Do you ever consider going to Hilton Head Island, SC for a fresh-out-of-the-oven French baguette or a raspberry tart that's perhaps gilded with gold flakes and filled with lemon curd? Maybe not, but you might want to reconsider because Hilton Head Social Bakery, with two locations on the island, has been baking that and much more since Chef Philippe Feret and his wife Marissa opened the bakery in 2016. Phillipe was an apprentice in his father's Paris bakery from the age of five, and he soon made restaurants his life, from the famous Parisian restaurant Taillevent, to New York City where he was enlisted to reopen Windows on the World in 1996. He subsequently served as executive chef at Tavern on the Green, The Regency Hotel, Cafe Centro and eventually owned his own restaurant and catering for 15 years. The bakery and his life on Hilton Head seems his most joyous chapter yet, using his father's croissant recipe, and also letting his creativity, enthusiasm for new flavor combinations, classic recipes, and the artistry of sugar work fill the pastry cases daily. He's embraced the island life and given up his chef's coat for shorts and t-shirts in the kitchen, but he admits he still dons it for special occasions.

I asked Tom to visit with us talk about the new stucco drainage system developed by Tamlyn. Having dealt with this issue for many years I was thrilled to see a system in place that finally addressed the common issues with this very good but volatile finish.
There is something about a design that can draw the eye. We often can't explain it, but we're drawn to an aesthetic or a certain item's look. For Tom Feret of Tamlyn and Cale Kids of Woodtone, the companies' partnership has done just that, with a Woodtone panel perfectly trimmed by a Tamlyn product at a recent trade show in Las Vegas turning plenty of heads. “(The panel) really pops in color. We wanted to promote that. We didn't want our trim in this case to overtake and be dominant. So, we chose a shadow effect using black anodized, which is very robust as far as longevity,” said Feret, the Sales Director at Tamlyn. “So, it actually created a shadow around that panel to let that panel really pop and sizzle. “It drew everybody's attention really well. It came off looking fantastic.” Of course, most trade shows currently are on hold because of the coronavirus pandemic, and both Feret and Kids say they've had to make some modifications to how they do business. Some of those effects, however, have had surprising positive benefits, such as taking the number of architects they can meet with from one or two at a time to 60 or 70 with online meetings. “It's that we're still able and have not felt the COVID effect like lots of industries,” said Kids, the sales manager for the state of Texas for Woodtone. “We believe the market is changing. Who would've thought a guy my age would've ever been able to get on a Zoom call?” But that's exactly what he and many others are doing, with the partnership creating showstopping materials continuing strong despite a pandemic.

This week on In the Drink, host Joe Campanale is joined in the studio by Thierry Valette of the Clos Puy Arnaud vineyard. Clos Puy Arnaud is mentioned in the Feret book (the bible of Bordeaux's Wines) in 1874 as a first growth. Thierry Valette has been producing here since 2000. The vineyard is run with the principles of an organic approach, and has been involved in biodynamic agriculture since 2004.
Background: Duchenne Muscular Dystrophy is an inherited degenerative neuromuscular disease characterised by rapidly progressive muscle weakness. Currently, curative treatment is not available. Approaches for new treatments that improve muscle strength and quality of life depend on preclinical testing in animal models. The mdx mouse model is the most frequently used animal model for preclinical studies in muscular dystrophy research. Standardised pathology-relevant parameters of dystrophic muscle in mdx mice for histological analysis have been developed in international, collaborative efforts, but automation has not been accessible to most research groups. A standardised and mainly automated quantitative assessment of histopathological parameters in the mdx mouse model is desirable to allow an objective comparison between laboratories. Methods: Immunological and histochemical reactions were used to obtain a double staining for fast and slow myosin. Additionally, fluorescence staining of the myofibre membranes allows defining the minimal Feret's diameter. The staining of myonuclei with the fluorescence dye bisbenzimide H was utilised to identify nuclei located internally within myofibres. Relevant structures were extracted from the image as single objects and assigned to different object classes using web-based image analysis (MyoScan). Quantitative and morphometric data were analysed, e. g. the number of nuclei per fibre and minimal Feret's diameter in 6 month old wild-type C57BL/10 mice and mdx mice. Results: In the current version of the module "MyoScan", essential parameters for histologic analysis of muscle sections were implemented including the minimal Feret’s diameter of the myofibres and the automated calculation of the percentage of internally nucleated myofibres. Morphometric data obtained in the present study were in good agreement with previously reported data in the literature and with data obtained from manual analysis. Conclusions: A standardised and mainly automated quantitative assessment of histopathological parameters in the mdx mouse model is now available. Automated analysis of histological parameters is more rapid and less time-consuming. Moreover, results are unbiased and more reliable. Efficacy of therapeutic interventions, e.g. within the scope of a drug screening or therapeutic exon skipping, can be monitored. The automatic analysis system MyoScan used in this study is not limited exclusively to dystrophin-deficient mice but also represents a useful tool for applications in the research of other dystrophic pathologies, various other skeletal muscle diseases and degenerative neuromuscular disorders.
