Abstract: Transient Receptor Potential (TRP) channels have evolved in eukaryotes to control various cellular functions in response to a wide variety of chemical and physical stimuli. This large and diverse family of channels emerged in fungi as mechanosensitive osmoregulators. The Saccharomyces cerevisiae vacuolar TRP yeast 1 (TRPY1) is the most studied TRP channel from fungi, but the molecular details of channel modulation remain elusive so far. Here, we describe the full-length cryo-electron microscopy structure of TRPY1 at 3.1 A resolution. The structure reveals a distinctive architecture for TRPY1 among all eukaryotic TRP channels with an evolutionarily conserved and archetypical transmembrane domain, but distinct structural folds for the cytosolic N- and C-termini. We identified the inhibitory phosphatidylinositol 3-phosphate (PI(3)P) lipid binding site, which sheds light into the lipid modulation of TRPY1 in the vacuolar membrane. The structure also exhibited two Ca2+-binding sites: one in the cytosolic side, implicated in channel activation, and the other in the vacuolar lumen side, involved in channel inhibition. These findings, together with data from molecular dynamics simulations, provide structural insights into the basis of TRPY1 channel modulation by lipids and calcium. Today we have Veronica Petrovsky with us, live in the studio to give us a detailed look into the life of a young woman who grew up seeing her mother purge, evidently leading to her eating disorder, anorexia nervosa. This is a must listen as we go into self-love, confidence, and so much more behind the deathly disorder.

In recent months, I have had a growing number of young women coming into my office for treatment with eating disorders, namely EDNOS, which stands for Eating Disorders Not Otherwise Specified, Anorexia Nervosa and Bulimia Nervosa. Before we start, I want to make it clear that EDNOS, Anorexia and Bulimia are hardly about food itself; food and the act of consuming it is just a byproduct. Rather, for some it is about achieving perfectionism, while for others it is about achieving the same kind of “high” one gets from cocaine, drugs, and other substances. The act of purging gives individuals a sort of “high” that keeps them going. Moreover, people that suffer from EDNOS, Anorexia and Bulimia use their power in being able to control their weight and appearance as a defense to hide the hurt, anxiety, fear, and OCD that goes on internally. To have the power to control one’s weight seems to substitute their lack in having control over past events and in other area of their lives. Anorexia and bulimia become an addiction. The tricky thing is, you can quit booze, you can quit drugs, and you can quit sex, but you can’t quit food. With us today, we have Elena Dunkle, an Anorexia Nervosa survivor and co-writer of “Elena Vanishing”, a memoir about her journey through recovery.
