N., Song, Y., Davies-Jenkins, C. W., Gong, T., Wang, G., Oeltzschner, G., Edden, R. A. E. Abstract: Purpose: To investigate the age-dependence of metabolite T1 relaxation times at 3T in both gray- and white-matter-rich voxels. Methods: This manuscript analyzes publicly available metabolite and metabolite-nulled (single inversion recovery TI = 600 ms) spectra acquired at 3T using PRESS localization. Voxels were placed in posterior cingulate cortex and centrum semiovale in 102 healthy volunteers across 5 decades of life (20s to 60s). All spectra were analyzed in Osprey v2.4.0. To estimate T1 relaxation times for tNAA2.0 and tCr3.0, the ratio of modeled metabolite residual amplitudes in the metabolite-nulled spectrum to the full metabolite signal was calculated using the single inversion recovery signal equation. Correlations between T1 and subject age were evaluated. Results: Spearman correlations revealed that estimated T1 relaxation times of tNAA2.0 (rs = -0.43; p less than 0.001) and tCr3.0 (rs = -0.23; p = 0.021) decreased significantly with age in white-matter-rich CSO, and less steeply (and not significantly) for tNAA2.0 (rs = -0.15; p = 0.136) and tCr3.0 (rs = -0.10; p = 0.319) in gray-matter-rich PCC. Conclusion: The analysis harnessed a large publicly available cross-sectional dataset to test an important hypothesis, that metabolite T1 relaxation times change with age. This preliminary study stresses the importance of further work to measure age-normed metabolite T1 relaxation times for accurate quantification of metabolite levels in studies of aging. Link to bioRxiv paper: http://biorxiv.org/cgi/content/short/2022.11.11.516194v1?rss=1 Authors: Gong, J., Stickland, R. C., Bright, M. G. Abstract: The blood flow response to a vasoactive stimulus demonstrates regional heterogeneity across both the healthy brain and in cerebrovascular pathology. The timing of a regional hemodynamic response is emerging as an important biomarker of cerebrovascular dysfunction, as well as a methodological confound within fMRI analyses. Previous research demonstrated that hemodynamic timing is more robustly characterized when a larger systemic vascular response is evoked by a breathing challenge, compared to when only spontaneous fluctuations in vascular physiology are present (i.e., in resting-state data). However, it is not clear whether hemodynamic delays in these two conditions are physiologically interchangeable, and how methodological signal-to-noise factors may limit their agreement. To address this, we generated whole-brain maps of hemodynamic delays in nine healthy adults. We assessed the agreement of voxelwise gray matter (GM) hemodynamic delays between two conditions: resting-state and breath-holding. We found that delay values demonstrated poor agreement when considering all GM voxels, but increasingly greater agreement when limiting analyses to voxels showing strong correlation with the GM mean time-series. Voxels showing the strongest agreement with the GM mean time-series were primarily located near large venous vessels, however these voxels explain some, but not all, of the observed agreement in timing. Increasing the degree of spatial smoothing of the fMRI data enhanced the correlation between individual voxel time-series and the GM mean time-series and did not substantially change the agreement in hemodynamic timing as a function of this relationship strength. These results suggest that signal-to-noise factors may be limiting the accuracy of voxelwise timing estimates and hence their agreement between the two data segments. In conclusion, caution must be taken when using voxelwise delay estimates from resting-state and breathing-task data interchangeably, and additional work is needed to evaluate their relative sensitivity and specificity to aspects of vascular physiology and pathology.

Will the Bitcoin price reach $5,000 or $10,000 before it drops and bring the cryptocurrency market capitalization to over $100 billion? Are altcoins starting to sell off into BTC? Would you watch this video to hear my opinions based on the data I see? The Bitcoin price reached a record high of over $2,700 today May 25, 2017! Meanwhile, most of the alternative cryptocurrencies dropped anywhere from 5% to 15% in the last 24 hours which I think is signaling the ceiling for the altcoin bubble. SELL is my recommendation for nearly all the altcoins at this point with holding Bitcoin providing risk of a drop and a potential reward of doubling or tripling itself in value as the altcoins sell. That said, I was totally wrong about selling Bitcoin before at $1700 and about selling my Dash masternode at $87 when the price was $150 yesterday. My own advice allowed me to miss out on earning another $50,000+ in profit WHICH I AM OKAY WITH because there is always more money to be made. Trading crypto can be an addiction because of the constant need to get gratification from making more money and the huge hangover when the high wears off into fear resulting in desperate selling measures! Learning when to sell and accepting missing out on potential gains in exchange for peace of mind has been a big challenge for me. I am grateful for the $75,000 I did make from buying Dash at $10 to $11 originally. Selling my Dash allowed me to discover another cryptocurrency which appears to not have been fully carried away by the bubble. I am ALL IN on the crypt I share in my video class at http://u.jerrybanfield.com/courses/allincrypto. The cryptocurrenty I share that I am buying in the course is still an amazing buy and I will give you a little more time to buy it while it is low before I start promoting it relentlessly which I think will help to bring the price up 10x within the next 10 months. For cashing out, I think now is the perfect time to put Bitcoin into the altcoin I have invested in before the speculators pump it up! If the price keeps dropping on the alt coin I am excited about, I am going to keep buying and buying anyway because I believe in the community! (raf)
