BUFFALO, NY- December 27, 2023 – A new #research paper was #published in Oncotarget's Volume 14 on December 20, 2023, entitled, “The pharmacodynamic and mechanistic foundation for the antineoplastic effects of GFH009, a potent and highly selective CDK9 inhibitor for the treatment of hematologic malignancies.” To evade cell cycle controls, malignant cells rely upon rapid expression of select proteins to mitigate pro-apoptotic signals resulting from damage caused by both cancer treatments and unchecked over-proliferation. Cyclin-dependent kinase 9 (CDK9)-dependent signaling induces transcription of downstream oncogenes promoting tumor growth, especially in hyperproliferative ‘oncogene-addicted' cancers, such as human hematological malignancies (HHMs). In this new study, researchers Fusheng Zhou, Lili Tang, Siyuan Le, Mei Ge, Dragan Cicic, Fubo Xie, Jinmin Ren, Jiong Lan, and Qiang Lu from GenFleet Therapeutics Inc. and Sellas Life Sciences Group aimed to summarize current knowledge underlying the mechanism of action (MOA) of GFH009 and explain its robust anti-cancer activity. “Understanding GFH009's MOA allows for a more optimal clinical development path, given the potential for meaningful benefits in patients with hematological malignancies.” GFH009, a potent, highly selective CDK9 small molecule inhibitor, demonstrated antiproliferative activity in assorted HHM-derived cell lines, inducing apoptosis at IC50 values below 0.2 μM in 7/10 lines tested. GFH009 inhibited tumor growth at all doses compared to controls and induced apoptosis in a dose-dependent manner. Twice-weekly injections of GFH009 maleate at 10 mg/kg significantly prolonged the survival of MV-4-11 xenograft-bearing rodents, while their body weight remained stable. There was marked reduction of MCL-1 and c-MYC protein expression post-drug exposure both in vitro and in vivo. Through rapid ‘on-off' CDK9 inhibition, GFH009 exerts a proapoptotic effect on HHM preclinical models triggered by dynamic deprivation of crucial cell survival signals. “Our results mechanistically establish CDK9 as a targetable vulnerability in assorted HHMs and, along with the previously shown superior class kinome selectivity of GFH009 vs other CDK9 inhibitors, strongly support the rationale for currently ongoing clinical studies with this agent in acute myeloid leukemia and other HHMs.” DOI - https://doi.org/10.18632/oncotarget.28543 Correspondence to - Jiong Lan - jlan@genfleet.com Sign up for free Altmetric alerts about this article - https://oncotarget.altmetric.com/details/email_updates?id=10.18632%2Foncotarget.28543 Subscribe for free publication alerts from Oncotarget - https://www.oncotarget.com/subscribe/ Keywords - cancer, GFH009, CDK9, leukemia, cell cycle About Oncotarget Oncotarget (a primarily oncology-focused, peer-reviewed, open access journal) aims to maximize research impact through insightful peer-review; eliminate borders between specialties by linking different fields of oncology, cancer research and biomedical sciences; and foster application of basic and clinical science. In honor of Hispanic Heritage Month, we're revisiting some of our favorite episodes! To close out HHM, here's How the Garcia Girls Lost Their Accents by Julia Alvarez. If you missed this episode the first time around, it's back with some bonus recommendations for readalikes. This episode we dive into Julia Alvarez's How the Garcia Girls Lost Their Accents, the 1991 reverse chronology story of four sisters who flee to the United States from the Trujillo regime in the Dominican Republic. As we look through Yolanda's eyes at the world of New York City and the Dominican Republic in the 60s, 70s, and 80s, we dissect Alvarez's depiction of class, race, acculturation, and machismo. We delve into the dynamics of sisterhood, boyfriends with ludicrous names, the ubiquity of certain aspects of adolescence, and complicated families. We also learn that some of us are Sandies but some of us are Lauras. Finally, we are taking it to the streets, er... stacks, with our new Person in the Stacks segment and asking what tastes like home. The eCW Podcast speaks with Geli King-Brown, senior director of quality management at Healing Hands Ministries (HHM), about how her organization is expanding its locations and services to extend healthcare to those who can't afford it. Geli discusses how the Patient-Centered Medical Home (PCMH) program has helped the organization standardize the care it provides by establishing policies, procedures, and workflows. By using eClinicalWorks® PCMH and HEDIS analytics, HHM can identify subgroups of their patient population with specific conditions or diseases and see the quality improvement process around them. With the embedded reports, Geli and her team can efficiently track and monitor their progress, share those results with their patients, and highlight healthcare champions at work. The game-changing result is that Geli and her team can identify what works and what doesn't while continuously striving to improve their care.
