exhibition style to his short program with an aggressive music choice and program. His free skate to “I'm Not a Vampire” may be his strongest yet. He did not perform the quad axel here or push to the limits he can on the technical side, but his victory was marked instead by more intensity and connection to the music than we've seen from him before. He's showing growth, and it's exciting to see. Yuma Kagiyama brought his finesse, his edges, his jumps, and his completeness as a skater to Lomabardia and looked absolutely divine as he took silver. As he did last season, his difficulty was somewhat dialed down here (its early season!), but that did not take away from the quality of his programs. In particular, his short program is a masterpiece that — once at its full power — will have audiences in the palm of his hand even more than they already are. A joy! Not too far behind the two of these men was Japan's Shun Sato, who looked better than ever after an off season where he obviously spent time working on his choreography and connection to the music. Worth noting is his score of 285.88, which got him a bronze here but would have taken gold over his countryman, Sota Yamamoto, at Nebelhorn Trophy. Yamamoto had a rough short program at Nebelhorn, but his exceptional free skate took him to victory. Right behind his was a strong performance from Gabriele Frangipani from Italy and an artistic dream from Deniss Vasiļjevs of Latvia. Ice Dance Division Of all the disciplines so far, we've seen the fewest of the top contenders in dance. Reigning world bronze medalists, Guignard & Fabbri of Italy, did make their debut with a fresh and fun rhythm dance. Their free skate seemed a bit less successful, but their scores were still magnificent and took them to an easy gold medal at Lombardia Trophy. The team nipping at their heels all last season, Lilah Fear & Lewis Gibson of Great Britain, looked amazing in their disco-themed rhythm dance. However, their free dance to a Beyonce medley seemed a little more safe than we've seen from them in past seasons. Still, their scores easily took them to victory at Nebelhorn Trophy and sets them up well as the season evolves. Our personal favorites so far this season have to be the two American teams of Christina Carreira & Anthony Ponomarenko, whose free dance to Carmen may well be their best to date, and Emilea Zingas & Vadym Kolesnik. Zingas & Kolesnik looked like a different team in their free dance, showing a new and captivating side to their skating. D., Schuman, E. M. Abstract: Brain function relies on communication via neuronal synapses. Neurons build and diversify synaptic contacts using different protein combinations that define the specificity, function and plasticity potential of synapses. More than a thousand proteins have been globally identified in both pre- and postsynaptic compartments, providing substantial potential for synaptic diversity. While there is ample evidence of diverse synaptic structures, states or functional properties, the diversity of the underlying individual synaptic proteomes remains largely unexplored. Here we used 7 different Cre-driver mouse lines crossed with a floxed mouse line in which the presynaptic terminals were fluorescently labeled (SypTOM) to identify the proteomes that underlie synaptic diversity. We combined microdissection of 5 different brain regions with fluorescent-activated synaptosome sorting to isolate and analyze using quantitative mass spectrometry 18 types of synapses and their underlying synaptic proteomes. We discovered ~1800 unique synapse-enriched proteins and allocated thousands of proteins to different types of synapses. We identify commonly shared synaptic protein modules and highlight the hotspots for proteome specialization. A protein-protein correlation network classifies proteins into modules and their association with synaptic traits reveals synaptic protein communities that correlate with either neurotransmitter glutamate or GABA. Finally, we reveal specializations and commonalities of the striatal dopaminergic proteome and outline the proteome diversity of synapses formed by parvalbumin, somatostatin and vasoactive intestinal peptide-expressing cortical interneuron subtypes, highlighting proteome signatures that relate to their functional properties. This study opens the door for molecular systems biology analysis of synapses and provides a framework to integrate proteomic information for synapse subtypes of interest with cellular or circuit-level experiments. Amen. Let's go for a walk. Let's go for a walk with Jesus. Jesus walks throughout the Gospels. Lent is the period of the Gospels when Jesus shares his ministry; as the story goes, Jesus walked for three years. Today, let us reflect on how we walk