With its interactor USP28, it is part of the mitotic surveillance pathway (MSP), a sensor that monitors the duration of cell division, promoting p53-dependent cell cycle arrest when a critical time threshold is surpassed. 53BP1 dynamically associates with kinetochores, being recruited during prophase, and then undergoing a time-dependent loss of affinity. However, the relevance of this behaviour remains unclear. Here, we identify CENP-F as an interaction partner and kinetochore receptor for 53BP1. By engineering human cells with a CENP-F point mutation, we demonstrate that preventing 53BP1 kinetochore localization does not reduce MSP proficiency. Strikingly, however, preventing the loss of 53BP1 from the kinetochore by inhibiting Polo-like kinase 1 (PLK1) restrains MSP activity, a phenomenon that is abrogated in the CENP-F mutant condition. Taken together, we demonstrate that kinetochore-loaded 53BP1 represents an MSP functionally inhibited state and that PLK1-dependent re-localization of 53BP1 represents an important layer of MSP regulation. Il titolo della decima puntata della terza stagione di J-TACTICS, trae spunto da: “Chi ha paura delle streghe”, (The Witches) che è un film del 1990 diretto da Nicolas Roeg e tratto dal romanzo per bambini Le streghe di Roald Dahl.Durante le vacanze estive in Norvegia con la famiglia, il piccolo Luke Eveshim ascolta le storie che nonna Helga gli racconta sulle vere streghe, raccomandandogli di fare molta attenzione, perché esse sono creature magiche e diaboliche che odiano i bambini e cercano di eliminarne il maggior numero possibile.Le streghe sembrano donne qualsiasi e vivono mimetizzate tra gli esseri umani, ma possono essere riconosciute da alcuni particolari: lunghi artigli per i quali indossano sempre i guanti, piedi senza dita che nascondono in scarpe larghe, assenza completa di capelli per cui mettono la parrucca e occhi sfumati di rossastro; inoltre sono dotate di un olfatto particolarmente sviluppato che consente loro di percepire l’odore di bambino, che per loro è nauseante.Facendo la nostra solita trasposizione dalla cinematografia al mondo del calcio, ed in modo particolare alle vicende juventine, potremo utilizzare il titolo e le vicende narrate nella pellicola per analizzare la partita giocata nel tardo pomeriggio di sabato al Ciro Vigorito di Benevento tra la squadra di casa e la Juventus.Una partita che, a detta di mister Pirlo, avrebbe dovuto sancire la fine degli esperimenti di formazione e ci avrebbe permesso di ammirare una squadra più sicura, determinata e convincente anche per dare continuità alla buona prova con il Cagliari in casa e dopo le incertezze manifestate qualche giorno prima con gli ungheresi del Ferencváros in Champions League.Tutte buone idee ed intenzioni che purtroppo non si sono minimamente palesate in quel di Benevento.I bianconeri danno vita infatti, ad una prova pressoché sconcertante, dopo un buon avvio culminato con il vantaggio del solito Alvaro Morata in versione bomber e re Mida con un gol ad ogni pallone giocabile che gli viene fornito.Lo spagnolo segna ancora, l’ottavo gol (con tre assist) che valgono il 48% dei gol totali della Juventus realizzati in questa stagione, Alvaro verrà poi espulso nel finale dopo il triplice fischio.Non basta il solito Morata, senza Cristiano Ronaldo (a riposo concordato e neanche partito per Benevento) la Juventus viene fermata sul pari da una bella partita della squadra di Inzaghi.Alla sua rete risponde Letizia con un gran destro al volo, 1-1 il risultato finale che lascia l’amaro in bocca, molti dubbi sulla formazione, sulla prestazione di alcuni singoli e una forte dose di arrabbiatura per i supporters bianconeri i quali vedono per l’ennesima volta i propri beniamini buttare via punti preziosi con squadre di caratura modesta se non addirittura mediocre, oramai una costante di questo avvio di stagione.Match da dimenticare per il giovane Frabotta, per il brasiliano Arthur il quale non riesce mai ad incidere e a proporre i cambi di passo che tanto necessari sarebbero a questa Juve, Ramsey autore di una prova soporifera ed incolore, non a caso sostituito al ’64 da Kulusevski ma autore anch’esso di una prova negativa in cui è apparso abulico, poco concentrato e fuori contesto.Mister Pirlo tuttavia nelle interviste post partita ha avuto parole di conforto per l’ex gialloblu: «E’ giovane, non può fare tutte le partite. Gioca in Nazionale, alla Juve, viene dal Parma, e il peso della palla è diverso. Impossibile farlo giocare sempre, anche lui è stanco. Without a sexually informed model, it is easy for therapists to pathologize sexual behaviors and not provide effective treatment. The term ‘sex addiction’ is thrown around like confetti, and that needs to stop! “We need to walk through these moments of being ‘ping-ed’ by your desire,” says Michael, and not stand in judgement of one’s sexual problematic behavior.