In this episode, we celebrate Hispanic Heritage Month (HHM) by gaining rich perspectives from our guests, Carolina Betancourt and Guillermo Guereque. Carolina was born in Venezuela and Guillermo in Mexico, and they have lived in the U.S. for many years. This conversation demonstrates the diversity of the Latin community and why HHM is so important to them, their community, and our country. LLR's podcast hiatus during every September is due to their involvement with Hell's Half Mile Film & Music Festival. Jay was able to introduce Party Nails to his hometown audience (her album You Don't Have To Come Home But You Can't Stay Here was his #2 record of 2020, which was covered in episode 12), while Deon spent his evenings creating a space suitable for a truly unique live music experience. It was one helluva weekend. After the rock-n-roll hangover dissipates, the Lightnin' Lickers will return with Episode 25, where they discuss their favorite records from bands filed under the letter E. Stay tuned! ___ HHM weekend's peak of music events for our 16th annual festival took place on Saturday, September 24th at the Dunlop Building (located at 517 Washington Ave., Bay City, MI), and per usual brought 3 amazing bands! This year's lineup featured Party Nails (Los Angeles), Ratboys (Chicago), and Sugar Tradition (Detroit), who recently opened for Jack White on his Supply Chain Issues Tour. As a festival, we were thrilled with the opportunity to use the Dunlop Building and transform it into a HHM-style venue for one evening, and bring 3 very different bands together to make it's mark on "Midtown Bay City"! It's Hell's Half Mile's sixteenth birthday, and we celebrated the only way we know how. The Art Department of Bay City provided stunning custom neon and unique art installations in the bar/lounge (former showroom) area, while Lightnin' Licks Vinyl Preservation Society provided venue layout, logistics, design and decorations (service garage). Dunlop Building owner Mike Beattie allowed HHM volunteers to showcase the history of the building, utilizing many promotional materials from the business' glory days as a Pontiac dealership and service center. Dunlop Pontiac was regularly voted as the #1 auto dealership in the Greater Bay region, selling both new and used vehicles to the community. Most longtime residents of Bay City have a connection to the building due to the once sizable workforce at nearby GM Powertrain, allowing family and friends to purchase their cars from the lot with GM discounts. It was important to HHM and Lightnin' Licks Vinyl Preservation Society that the aesthetics of the venue reflected the history of the building, marrying the city's auto manufacturing past with it's cultural present, as any space where working people spent their time away from their families deserves to be treated with respect. Music included in the video is that of Party Nails, Ratboys, and Sugar Tradition. Not streaming on Spotify? #Join Jamie & Simon as they Rewind cult TV Show Better Call Saul. This week we deep dive S4 E1 Smoke and S4 E2 Breathe and explore the show's symbolism, symmetry and plot twists From Wikipedia: S4 E1 "Smoke" In a flash forward, following his collapse while working at the Omaha Cinnabon, a nervous "Gene" has a worrying encounter with a hospital clerk who needs his personal information for billing purposes. During his taxi ride to the mall to retrieve his car, Gene becomes suspicious of the driver, whose cab displays an Albuquerque Isotopes air freshener. He exits before reaching the mall and walks around the corner out of the driver's sight. In 2003, Jimmy and Kim learn of Chuck's death, and believing himself responsible, Jimmy falls into depression. Howard arranges Chuck's funeral, and later admits that he believes his decision to dismiss Chuck from HHM caused Chuck's suicide. Howard explains that Chuck's malpractice insurance rates had become exorbitant – not realizing Jimmy was to blame for reporting Chuck's instability to the insurers. Jimmy allows Howard to take the blame