Abstract: Obesity substantially increases the risk of type 2 diabetes, cardiovascular disease, and other diseases, making it a leading preventable cause of death in developed countries. It has a strong genetic basis, with obesity-associated genetic variants preferentially acting in the brain. This includes the hypothalamic pro-opiomelanocortin (POMC) neurons that inhibit food intake and are stimulated by drugs that agonise glucagon-like 1 peptide receptor (GLP1R) including Semaglutide (Ozempic/Wegovy). We therefore hypothesised that drugs which selectively activate human POMC neurons would suppress appetite and promote weight loss, and that focusing on drugs already approved for use would facilitate rapid clinical translation. We therefore generated POMC neurons from human pluripotent stem cells (hPSCs) and identified enriched genes that were genetically associated with obesity and targeted by approved drugs. We found that human POMC neurons are enriched in GLP1R, reliably activated by Semaglutide, and their responses are further increased by co-administration of Ceritinib, an FDA-approved drug potently and selectively inhibiting anaplastic lymphoma kinase (ALK). Ceritinib reduced food intake and body weight in obese but not lean mice, and upregulated the expression of GLP1R in the mouse hypothalamus and hPSC-derived human hypothalamic neurons. These studies reveal a new potential therapeutic strategy for reducing food intake and body weight, and demonstrate the utility of hPSC-derived hypothalamic neurons for drug discovery. Goleador innato, el instinto de anotador lo llevó lejos y a logar no sólo campeonar, sino a ser el último gran killer del área que tuvieron los Pumas de la universidad. Jugó en varios equipos, y muchos de ellos de gran envergadura. En Argentina en Estudiantes de la Plata, Independiente de Avellaneda y Boca Juniors. Saliendo de su país natal, fue al Sporting de Lisboa, donde además jugó Champions. En España Villarreal y Tenerife fueron los que contaron con sus magníficos goles. Y en México, Pumas, Toluca, Atlas, Pachuca y Estudiantes Tecos fueron los que lograron contarlo en sus filas. En su actual etapa como técnico, ha pasado por Tepatitlán, Venados y Pumas.

Abstract: The emergence of epigenetic predictors was a pivotal moment in geroscience, propelling the measurement and concept of biological ageing into a quantitative era. However, while current epigenetic clocks have shown strong predictive power, they do not reflect the underlying biological mechanisms driving methylation changes with age. Consequently, biological interpretation of their estimates is limited. Furthermore, our findings suggest that clocks trained on chronological age are confounded by non-age-related phenomena. To address these limitations, we developed a probabilistic model that describes methylation transitions at the cellular level. Our approach reveals two measurable components, acceleration and bias, that directly relate to perturbations of the underlying cellular dynamics. Acceleration is the proportional increase in the speed of methylation transitions across CpG sites, whereas bias is the degree of global change in methylation affecting all CpG sites uniformly. Using data from 7,028 participants from the Generation Scotland study, we found the age acceleration parameter to be associated with physiological traits known to impact healthy ageing. Furthermore, a genome-wide association study of age acceleration identified four genomic loci previously linked with ageing. Copy rights belong to original authors. Abstract: The liver has a unique ability to regenerate however in the setting of acute liver failure (ALF) this regenerative capacity is often overwhelmed and emergency liver transplantation is the only curative option. To advance our understanding of human liver regeneration and to inform design of pro-regenerative therapies, we use paired single-nuclei RNA sequencing (snRNAseq)combined with spatial profiling of healthy and ALF explant human livers to generate the first single-cell, pan-lineage atlas of human liver regeneration. We uncover a novel ANXA2+ migratory hepatocyte subpopulation which emerges during human liver regeneration, and a corollary migratory hepatocyte subpopulation in a mouse model of acetaminophen (APAP)-induced liver regeneration. Importantly, interrogation of necrotic wound closure and hepatocyte proliferation across multiple timepoints following APAP-induced liver injury in mice demonstrates that wound closure precedes hepatocyte proliferation. 