Abstract: Objective: Macrophages play a central role in the onset and progression of vascular disease in pulmonary hypertension (PH) and cell-based immunotherapies aimed at treating vascular remodeling are lacking. This work evaluates the effect of pulmonary administration of macrophages modified to have an anti-inflammatory/pro-resolving phenotype in attenuating early pulmonary inflammation and progression of experimentally induced PH. Approach and Results: Mouse bone marrow derived macrophages (BMDMs) were polarized in vitro to a regulatory (M2reg) phenotype. M2reg profile and anti-inflammatory capacity were assessed in vitro upon lipopolysaccharide (LPS)/interferon-{gamma} (IFN{gamma}) restimulation, before their administration to 8- to 12- week-old mice. M2reg protective effect was tested at early (2 to 4 days) and late (4 weeks) time points during hypoxia (8.5% O2) exposure. Levels of inflammatory markers were quantified in alveolar macrophages and whole lung, while PH development was ascertained by right ventricular systolic pressure (RSVP) and right ventricular hypertrophy (RVH) measurements. Bronchoalveolar lavage (BAL) from M2reg-transplanted hypoxic mice was collected, and its inflammatory potential tested on naive BMDMs. M2reg macrophages demonstrated a stable anti-inflammatory phenotype upon a subsequent pro-inflammatory stimulus by maintaining the expression of specific anti-inflammatory markers (Tgf{beta}, Il10 and Cd206) and downregulating the induction of proinflammatory cytokines and surface molecules (Cd86, Il6 and Tnf). A single dose of M2regs attenuated the hypoxic monocytic recruitment and perivascular inflammation. Early hypoxic lung and alveolar macrophage inflammation leading to PH development was significantly reduced and, importantly, M2regs attenuated RVH, RVSP and vascular remodeling at 4 weeks post treatment. Conclusion: Adoptive transfer of M2regs halts the recruitment of monocytes and modifies the hypoxic lung microenvironment, potentially changing the immunoreactivity of recruited macrophages and restoring normal immune functionality of the lung. These findings provide new mechanistic insights on the diverse role of macrophage phenotype on lung vascular homeostasis that can be explored as novel therapeutic targets. Abstract: Inflammation in the central nervous system (CNS) can impair the function of neuronal mitochondria and contributes to axon degeneration in the common neuroinflammatory disease multiple sclerosis (MS). Here we combine cell type-specific mitochondrial proteomics with in vivo biosensor imaging to dissect how inflammation alters the molecular composition and functional capacity of neuronal mitochondria. We show that neuroinflammatory lesions in the mouse spinal cord cause widespread and persisting axonal ATP deficiency, which precedes mitochondrial oxidation and calcium overload. This axonal energy deficiency is associated with impaired electron transport chain function, but also an upstream imbalance of tricarboxylic acid (TCA) cycle enzymes. Among these, isocitrate dehydrogenase 3 (Idh3), a rate-limiting enzyme of the TCA cycle, is depleted in neuronal mitochondria in experimental models and in MS lesions. Notably, viral overexpression of an Idh3 subunit ameliorates the axonal energy deficits in neuroinflammatory lesions, suggesting that TCA cycle dysfunction in MS may be amendable to therapy. Abstract: Amyotrophic lateral sclerosis (ALS) is a fatal neuromuscular disease that affects all voluntary muscles in the body, leading to paralysis and respiratory failure, while the extraocular muscles (EOMs) are largely spared even at the end-stage of ALS. Through whole-mount muscle imaging, we detected severe denervation along with depletion of Pax7+ satellite cells (SCs) peri-neuromuscular junction (NMJ) in hindlimb and diaphragm muscles of end-stage SOD1G93A mice (a familial ALS mouse model), but not in EOMs. Upon isolating SCs from different muscles using fluorescence activated cell sorting (FACS), the FACS profiles of hindlimb and diaphragm SCs of G93A mice exhibited activation and depletion phenotypes but not in wildtype controls. Importantly, both wildtype and G93A EOM SCs exhibited spontaneous activation behavior without significant differences in abundance. Examination of Pax7+ and Ki67+ cell ratios and RNA-Seq of SCs cultured in growth and differentiation medium revealed that EOM SCs maintained renewability and stemness better than diaphragm and hindlimb counterparts, especially in differentiation-favoring environments. Comparative functional annotation analyses indicate enrichment of axon guidance molecules, such as Cxcl12, in cultured EOM SCs. In neuromuscular coculture experiments, overexpressing Cxcl12 in G93A hindlimb SC-derived myotubes enhanced motor neuron axon extension and improved innervation, partially replicating the multi-innervation property of EOM SC-derived myotubes. The unique SC homeostasis regulation and the production of axon guidance molecules including Cxcl12 may explain the ALS resistant nature of EOMs. Intriguingly, feeding G93A mice with sodium butyrate extended the life span of G93A mice, alleviated NMJ denervation and SCs depletion. Butyrate treatment promoted renewability and stemness