and quickly regains his happy-go-lucky demeanor. Mike performs actual security and safety consulting for Madrigal Electromotive and details a list of violations he uncovered on his first site visit. Following Hector's stroke, the cartel bosses direct Nacho and Arturo to take over Hector's drug operation. Gus remains suspicious of Nacho and has Victor follow him. S4 E2 "Breathe" With his law license suspended, Jimmy interviews for a job as a salesman at Neff Copiers and spots an opportunity to steal a valuable Hummel figurine. Rebecca and Kim meet with Howard to review the disposition of Chuck's estate. Kim is unaware of Jimmy's role in the events which led to Chuck's death and has an explosive confrontation with Howard in which she lambastes him for treating Jimmy poorly ever since. Lydia tells Mike that his security consulting is a paper transaction to facilitate the laundering of the money he stole from the Salamancas. Mike insists on continuing actual inspections, arguing they provide a plausible explanation in case anyone looks into the payments. Gus discreetly pays a skilled doctor to help Hector recover from his stroke and learns Nacho was responsible for it. When Arturo and Nacho come to his facility to acquire more drugs, Arturo strong arms his way into taking more than he is supposed to. He brags about his success as they leave, but Gus ambushes and kills him. Gus then informs Nacho he knows Nacho attempted to murder Hector, but the Salamancas do not, and because of this knowledge, Nacho is now under Gus's control.
Join Simon & Jamie as they deep dive S5 E5 "Dedicado a Max" and S5 E6 "Wexler v. Goodman" and explore the show's symbolism, symmetry and plot twists From Wikipedia: S5 E5 "Dedicado a Max" Mike wakes up on a ranch in Mexico which has ties to Gus, including a fountain dedicated to Max. Dr. Barry Goodman treats him and recommends he stay and heal. Gus later arrives and asks for Mike's help in Gus' war against the Salamancas. Mike refuses to engage in killing simply to further Gus' aims, but Gus says he wants Mike because Mike understands Gus' need for revenge. Jimmy as Saul creates delays in Mesa Verde's eviction of Acker. Kim tries to recuse herself by claiming a conflict of interest, but Kevin insists she remain and refuses any compromises. Howard asks Jimmy to accept his offer to join HHM, and Jimmy claims he is still considering it. Jimmy suggests there are no options left for Acker but to find "dirt" on Kevin and tries to persuade Kim to move on, but she decides to proceed. Jimmy hires Sobchak who thinks his search of Kevin's home revealed nothing damaging, but Kim's knowing smile as she looks through Sobchak's photos indicates she notices something useful. Rich Schweikart suggests that Kim should disengage from Mesa Verde's business, correctly deducing her heart is not in it, but she angrily refuses. S5 E6 "Wexler v. Goodman" Kim tells Jimmy she does not want to follow through on blackmailing Kevin and wants to reach a settlement between Mesa Verde and Everett Acker. Jimmy agrees, but at the meeting to finalize the settlement, he stuns everyone by asking for $4 million. Kim's insight into Sobchak's photos is that Mesa Verde's logo was inspired by a photograph the bank never obtained permission to use. Jimmy pressures Kevin by threatening an injunction against displaying the logo and threatening to run TV ads seeking plaintiffs for class action suits against the bank which negatively depict Kevin's father. Kevin meets privately with Jimmy and accepts a deal to pay Acker and the photographer. Kim angrily confronts Jimmy, upset that he made her the "sucker" for his con. She suggests getting married. Gus, Victor, and Mike meet with Nacho, who reports Lalo's plan to reveal the locations of Gus's street dealers to the police. Gus directs Victor to give up low-level employees while ensuring nothing leads back to Gus. Mike secretly points the police to Lalo's involvement in the murder of Fred the money wire clerk,[c] and they converge on Lalo while he is driving.

