In this episode, we discuss the gut-immune axis and Small Intestinal Fungal Overgrowth (SIFO), with a mechanistic focus on immune activation and epithelial barrier disruption. We detail contributing factors to and symptoms of SIFO. We further discuss the anatomical positioning and immunological functions of mast cells within the lamina propria, highlighting mast cell activation syndrome, while detailing how SIFO and SIBO can serve as upstream triggers for mast cell activation.Topics:1. The Gut-Immune Axis and Mast Cells - The intestinal epithelium forms the innermost selective barrier, coated in a protective mucus layer.- Immediately beneath lies the lamina propria, rich in immune cells including mast cells. 2. Mast Cells in the Lamina Propria - Mast cells are positioned near nerves, capillaries, lymphatics, and epithelial cells within the lamina propria.- Their anatomical location enables rapid immune surveillance and inflammatory response.- Mast cell granules store histamine and more. 3. Mast Cell Mediators - Histamine: Biogenic amine, inflammatory signaling.- Tryptase: A serine protease.- Cytokines and lipid mediators also released upon activation. 4. Mast Cell Activation Syndrome (MCAS) - Systemic: skin, respiratory tract, cardiovascular system, nervous system, and more.- The gastrointestinal tract.- Common triggers for mast cell activation. 5. Transition to Small Intestinal Fungal Overgrowth (SIFO) - Overgrowth of fungal organisms, often candida species, within the small intestine.- Contributing factors including hypochlorhydria, slowed motility, immune suppression, disrupted bacterial populations.- Overlap: SIFO, SIBO  6. Hypochlorhydria and the Stomach's Defense Role - Parietal cells secrete hydrochloric acid (HCl) and intrinsic factor.- HCl denatures proteins and sterilizes ingested pathogens.- Chief cells secrete pepsinogen, which becomes pepsin in acidic conditions to digest proteins.- Consequences of Reduced Gastric Acidity  7. The Role of Intestinal Motility - Enteric nervous system (ENS), myenteric and submucosal plexuses.- Coordinated contractions preventing stasis and microbial overgrowth. 8. Common Symptoms of SIFO and clinical overlap with SIBO 9. Candida and Fungal Pathophysiology in the Gut - Yeast form, regulated by microbial competition and immune defenses.- Hyphal transformation.- Degrade the mucus layer, disrupt epithelial integrity, trigger inflammation. 10. Secretory IgA and Mucosal Defense - Chronic stress, immune dysfunction, dysbiosis can lower sIgA levels and weaken mucosal immunity. 11. SIFO, SIBO, and Mast Cell Activation  12. More efficient and longer-lasting fuel cells are essential for fuel cell-powered heavy-duty hydrogen vehicles to be an alternative to combustion fuelled counterparts. Researchers at Chalmers University of Technology, Sweden, have developed an innovative method to study and understand how parts of fuel cells degrade over time. This is an important step towards the improved performance of fuel cells and them becoming commercially successful. How fuel cells degrade over time Hydrogen is a fuel alternative that is becoming increasingly interesting for heavy-duty vehicles. Hydrogen-powered vehicles only emit water vapour as exhaust, and if the hydrogen is produced using renewable energy, it is completely free of carbon dioxide emissions. Unlike battery-powered electric vehicles, hydrogen-powered vehicles do not need to burden the electricity grid, as hydrogen can be produced and stored when electricity is cheap. For some hydrogen-powered vehicles the propulsion comes from a so-called fuel cell. However, hydrogen-fuel-cell-powered vehicles are limited by a relatively short lifespan, because fuel cell components, such as electrodes and membranes, degrade over time. It is this problem that the recent study addresses. Researchers at Chalmers University of Technology have developed a new method for studying what affects the ageing of fuel cells by tracking a specific particle in the fuel cell during use. The team of researchers have studied an entire fuel cell by taking it apart at regular intervals. Using advanced electron microscopes, they have then been able to follow how the cathode electrode degrades in specific areas during the cycles of use. Previous studies have been done on so-called half-cells, which are similar (but not the same as) half of a fuel-cell and are carried out under conditions that differ significantly from the real fuel cell. Better understanding with new experimental method "It has previously been assumed that the performance would be affected by the fuel cell being disassembled and studied in the way we have done, but it turned out that this assumption is not correct, which is surprising," says research leader Björn Wickman, Associate Professor at the Department of Physics at Chalmers. The researchers at Chalmers have been able to explore how the material in the fuel cell degrades at both the nano and micro level, and pinpoint exactly when and where the degradation occurs. This provides valuable information for the development of new and improved fuel cells with a longer lifespan. "From previously only looking at how the fuel cell has aged after use, we have now been able to look into the middle stage," says