of cultured G93A hindlimb and diaphragm SCs, as well as Cxcl12 expression. Thus, butyrate-induced EOM SC-like transcriptomic patterns may contribute to its beneficiary effects observed in G93A mice. Copy rights belong to original authors. Abstract: In Amyotrophic Lateral Sclerosis (ALS) motor neuron disease, mutations in proteins that upon stress localize within cytoplasmic protein inclusions called Stress Granules (SGs), are linked to the formation of aberrant inclusions, which are related to neuronal cell death. By combining studies in human cells and C. elegans including the use of Nanobodies, we found that inhibition of NEDP1, the enzyme responsible for the processing and deconjugation of the Ubiquitin-like molecule NEDD8 from substrates, promotes the elimination both of physiological and pathological SGs. The hyper-NEDDylation of Poly-(ADP-ribose) polymerase-1 enzyme upon NEDP1 inhibition compromises PAR production and is a key mechanism for the observed SG phenotype. Importantly, the above-described effects are related to improved cell survival in human cells, and in C. elegans, NEDP1 deletion ameliorates ALS-phenotypes related to animal motility. Our studies reveal NEDP1 as potential therapeutic target for ALS, based on the elimination of aberrant SGs. Copy rights belong to original authors. SIS talk is back : April is financial awareness month and also Sexual assault & Child Abuse Awareness month. We both believe that both topics are needed to improve our communities and individual families. We cannot heal or break generational curses without having these conversations. Nick Grono is an Australian (ex-pat) human rights campaigner who heads the Freedom Fund in London – the world's first private donor fund dedicated to ending slavery. Nick speaks with Michelle Michels about the work 'The Freedom Fund' is doing throughout modern slavery affected countries to assist in reducing the incidence of human trafficking into modern slavery. Nick is also the co-chair of the Jo Cox Foundation, and a board member of Girls Not Brides, the Global Partnership to End Child Marriage. Nick was the inaugural CEO of the Walk Free Foundation, a leading international actor in the fight against modern slavery. Prior to this, Nick was the Deputy President and COO of the International Crisis Group (ICG), the world’s leading conflict prevention NGO. Abstract: Diet is proposed to affect brain aging. However, the causality and mechanism of dietary effects on brain aging are still unclear due to the long time scales of aging. The nematode Caenorhabditis elegans (C. elegans) has led aging research because of its short lifespan and easy genetic manipulation. When fed the standard laboratory diet, Escherichia coli (E. coli), C. elegans experiences an age-dependent decline in temperature-food associative learning, called thermotaxis. To address if diet ameliorates this decline, we screened 35 different lactic acid bacteria as alternative diets. We found that Lactobacilli in a clade enriched with heterofermentative bacteria ameliorated age-dependent decline. On the other hand, homofermentative Lactobacillus species did not show this beneficial effect. Lactobacilli affected the thermotaxis of aged animals through DAF-16, an ortholog of mammalian FOXO transcription factor, while the effect on the thermotaxis was independent of the lifespan and locomotion. Our results demonstrate that diet can impact brain aging without changing the lifespan and that bacterial screen using C. elegans is a powerful approach to investigate age-dependent behavioral decline. Abstract: Epidemiological studies suggest a link between type-2 diabetes and Parkinsons disease (PD) risk. Treatment of type-2 diabetes with insulin sensitizing drugs lowers the risk of PD. We previously showed that the insulin sensitizing drug, MSDC-0160, ameliorates pathogenesis in some animal models of PD. MSDC-0160 reversibly binds the mitochondrial pyruvate carrier (MPC) protein complex, which has an anti-inflammatory effect and restores metabolic deficits. Since PD is characterized by the deposition of alpha-synuclein (aSyn), we hypothesized that inhibiting the MPC might directly inhibit aSyn aggregation in vivo in mammals. To answer if modulation of MPC can reduce the development of aSyn assemblies, and reduce neurodegeneration, we treated two chronic and progressive mouse models; a viral vector-based aSyn overexpressing model and a pre-formed fibril (PFF) aSyn seeding model with MSDC-0160. These two models present with distinct types of aSyn pathology but lack inflammatory or autophagy deficits. Contrary to our hypothesis, we found that a modulation of MPC in these models did not reduce the accumulation aSyn aggregates or mitigate neurotoxicity. Instead, MSDC-0160 changed the post-translational modification and aggregation features of the aSyn. Abstract: Troxerutin (TXR), a phytochemical reported to possess anti-inflammatory and hepato-protective effects. In this study, we aimed to exploit anti-arthritic properties of TXR using an adjuvant induced arthritic (AIA) rat model. AIA induced rats showed highest arthritis score at disease onset and by oral administration of TXR (50, 100, 200 mg/kg body weight), reduced to basal level in a