An informant makes a telephone call to report their presence. Lalo directs Jimmy to a remote pickup site and pays him $100,000 to transport the cash. Kim says she does not want him to go because he is a lawyer, not a "bagman" for drug dealers, but he says it will be an easy job. The Cousins deliver two bags of cash to Jimmy, then depart. As Jimmy starts his return trip, he is cut off by several gunmen. They take the money and prepare to kill him but are attacked by an unseen sniper. All but one are killed and the survivor escapes. The shooter is Mike, who drives off with Jimmy and the cash. Jimmy's car breaks down, so they push it over the edge of the road, then walk cross-country to avoid the gunman. They camp overnight, then continue their trek. When Jimmy fails to return, Kim asks Lalo for his location, but Lalo refuses. The gunman continues searching, so Jimmy acts as a decoy and draws him in. He then uses his ill-gotten gains to compete with HHM by creating a billboard which mimics HHM's logo and Howard's signature look. This results in a cease and desist order and Jimmy is required to remove the advertisement. He tries to generate sympathetic media coverage by faking the rescue of the worker who is removing the billboard. To Howard's disgust, Jimmy's scam generates positive local news coverage and new clients. Jimmy unsuccessfully attempts to conceal his deceit from Chuck.

In 2002, Jimmy continues to work in elder law. While assisting a resident of the Sandpiper Crossing home with her will, Jimmy discovers that the company overcharges residents for expenses. After speaking to several other residents, Jimmy suspects elder abuse and presents a hastily written demand letter warning Sandpiper against destroying evidence. He is escorted from the facility, but can hear the staff shredding documents, so he searches the dumpster and finds the shreds. Jimmy and Chuck put the pieces together and recover an incriminating document. Realizing the potential of the case as a class action lawsuit, Chuck agrees to aid Jimmy. In the excitement, Chuck goes to Jimmy's car for more documents and is shocked to realize he went outside without suffering any electromagnetic hypersensitivity symptoms.

Sandpiper buries them in paperwork, and Chuck convinces Jimmy to bring the case to HHM, which can handle large lawsuits. Hamlin is eager to take it but wants to prevent Jimmy's participation by buying him off with a small of counsel fee and a percentage of the judgment. Jimmy refuses and later discovers Chuck braved his electromagnetic hypersensitivity symptoms to make a pre-meeting phone call to Howard, in which he asked Howard to prevent Jimmy's participation. Chuck admits he has never regarded Jimmy as a peer, owing to his conman's past and unconventional legal training, also admitting it was he who insisted HHM not hire Jimmy back when he passed the bar exam. Jimmy stops taking care of Chuck and allows HHM to take the case. Mike serves as a bodyguard for pharmaceutical employee Daniel Wormald, who sells Nacho stolen pills. The payment is $20 short, and Mike insists Nacho pay in full. When Daniel questions his willingness to break the deal over a small amount, Mike explains he researched Nacho ahead of time, knew he was conducting the transaction without the knowledge of his bosses, and thus had an incentive to ensure things went smoothly.

Jimmy's agreement to do a con with Marco leads to a week of scams, which ends after Marco dies. Kim calls Jimmy to say the Sandpiper case is so big HHM intends to work with Davis & Main of Santa Fe. D&M's partners know Jimmy has a good rapport with the elderly clients, so they want to hire him. Jimmy returns to Albuquerque to meet with them but hesitates and then drives out of the courthouse parking lot. He stops to ask Mike why they did not keep the Kettleman money. Mike recalls that Jimmy wanted to do the "right thing", and Mike merely did the job for which Jimmy hired him. Jimmy assures Mike that he will not make that mistake again and drives off.