with Jesus. What does it mean to Walk? We all know how to walk... or so we think. There are different ways we walk...we walk towards, we walk away, we walk in circles. Our spiritual lives and Lent have all these manners of walking. We walk towards Easter, we know the path and the ending of Jesus's story. During Lent, I sense we try to rush ahead to Easter and not stay in the story. Yet the walk towards Easter is slow and deliberate. We come to know Jesus's ministry and, through each pause in the journey, to understand more deeply the person and power of Jesus. We walk away by sinning and ignoring Jesus. Have you ever tripped when you were walking? I have tripped many times, one fall resulted in a concussion. I was not in the walk, I was somewhere else, I crashed. In our spiritual life, when we are distracted or pay attention elsewhere, we sin, and sometimes we crash. Like having a concussion, our souls' awareness and the orientation of our soul are jumbled. We are not looking into our soul and so we trip into sins of both omission and commission. We walk in circles when we act as if we know where we are going in our spiritual lives and are honestly lost. Walking in our spiritual lives is like walking in the woods, a place of wonder and worry because we can see only so far ahead. In our spiritual walk in the woods, are we entering a dark place that fills us with worry or a light space that sparks our wonder? Sometimes we find ourselves overtaken by wonder or by worry. The wonder occurs when we are surprised by delight and the worry when unexpected concerns surface. This wonder/worry tension is also the reality of our spiritual life. When we focus on ourselves we are worried, when we focus on Jesus we feel awe and wonder. How are we With Jesus? Being with someone can ease the wonder/worry dilemma. We can share the wonder and lessen the worry. We are not alone. "With" can have many meanings - I want to focus on two - the act of accompaniment, and the difference between empathy and sympathy. As some of you may know, I work at Georgetown University, which is a Catholic and Jesuit institution. The Jesuit expression of charism or purpose has evocative language about being in relationship with God. The word "accompaniment" is often used. It means to be alongside someone in their journey, not to change the journey but to accept the journey. True accompaniment also means to not be in control. When we accompany someone, we give over to the experience - we do not use active verbs - direct, determine, guide, define. But how do we give over to accompaniment? We can understand accompaniment by the contrast between empathy and sympathy. Is our connection by knowing of the other person's circumstance? When we have empathy, we understand the circumstance of the other...but with some degree of distance and disengagement. Or is our connection by feeling the reality of the other person? When we have sympathy, we experience another's world. We identify with the person. Lent calls us into sympathy. We move in response to Jesus. Jesus breathes in, we breathe in... Jesus breathes out, we breathe out. We mirror Jesus's experience. We are not setting the path of the journey, no matter how many Lents we observe, no matter how often we read the Gospels. We are following in Jesus's steps. How do we come to know Jesus? Jesus is revealed to us as we walk with him through Lent. And we are revealed to him in turn by how we engage. We do not have a speaking part in the Gospels, yet we have a significant role in bringing the Gospels into this world. Jesus is both the path and the destination of Lent. As the path of Lent, Jesus's walk and encounters and lessons are demanding. Lent is not a stroll, it is a real honest journey. And as the destination, Jesus brings us to Christ, who gives us salvation. How do we encounter Jesus in the words of the Gospels, and in the actual lived Gospels of our lives? Every day, we Walk with Jesus...we encounter Jesus in the strangers we meet and particularly in the people we know most deeply. I will close with a story about accompaniment. In these weeks of Lent, I am mindful of accompanying both my 92-year-old mother-in-law and a work colleague. Each is dealing with physical diminishment, one from old age and one from a degenerative disease. Both are people who are "doers". If they could, each would be very active walkers, with a purpose, a quick pace, a destination, and a planned arrival time. Now they are moving slowly towards death. Jesus is walking with them. And I am accompanying them. My mother-in-law resents the help she needs, she is polite but very saddened. She was always the helper. My colleague is dealing with being rather than with doing. His work identity, and thus much of his own sense of