dose dependent manner. Isobaric tag for relative and absolute quantitative (iTRAQ) proteomics tool was employed to identify deregulated joint homogenate proteins in AIA and TXR treated rats to decipher probable mechanism of the TXR action in arthritis. iTRAQ analysis identified a set of 434 joint homogenate proteins with 65 deregulated proteins (log2 case/control [≥]1.5) in AIA. Expressions of a set of important proteins (AAT, T-kininogen, vimentin, desmin, nucleophosmin) that could classify AIA from healthy were validated using Western blot analysis. Western blot data corroborated proteomics findings. In silico protein-protein interaction study of joint homogenate proteome revealed that complement component 9, the major building blocks of the membrane attack complex (MAC) responsible for sterile inflammation, gets perturbed in AIA. Our dosimetry study suggests that a TXR dose of 200 mg/kg body weight for 15 days is sufficient to bring the arthritis score to basal levels in AIA rats. We have shown the importance of TXR as an anti-arthritis agent in AIA model and after additional investigation its arthritis ameliorating properties could be exploited for clinical usability. Ameliorated is often, but not always, used in a medical context. When I was ill, I found that chicken soup ameliorated my cold. With every sip, I could feel my symptoms slowly fade away.

Yarn has played a significant part of life since then, as clothing and bedding, decoration, comfort, and art form. It's also traditionally a very female activity, in the western world, although I believe in South America knitting is one of those hard core macho things that men get together to do. Which goes to show that like most gender things it actually has more to do with culture than physicality.Our female ancestors then, for most of human history, were involved with yarn. A woman might be buried with her distaff even. Spinning, weaving, making and mending are traditional women's work. The methods we have, from the spindle and spinning wheels to knitting and crochet also connect us to the people who invented, developed and perfected them. Like so many of our ancestors, the yarn innovators are largely unknown, and there were probably a great many of them.Every woman who passes down this skill to a girl, is part of a huge web of weaving tradition. I learned knitting from my mother, crotchet from the mother of a boyfriend, spinning from several friends, although I'm not very good at it! I learned naalbinding (which might not be how you spell it!) to make traditional Viking socks, which take forever but are stunningly substantial. The wool work doesn't come alone though. Alongside it come the tales of other knitters and crafters, family myths and anecdotes. While the hands are busy, the mind has plenty of room to wander.I find wool work incredibly soothing. If I'm rattled, my mind in chaos and my body weary, then to sit down with wool is one of the best therapies. The rhythm of it is innately soothing. Watching small pieces of creativity form between my fingers is affirming, settling. The stories and inspiration of other wool workers are very much with me, and I feel part of something much bigger than I am. There's a power in weaving, knitting, spinning – the power to take the raw materials of nature and fashion them into something essential for life in a cold climate. Wool for our ancestors must have contributed to survival, a vital part of culture that made human life viable in tough locations.I heard a story once, that Arran sweaters have complex designs because each family had its own pattern. The women made them so that if a drowned sailor washed ashore, they could figure out who he was, who his people were. I can't imagine making a jumper for my man that would mark him as mine in case he died, that must take a certain kind of pragmatic courage.Modern life is full of things that claim to be convenient, but that take far more from us than they give. I know a lot of women who are returning to wool, or learning it anew, because it makes sense to them on an emotional level as well as a practical one. Being able to do the essential things of life matters and in turn makes your life feel more real. Ancestral pursuits like this bring a sense of stability, rootedness and belonging. We need that. In cultures full of uprooted people, in times where continuity and certainty are hard to come by, reaching back into the past makes a lot of sense. The ancestors are all there, behind us. A sense of belonging is a precious thing to have when everything else feels a bit fragile and uncertain. Between climate change and the world economies, and the insane priorities of politicians, life for any aware person is not comfortable, even in our relatively insulated western world.The wool tribe is there, for anyone willing to pick up the threads and find out how to make them. Your grandmothers will have done it, and your great grandmothers. The odds are you had some distant grandfathers who kept or sheared sheep, too. Go back far enough and most of us have that, in fact. The threads of wool can also be threads of continuity, safety lines to remind us of where we came from and how we connect to the other creatures, and to the earth.