When he is finally let out, he leaves behind the words "SG WAS HERE" carved in the wall. In 2002, Jimmy decides to decline Davis & Main's employment offer and closes down his practice. Kim confronts Jimmy about his odd behavior, but Jimmy is adamant about leaving the practice of law. Instead, he persuades Kim to help him con a stock trader into buying them expensive drinks and food, and then paying their tab. Thrilled by the experience, Kim spends the night with Jimmy. Daniel ("Pryce") fires Mike, since he believes he no longer needs Mike as a bodyguard. Nacho takes advantage of Mike's absence to obtain Pryce's real name and address. Pryce's house is burglarized and he calls the police. The responding officers are suspicious about the nature of the burglary and investigate further, finding an empty hidden compartment behind his couch. After reconsidering the offer to join Davis & Main, Jimmy decides to accept. E2 Cobbler from Wikipedia: Howard visits Chuck and expresses concern about Jimmy's employment at Davis & Main. Mike encounters Daniel (Pryce) when he arrives at the police station for an interview about his stolen baseball cards. Mike warns Pryce that the police must suspect that he is a drug dealer and offers to find the cards himself to keep Daniel from talking to the police. Mike tracks down Nacho and threatens to inform Tuco about Nacho's secret drug deals if Nacho does not return the cards. Nacho agrees to return Daniel's cards plus $10,000 in exchange for Daniel's Hummer. Chuck visits the HHM office, where he sits in on a firm meeting between HHM and D&M, which unnerves Jimmy. Jimmy receives a call from Mike requesting that he represent Daniel. Jimmy deflects police attention from Daniel by telling investigators the secret compartment in his living room was used to hold fetish videos. When Jimmy tells Kim about staging a video with Daniel as part of his effort to convince the police, Kim voices disapproval and says Jimmy's willingness to engage in unethical behavior jeopardizes his position at D&M.

Chuck's later attempt to imitate Jimmy falls flat. In the present, Jimmy meets Kim in the HHM document room and proposes she sue the firm. Kim refuses, pointing out that no firm would ever hire her again, and tells Jimmy to worry about his job while she worries about hers. Kim tries to get out of the document review room by working her professional and school contacts to land a major new client. She succeeds in securing the business of Mesa Verde Bank, a local institution planning to expand regionally. Howard is pleased to have the lucrative new client, but does not give Kim credit, and keeps her working in document review. Chuck tells Kim that when the McGills owned a store in Cicero, Jimmy supposedly embezzled money, which led to the store's failure. Having warned Kim about Jimmy, Chuck promises to approach Howard about moving her out of document review. Mike is approached by Tuco's uncle, Hector Salamanca, who offers Mike $5,000 to claim the gun Tuco was carrying at Mike's assault was Mike's, which will reduce Tuco's jail sentence. S2 E6 Bali Ha'i Jimmy finds it difficult adjusting to his new job at D&M, and is unable to sleep in his corporate apartment, finding comfort only when he returns to his old boiler room office. With Chuck's help, Kim is reinstated to her old position at HHM, but she is treated coldly by Howard who gives her the most humiliating and menial assignments, such as arguing unwinnable motions in court. Kim is approached by Rich Schweikart of Schweikart & Cokely, who tells her he was impressed with her performance in arguing a motion she was sure to lose, and offers to hire her at S&C. Unsure of what to do, Kim relieves her stress by running another con with Jimmy, fooling an investor into giving them a $10,000 check, which Kim keeps as a souvenir. Mike refuses Hector's deal, but Hector wages an intimidation campaign, including having subordinates break into Mike's house to scare him, and Leonel and Marco Salamanca silently threatening to kill Kaylee. Mike agrees to say the gun was his, but demands $50,000, to which Hector agrees. Mike gives $25,000 to Nacho and explains that since Tuco's sentence will be reduced, Mike has failed to uphold his part of their deal, so he is obligated to return what Nacho paid.