4-D intravital imaging of APAP-induced mouse liver injury identifies motile hepatocytes at the edge of the necrotic area, enabling collective migration of the hepatocyte sheet to effect wound closure. Depletion of hepatocyte ANXA2 expression reduces HGF-induced human and mouse hepatocyte migration in vitro, and abrogates necrotic wound closure following APAP-induced mouse liver injury. Taken together, our work dissects unanticipated aspects of liver regeneration, demonstrating an uncoupling of wound closure and hepatocyte proliferation and uncovering a novel migratory hepatocyte subpopulation which mediates wound closure following liver injury. Therapies designed to promote rapid reconstitution of normal hepatic microarchitecture and reparation of the gut-liver barrier may open up new areas of therapeutic discovery in regenerative medicine. Lu, Alexandra M. Binder, Joshua Zhang, Qi Yan, Alex P. Reiner, Simon R. Cox, Janie Corley, Sarah E. Harris, Pei-Lun Kuo, Ann Z. Moore, Stefania Bandinelli, James D. Stewart, Cuicui Wang, Elissa J. Hamlat, Elissa S. Epel, Joel D. Schwartz, Eric A. Whitsel, Adolfo Correa, Luigi Ferrucci, Riccardo E. Marioni, and Steve Horvath from the University of California Los Angeles, Altos Labs, University of Hawaii at Manoa, Fred Hutchinson Cancer Research Center, University of Edinburgh, National Institute on Aging, Gillings School of Global Public Health, University of North Carolina at Chapel Hill, Harvard T.H. Chan School of Public Health, University of California – San Francisco, and the University of Mississippi Medical Center previously described a DNA methylation (DNAm) based biomarker of human mortality risk DNAm GrimAge. In their current study, the researchers describe version 2 of GrimAge (trained on individuals aged between 40 and 92) which leverages two new DNAm based estimators of (log transformed) plasma proteins: high sensitivity C-reactive protein (logCRP) and hemoglobin A1C (logA1C). “To arrive at version 2 of GrimAge, we developed two additional DNAm based surrogates for plasma proteins that are widely used in the clinic (DNAm logCRP and DNAm logA1C).” The team evaluated GrimAge2 in 13,399 blood samples across nine study cohorts. After adjustment for age and sex, GrimAge2 outperforms GrimAge in predicting mortality across multiple racial/ethnic groups (meta P=3.6×10-167 versus P=2.6×10-144) and in terms of associations with age related conditions such as coronary heart disease, lung function measurement FEV1 (correlation= -0.31, P=1.1×10-136), computed tomography based measurements of fatty liver disease. The researchers presented evidence that GrimAge version 2 also applies to younger individuals and to saliva samples where it tracks markers of metabolic syndrome. DNAm logCRP is positively correlated with morbidity count (P=1.3×10-54). DNAm logA1C is highly associated with type 2 diabetes (P=5.8×10-155). DNAm PAI-1 outperforms the other age-adjusted DNAm biomarkers including GrimAge2 in correlating with triglyceride (cor=0.34, P=9.6×10-267) and visceral fat (cor=0.41, P=4.7×10-41). Overall, the team demonstrated that GrimAge version 2 is an attractive epigenetic biomarker of human mortality and morbidity risk. “GrimAge2 will not replace existing clinical biomarkers. C., Marioni, R. E., Mur, J. Abstract: Genetic variation in VKORC1 is associated with differences in the coagulation of blood and consequentially with sensitivity to the drug warfarin. Variation in VKORC1 has also been linked to parental dementia. However, it is unclear whether the relationship persists for the diagnosis in patients themselves, whether the association holds only for certain forms of dementia, and if those taking warfarin are at greater risk. Here, we use data from 211,423 participants from UK Biobank to examine the relationship between VKORC1, risk of dementia, and the interplay with warfarin use. We find that the T-allele in rs9923231 confers a greater risk for vascular dementia (OR=1.28, p=0.0069), but not for general dementia (OR=1.04, p=0.21) or Alzheimer dementia (OR=1.05, p=0.35), and that the risk of vascular dementia is not affected by warfarin use in carriers of the T-allele. Our study reports for the first time an association between rs9923231 and vascular dementia, but further research is warranted to explore