self, has been anchored in getting things done and marking time by accomplishments. Now he says, he just "is". Both my mother-in-law and my colleague are sanctified people. They are in sympathy with Jesus. Their breaths of yearning and of sorrow mirror Jesus's breaths through Lent. I am honored to accompany them, to walk with them, to walk with Jesus. As we continue with the rigors of Lent, with the temptations, with the times of lonely solitude, with the moments of feeling misunderstood and slighted...all which Jesus experienced...know we are walking with Jesus and Jesus is walking with each of us. Step by step, stumble by stumble, blessing by blessing...we are walking towards salvation. My beloved friends, keep walking with Jesus, keep walking. Amen. Transcript: [Eugenie Dieck]: May the words of my mouth and the meditations of our hearts be acceptable in your sight O Lord, our strength and our Redeemer. Amen. [The Rev. Jarrett Kerbel]: Please be seated. [Eugenie Dieck]: Today is the tenth Sunday of Pentecost. Being deep into a liturgical season reminds me of being in school, in the middle of the semester and a bit bored with the content. Then, when writing a term paper, suddenly realizing the content had become very complex. It's time to pay attention. While writing this sermon, or honestly multiple drafts of this sermon, this student of the Pentecost course called “Building the Church” realized the content is quite challenging. My recognition of the complexity began with the first review of this Sunday's lessons. They confounded me. The lessons seem to be a jumble. The Old Testament is about David's lustful sinfulness, the Psalm is a robust confession, the Epistle is about how the Church is formed through its members working together, and the Gospel has Jesus saying “I am the bread of life.” Yes, a jumble of readings, unless one probes them for the lessons they contain. These readings lay out two tensions to be resolved by us to build God's Church. First Tension: I vs We. Second Tension: Want vs Need. Think about most of life's dilemmas. They can be reduced to addressing these tensions: I vs. we, want vs. need. Let's go back to the lessons to see how these tensions surface. In the Old Testament, we encounter David's horrible act of murdering Uriah and then bringing Uriah's widow to his house, to bear him a son. David was the king for a nation, he put himself before others, disregarding the leadership role given to him by God. David wanted this woman. He had multiple wives who had been given to him, and he did not need another wife. In the Psalm, we hear David's penitential confession and his request for forgiveness. Here, David is sublimating his ego to God. David is letting go of the “I.” What David wants is the same as what David needs: to be purged of his sins. The Epistle speaks to leading a life worthy of the calling to which one has been given. We have to read carefully about being called. We do not determine our call, God calls us. A calling is not what an individual wants. Instead, a calling is what the Church needs from the individual. Following these three readings, we hear the Gospel. We should, at this point, be primed for the nuances of John. This reading is a riddle about want vs need, about the individual's will vs God's will. The crowd wants signs from Jesus so badly they get into boats to look for Jesus. When they find Jesus, he is frustrated and responds to them, saying, essentially, “enough signs.” Jesus urges the crowd to do what is needed: to believe. As members of the Church, we are caught in the challenges of I vs. We and Needs vs. Want. Moving from “I Want” to “the Church Needs” is hard work; we must both discern and accept God's call. Here's how I learned the difference between “I want” and “the Church Needs.” When our son, Brendan, was in elementary school, he had significant learning differences. These differences made it very challenging for Brendan to be comfortable in groups. He could not follow multi-step directions or adapt to quick social interactions. Brendan refused to go to Sunday school, because it all moved too fast for him. He wanted me to go with him. However, when our sons were young, I really loved Adult Class. I got to be with adults, and talk about God, and not be with children. While I very much loved our sons, I really did not like groups of children. Guess what happened. I became a Sunday school teacher…for 8 years. That is a long time to do something that you do not want to do. Through the sad and tearful eyes of a 5 year old, God called me to be a Sunday school teacher. To be honest, I felt no differently about teaching Sunday school at the end of the 8 years, than I did when I started. I did not like it. The lesson I learned was that the Church was