At an HHM meeting, Jimmy presents his client outreach report, and informs the group that he has signed up several new clients for the class action lawsuit. Chuck is suspicious of Jimmy's methods, but Jimmy deflects the question. Kim warns Jimmy to keep his methods legitimate, since she recommended him to Davis & Main, and his actions will reflect on her judgment. Stacey voices concern to Mike about gunshots she heard in her neighborhood over the previous two nights. Mike does overnight surveillance without Stacey's knowledge and does not hear or see anything out of the ordinary. At work the following morning, Stacey calls Mike. He rushes to her house, where she says there were three more gunshots the night before and points out a chip in the corner of her outside wall that she tearfully insists is from a bullet. E4 - Gloves Off Jimmy meets with the Davis & Main partners, who are angry at him for running a TV commercial without their consent, but Cliff decides to give Jimmy a second chance. Kim meets with Howard and Chuck, and Howard reprimands her for her failure to inform them about Jimmy's commercial, then demotes her to entry level work in the firm's document review office. Mike and Nacho go over plans to kill Tuco, due to Nacho's concern about Tuco's erratic behavior and the possibility of Tuco finding out about his secret deals. Instead, Mike stages a fender bender by hitting Tuco's car with his, then goads Tuco into attacking him. Jimmy disbelieves him and tries to warn his father, but his father is more concerned that suspicion could lead him to not help someone who might truly be in need. When the grifter uses mostof the money given by Jimmy's father to buy cigarettes, Jimmy pockets the money. In 2002, Jimmy helps represent Mike when he claims to the district attorney that the gun in his recent altercation with Tuco did not belong to Tuco. Jimmy decides to quit D&M, but learns that if he quits, he will have to repay the firm's signing bonus. Jimmy discovers his contract allows him to keep the bonus if he is fired without cause. He takes several actions designed to be irritating to his D&M co-workers, and Cliff eventually fires him after saying that losing the bonus money is worth it to be rid of him. Jimmy attempts to convince Kim to become a partner in their own law firm. Kim agrees only on the condition that Jimmy play it "straight and narrow". Kim later approaches Jimmy with a compromise, suggesting they start separate solo firms, but share office space so that they can split expenses and lend each other support. Mike promises to buy Stacey a new house in a safer neighborhood and begins surveilling Hector's restaurant. S2 E8 - "Fifi" Kim tells Howard she is leaving HHM. Howard wishes her well, and immediately after Kim leaves, they each race to secure the Mesa Verde account. Kim meets with Kevin and Paige, who agree that Kim will be Mesa Verde's outside counsel. Kim and Jimmy set up their practices in a re-purposed dentists' office. Howard informs Chuck of Kim's resignation, the loss of Mesa Verde and Kim's teaming with Jimmy. Chuck damns Kim with faint praise at a meeting with Kevin and Paige, causing Kevin to keep Mesa Verde's business at HHM. Jimmy perpetrates a ruse at a U.S. Air Force base in order to gain access to the B-29 bomber FIFI and shoot video footage for use in TV ads promoting his law practice. Kim tells Jimmy she lost Mesa Verde. Ernesto tells Jimmy Chuck is ill from leaving his house to meet with Kevin and Paige and Jimmy says he will stay with Chuck. While Chuck is asleep Jimmy takes Mesa Verde documents to a copy store, alters them, then replaces them in Chuck's files. He stores legal documents at Chuck's house in a plan to get Chuck interested in cases so he will regain his interest in practicing law. Jimmy asks Kim to join him in a law partnership, but she cites her loyalty to HHM as her reason for declining. The Kettlemans fire HHM and hire Jimmy after Kim tries to get them to accept a plea bargain that includes jail time for Craig. Jimmy also tries to get them to accept, but Betsy blackmails Jimmy by pointing out that the "retainer" the Kettlemans paid him could be considered a bribe.