IM David Vigorito is a Massachusetts-based chess player, author and trainer. As a player, David been the state champion of Massachusetts, New Hampshire and Nevada. He has also competed in the U.S. Championship and has been among the top 50 player in the US. David and his publisher, Quality Chess, have just released his 9th book, Playing the Najdorf. Welcome to episode 138 of the Sexology Podcast! Today I welcome Michael Vigorito to the podcast. In this episode he speaks to me about out of control sexual behaviour, examining the driving force behind it and understanding the best practices to deal with this issue. Mr. Vigorito is a sex therapist, author and consultant. As a psychotherapist, he provides individual, couples and group psychotherapy for a range of sexual and behavioral health concerns. He developed a sexual health assessment and treatment protocol with Douglas Braun-Harvey, which was published in Treating Out of Control Sexual Behavior - Rethinking Sex Addiction (2016) (SSTAR 2018 Health Professional Book Award Winner). As a sexual health consultant for Behavioral Health Systems Baltimore and University of Maryland - Prevention Research Center, Mr. Vigorito designed and implemented capacity-building initiatives to integrate sexual health into mental health and substance use disorder treatment. These programs offered trainings and technical assistance in conducting routine sexual health conversations, sexual risk-reduction counseling and LGBTQ cultural competency within behavioural health systems of care. Laura Adams is the President and CEO of the Rhode Island Quality Institute (RIQI), a center of collaborative innovation that advances health and healthcare transformation. RIQI is the only organization in the nation to win all three of the major HITECH health IT grants which funded CurrentCare (the statewide health information exchange), the RI Regional Extension Center and the RI Beacon Community, bringing in $27M in funding. She was recently appointed to the Governor's Workgroup on Healthcare Innovation. Laura served on the ONC's HIT Policy Committee's Governance Panel for the Nationwide Health Information Network. She is a member of the Health Information Management and Systems Society (HIMSS) Center for Patient and Family Centered Care Advisory Group and chaired the Institute of Medicine's (IOM) Planning Committee for the “Digital Infrastructure for Population Health and a Learning Healthcare System” workshop series. Laura led the governance consulting for the Improving Performance in Practice (IPIP) Initiative, a RWJF-funded collaboration among the American Board of Medical Specialties, and the Boards and Societies of Internal Medicine, Family Physicians and Pediatrics. She traveled in the U.S. and Europe with W. Edwards Deming in the study of statistical-based quality improvement. She was Founder, President and CEO of Decision Support Systems, a New York-based company specializing in Internet-based healthcare decision support. Laura has been a faculty member of the Institute for Healthcare Improvement (IHI) in Boston since its inception. She directed the IHI Idealized Design of the Intensive Care Unit project and served as faculty in the VHA's ICU improvement collaborative. She was among the first to bring the principles of healthcare QI to the Middle East, in conjunction with Donald Berwick, MD and the Harvard Institute for Social and Economic Policy in the Middle East. She served as IHI faculty at the inaugural IHI Middle East Forum on Quality Improvement in Healthcare in Doha, Qatar in 2013. Her publications include co-authoring with Gustafson, et al, Developing & Testing a Model to Predict Outcomes of Organizational Change, Health Services Research, 38(2), 751-776 and co-authoring “Collaborating with Consumers to Advance Health Knowledge and Improve Practice” Herzlinger, R, Consumer-Driven Health Care: Implications for Providers, Payers and Policymakers, 2004. She also co-authored “A Draft Framework for Measuring Progress Towards the Development of a National Health Information Infrastructure, BMC Medical Informatics Decision Making, June, 2005. Laura co-authored “Improving comfort and communication in the ICU: a practical new tool for palliative care performance measurement and feedback”, Nelson, J., Mulkerin, C., Adams, L. and Pronovost, P.; Quality and Safety in Health Care (QSHC), Aug 2006. She co-authored with Peter Pronovost, MD, et.al. in The Rhode Island ICU Collaborative: A Model for Reducing CLABSI and Ventilator-associated Pneumonia Statewide, QSHC, 2010;19:555-561. She also co-authored publication with M. Vigorito and B. Sexton entitled Collaborative Improvement in Sepsis Identification & Treatment, JQPS, Vol. 37, No. 11, Nov. 2011. RIQI won the 2013 National Council for Community Behavioral Health Excellence Award for Impact in serving those with behavioral health and substance abuse challenges. Laura has received citations from the RI Congressional delegation for her visionary leadership and contributions to improving the healthcare system in Rhode Island.