doctoral student Linnéa Strandberg at Chalmers. "Being able to follow a single, chosen particle within a specific area, provided a much better understanding of the degradation processes. Greater knowledge of these is an important step on the way to designing new materials for fuel cells or to adjust the control of the fuel cell." New method paves way for longer lasting fuel cells The U.S. Department of Energy (DOE) has pointed out that improved lifetime of fuel cells is one of the most important goals to reach before fuel cell-powered hydrogen vehicles can become commercially successful. According to the industry, a truck needs to be able to withstand 20,000 - 30,000 hours of driving over its lifetime, which a fuel cell-powered hydrogen truck cannot achieve today. "We have now laid a foundation on which to build for the development of better fuel cells. Now we know more about the processes that take place in the fuel cell and at what point over the lifetime of the fuel cell they occur. In the future, the method will be used to develop and study new materials that can give the fuel cell a longer lifespan," says Björn Wickman. Facts: How a fuel cell works The core of a fuel cell consists of three active layers, two electrodes - anode and cathode respectively - with an ion-conducting membrane in the middle....

One of the primary reasons why men struggle with women is because we use logic to try and appeal to them. Surely by giving her everything she wants and being a good boy, she's going to fall in love with me, right? Nothing could be further from the truth, when you over-supplicate and confirm to a woman, she ends up losing both respect and attraction for you. All you will do is turn into someone you are not and end up miserable for it. So in today's episode, Mark is going to equip you with the tools you need to identify when a woman is trying to make you conform to her too much, so you can remain steadfast in your frame and ensure it's HER that's stepping into your world and not the other way around. In this episode, we explore the destructive impact of trawling, an unsustainable fishing method that is contributing to overfishing and the degradation of the ocean. We discuss how trawling destroys the ocean floor and its potential effects on climate change. Additionally, we delve into a new study that reveals the significant carbon release associated with bottom trawling. This episode highlights the urgent need to address the environmental consequences of fishing practices and offers insights on how individuals can take action to protect the ocean. While efforts are being made to conserve fisheries and promote sustainable fishing practices, certain methods, such as trawling, remain highly unsustainable. In this episode, we delve into the destructive nature of trawling and its impact on the ocean floor, as well as its contribution to climate change. Trawling involves dragging a massive net along the ocean floor, scraping and collecting everything in its path. This method not only destroys the surrounding environment but also releases significant amounts of carbon into the atmosphere. A recent study has estimated that trawling releases approximately 370 million tons of carbon dioxide annually, making it a significant contributor to global warming. The study, conducted by a global team of climate and ocean experts, analyzed data from 1996 to 2020 and found that trawling released a staggering 8.5 to 9.2 billion tons of carbon dioxide into the atmosphere during that period. The researchers likened trawling to marine deforestation, causing irreparable harm to the climate, society, and wildlife. The carbon released by trawling not only contributes to global warming but also leads to local acidification, reducing the ocean's capacity to absorb carbon. Sediment plumes created by trawling can be seen from space, similar to vapor trails left by aircraft. The study identified trawling hotspots, including the East China Sea, Baltic Sea, North Sea, and Greenland Sea, as significant sources of climate emissions. It is alarming to note that over half of the carbon released by trawling enters the atmosphere within a decade. This means that even if trawling were to cease immediately, the carbon emissions from past trawling activities would continue to impact the climate for years to come. The study also highlighted the fact that 75% of fishing vessels are not publicly tracked, indicating that the estimates of carbon emissions from trawling may be conservative. To address this issue, the study authors emphasize the need for countries to include trawling emissions in their climate action plans. By regulating and reducing trawling activities, immediate benefits can be achieved in terms of carbon emissions reduction. Failure to take action will result in the continued release of carbon dioxide into the atmosphere, exacerbating the effects of climate change. In conclusion, trawling is a highly destructive fishing method that not only destroys marine habitats but also releases significant amounts of carbon dioxide into the atmosphere. It is crucial for countries to prioritize the regulation and reduction of trawling activities to mitigate the impacts of climate change and protect marine ecosystems.