not about me. What mattered was that a little boy happily went to Sunday school, he belonged, and he grew in God's teachings. Brendan, now 30, is a generous, patient man who teaches middle schoolers with learning differences. Over the waning days of summer, let's take time to consider the coming year at St Martin's. Let's reflect on the tension between “I want” and “the Church needs.” In our careerist world, and our American sense of personal fulfillment, how are we called to do the work of the Church? Responding to God's call may require us to be uncomfortable and stay with the discomfort, even for 8 years. It is right for the 10th Sunday of Pentecost for us to resolve the tensions between “I want” and “the Church needs” to work hard at building the Church. Amen. Our sermon for the Day of Pentecost is given by Eugenie Dieck, a licensed lay preacher. She talks about daring to be wonderfully awkward as we go out and share the gifts we have received, particularly the gift of our faith. Today's readings are: Acts 2:1-21 Psalm 104:25-35, 37 Romans 8:22-27 John 15:26-27; 16:4b-15 Readings may be found on LectionaryPage.net for the Day of Pentecost, Year B Transcript: [Introductory music] [Eugenie Dieck] May the words of my mouth and the meditations of our hearts be acceptable in your sight, O Lord, our Rock and our Redeemer. (Psalm 19:14) Let's begin with a hypothetical moment. You have just been given a gift that grows if you share the gift with others. What will you do? My guess is that you will share the gift. However, sharing can be hard. We hope some of the gift will be left for us. And we have to trust that other people will want the gift. Finally, we have to maintain our generosity if the gift is refused. Today is Pentecost Sunday when the apostles, and all of us, are given the gift of the Holy Spirit. Pentecost Sunday, for all its drama of wind, and flames, and speaking in tongues, is really straightforward – Pentecost Sunday is about how the Holy Spirit enables us to share the gift of Jesus Christ with others. Our work, as the laity, is not primarily in this church building and on this campus. Our work is in the world. We come to St. Martin's to be together and to be renewed. We leave our church home to go out into the world to do the work God has given us to do. Yes, this work involves good works and our daily labors, but it also involves the work of proclaiming the message of Jesus Christ, our Lord and Savior. As Christians, we do our work in a world that can be indifferent, or even hostile, to the gift of Jesus Christ. Talking about our beliefs, and taking actions based on these beliefs, can be AWKWARD. The word “awkward” means to cause difficulty, to deal with something that is hard and may even be embarrassing. I assert that if we are truly evangelizing, we are intentionally being awkward. We are causing difficulty. Let's embrace the awkward, take joy in the awkward, because we are sharing, through the gift of the Holy Spirit, the gift of Jesus Christ. Here's how I live into the wonderfulness of the awkward...here's my evangelism story. I preach at St. Martin's, and I tell people outside of St. Martin's that I am a preacher. Both preaching and talking about preaching are evangelism. As you might guess, when I tell people that I preach many of the responses are a combination of confused looks, long silences, and earnest questions: Why do you preach? What do you talk about? Do sermons make a difference? Aren't you scared? Why do I preach? I preach to sing a song of praise to God. Preaching is my aria of belief. When I preach, I share God's love. The love I have for God and God has for me is real love – with sorrows and joys, failures and achievements. Most of all, this love is unconditional and constant because of the promise of salvation. I preach to encourage each of you to share God's love, to sing your song of praise. The pivot of Pentecost is that the Apostles were free to go into the world and declare the message of Jesus Christ. How do you declare? That is not a hypothetical question. I ask each of you “what is your declaration of faith?” Do others – your family, friends, acquaintances, and colleagues – know how much being a Christian, an Episcopalian, and a member of St. Martin's means to you? What do I talk about when I preach? Sometimes when I preach, I start softly and gently for tentative folks. I will talk about God. I talk about hope. I talk about ways to love others when it is hard to be loving. Other times, I share the whole massive gift -- I talk about Jesus and the Trinity, about salvation and the resurrection, about sin, repentance and forgiveness. My enthusiasm overtakes me, and I admit my enthusiasm may overwhelm others. I startle some people on purpose to create the