potential mechanisms and specify the relationship between rs9923231 and features of vascular dementia. It’s not uncommon to read comparisons between Albert Einstein and Paul Marioni, artist and one of the founders of the Studio Glass movement, many based on their shared lifelong fascination with light. Known as an innovator in the glass world, Marioni has been pushing the limits of his medium for five decades, redefining what is possible not only in process but content. He says: “I work with glass for its distinct ability to capture and manipulate light. While my techniques are often inventive, they are only in service of the image.” A surrealist whose work addresses issues of nature, identity, and emotion, Marioni relies upon dreams as well as political and social convictions to make statements, causing us to forget the unfair advantage that working with glass affords. Using material that is inherently beautiful, the artist inspires people to think rather than telling them what to think. Marioni’s work can be found in collections including the Smithsonian American Art Museum, Washington, D.C.; The Corning Museum of Glass, Corning, New York; and the Museum of Arts and Design, New York, New York. Marioni, who graduated in 1967 from the University of Cincinnati, Ohio, is a Fellow of the American Crafts Council and Glass Art Society Lifetime Achievement Award recipient. He has received three fellowships from the National Endowment of the Arts and has taught at schools worldwide including the Penland School of Crafts, Bakersville, North Carolina; Pilchuck Glass School, Stanwood, Washington; the Glass Furnace, Istanbul, Turkey; and more recently at Soneva Art and Glass in the Maldives. At 77, Marioni remains passionate about the “road show,” a grassroots effort started by artists like himself, Fritz Dreisbach, Dale Chihuly, and Richard Marquis, to spread knowledge and enthusiasm about glass to anyone who showed interest. “I’ve worked in glass all but three years of my life. What was I thinking to get into a field with no history, no books, no teaching? Obviously I wasn’t thinking. But we built the Studio Glass movement on cooperation, not competition, because there was no past. There was nothing for us to get. And we’ve barely scratched the surface of what can be done.” In addition to gallery work, Marioni has produced over 100 public and private commissions in both cast glass and terrazzo. From his studio in Mexico, the artist currently works on the biggest commission of his career for the $52 million Bellevue, Washington, light rail station. Its train serves business powerhouses of the Pacific Northwest including Microsoft and Boeing. Selected as lead artist for the project through a national competition, Marioni is designing 3000+ square feet of art glass for the platforms as well as the terrazzo floors.

On May 12 at Chihuly Garden and Glass in Seattle, Washington, a full capacity audience of 275 guests attended Pilchuck Celebrates and toasted the 2016 Honorees. Glassblowing pioneer Dante Marioni, 52, was awarded The Libenský | Brychtová Award, which acknowledges extraordinary talent and high achievement in the world of contemporary glass. Marioni took the glass world by storm at age 19 with his exacting and intuitive command of both Venetian design and technique. The resulting monumental Post Modern sculpture referred to vessels and paid homage to glassblowing itself. His early aesthetic signature included graceful form, impeccable finish, and the use of solid opaque colors, all of which made his work instantly recognizable. Always looking for a new challenge, Marioni’s series are short-lived. The natural evolution of process and form led to an aesthetic shift away from opaque, bold color to an exploration of pattern created through Venetian techniques such as reticello and latticino in transparent glass. Marioni’s current work beguiles the viewer with its intricacy, beauty, and perfection of form, revealed in sculptural vessels that are inspired by the stylized leaf as found in the decorative arts. Marioni’s work will be exhibited in Classical, Native, and Pop Cultures, on view September 2 – 30 at Schantz Gallery, Stockbridge, Massachusetts, along with the work of Preston Singletary and Richard Marquis. His solo exhibition, In a Breath, opens November 4 at Ken Saunders Gallery, Chicago, Illinois, during the first week of SOFA and runs through December 2016.