On this week's show: Comparing human-dog bonds with human-wolf bonds, and monitoring termite decay rates on a global scale First up on the podcast this week, Online News Editor David Grimm talks with host Sarah Crespi about the bonds between dogs and their human caretakers. Is it possible these bonds started even before domestication? Also this week, Sarah talks with Amy Zanne, professor and Aresty endowed chair in tropical ecology in the Department of Biology at the University of Miami. They discuss a global study to determine whether climate change might accelerate the rate at which termites and microbes break down dead wood and release carbon into the atmosphere. This week's episode was produced with help from Podigy. In Degrade and Destroy: The Inside Story of the War Against the Islamic State, from Barack Obama to Donald Trump (FSG, 2022), Wall Street Journal national security correspondent Michael R. Gordon reveals the strategy debates, diplomatic gambits, and military operations that shaped the struggle against the Islamic State. With extraordinary access to top U.S. officials and military commanders and to the forces on the battlefield, Gordon offers a riveting narrative that ferrets out some of the war's most guarded secrets. Degrade and Destroy takes us inside National Security Council meetings at which Obama and his top aides grapple with early setbacks and discuss whether the war can be won. It also offers the most detailed account to date of how President Donald Trump waged war--delegating greater authority to the Pentagon but jeopardizing the outcome with a rush for the exit. Drawing on his reporting in Iraq and Syria, Gordon documents the closed-door deliberations of U.S. generals with their Iraqi and Syrian counterparts and describes some of the toughest urban battles since World War II. As Americans debate the future of using force abroad, Gordon's book offers vital insights into how our wars today are fought against militant foes, and the enduring lessons we can draw from them. In 2014, as Islamic State insurgents took control of the Iraqi city of Mosul, President Barack Obama made the decision to send troops back to Iraq. Within five years, through the work of the United States and its partners, the organization was largely dismantled. What was the nature of the U.S. struggle against the Islamic State? Which decisions were instrumental to its success? And how did the U.S. coordinate with partners in the region. To discuss these issues, former Lawfare associate editor Bryce Klehm spoke with Michael Gordon, a national security correspondent for the Wall Street Journal, about his new book, “Degrade and Destroy,” Gordon's fourth book on Iraq. They covered a range of topics, including the status of forces agreement, or SOFA, the Trump administration's counter-Islamic State strategy, and the challenges for journalists embedding with coalition forces. To be disgruntled at anything that happens is a kind of secession from Nature, which comprises the nature of all things. When it turns its back on another person or sets out to do it harm, as the souls of the angry do. When it is overpowered by pleasure or pain When it puts on a mask and does or says something artificial or false When it allows its action and impulse to be without a purpose, to be random and disconnected: even the smallest things ought to be directed toward a goal. But the goal of rational beings is to follow the rule and law of the most ancient of communities and states." -- Meditations: A New Translation (the book I read these meditations from) --> [link]