AWKWARD. Not for me, but for God. When the apostles spoke in tongues, they were giving away their belief, they were sharing the gift and they were letting go…they were speaking as the Holy Spirit directed, in languages that could be understood by others. The apostles were proclaiming Jesus Christ and they were not in control – the apostles were experiencing the AWKWARD. I invite you into the AWKWARD – Are you willing to be awkward for Jesus Christ? Do sermons make a difference? That is not a question I can answer definitively. The difference a sermon makes doesn't necessarily happen on a given Sunday or in the same way for everyone. Some years ago, I gave a sermon that evoked extreme reactions – one person said they were deeply, profoundly blessed by the sermon and another person shared that the sermon filled them with rage. Confusing…perhaps at first, but not really when we consider how faith lives in each of us. I thanked each person for their honesty and suggested they weave their feelings of joy or rage back into their prayer life. The sermon I preached made a difference only because it prompted further conversations with God. The sermon is never about the sermon itself. A sermon is like the apostles speaking in tongues – one person experiences great joy, another person erupts with anger. Both of these emotions are real experiences of faith. The purpose of a sermon is to share faith with people; God will take it further. So, what language has the Holy Spirit given to you? To whom are you called to proclaim the gifts of faith? Am I scared when I preach? This is such a great question, as it asks, “what are you afraid of?” Are you afraid of what others may think? Are you afraid of failing? Are you just afraid? I am not afraid to preach. I feel full of courage and joy to talk about God, in God's house, surrounded by friends. Preaching is not scary. What is scary is to be a proclaiming Christian in a hostile world. Proclaiming, “I believe in Jesus Christ.” Proclaiming, “All that I am and all that I may become is because of the courage given to me by the Holy Spirit.” Proclaiming, “I am a breath of love in this world because God's full breath of creation brought me into existence.” Proclaiming is scary and exhilarating and awkward. Rational people do not say these things. Faithful people do, this faithful person does. Perhaps faith is not rational and that is part of the gift. Faith is transformative and transporting, which is wonderfully scary. What is scary for you? How do you proclaim your faith? Let's go back to the trick hypothetical question…what are we going to do with the gift that grows when we share with others? What ARE we going to do with our faith in Jesus Christ? Someone may have shown you the way or may be supporting your faith right now. Others need your faith, no matter how tentative or challenged you think your faith may be. Your faith might be a small light and not a huge flame, but your faith really matters. Your faith may be the gift to bring someone out of darkness. Today, my proclamation is emphatic. Your proclamation may be a gentle whisper. All proclamations are gifts worth sharing. At the end of today's service, we will leave this comforting place and encounter a world hostile to Jesus Christ. As a Christian, you may be afraid and feel awkward, I am afraid and awkward much of the time, so I trust God. I urge you to trust God, to have enough trust to be like the Apostles drunk with belief, and love, and hope, this morning and every morning. Let us go forth from this place to do the work God has given us to do. Proclaim the gift of Jesus Christ, share the gift of faith with abandon. Be awkward and be fearless. Abstract: Although significant efforts have allowed the identification of mRNAs in the processes of excitatory neurons, it is still unclear whether inhibitory neurons also localize a large population of mRNAs. In addition, the variability in the mRNA population within and between cells is not known. Here we developed a method for the transcriptomic characterization of a single neuron's subcellular compartments, combining laser capture micro dissection with scRNAseq. We separately profile the dendritic and somatic transcriptomes of individual rat hippocampal neurons and investigate the relation in mRNA abundances between the soma and dendrites of single glutamatergic and GABAergic neurons. We found that, like their excitatory counterparts, interneurons contain a rich repertoire of ~4000 mRNAs. Finally, using cell-type specific metabolic labelling of isolated neurites, we demonstrated that the processes not only of Glutamatergic but also of GABAergic neurons are capable of local translation, suggesting mRNA localization and local translation is a general property of neurons.
